    Exercise-induced respiratory muscle work: Effects on blood flow, fatigue and performance

The official published version can be obtained from the link below.In healthy subjects, heavy intensity endurance exercise places substantial demands on the respiratory muscles as breathing frequency, ventilation and the work of breathing rise over time. In the highly trained subject working at high absolute work rates, the ventilatory demand often causes varying degrees of expiratory flow limitation, sometimes accompanied by lung hyperinflation and, therefore, increased elastic work of breathing. Time-dependant increases in effort perceptions for both dyspnea and limb discomfort accompany these increased ventilatory demands. Similar responses to endurance exercise but at much lower exercise intensities also occur in patients with COPD and CHF. Note that these responses significantly influence exercise performance times in both health and disease. This effect was demonstrated by the marked reductions in the rate of rise of effort perceptions and the enhanced exercise performance times elicited by unloading the respiratory muscles using pressure support ventilation or proportional assist mechanical ventilation. In healthy fit subjects, unloading the inspiratory work of breathing by about one half increased performance by an average of 14% (Harms et al. 2000), and in CHF and COPD patients performance time more than doubled with respiratory muscle unloading (O’Donnell et al. 2001). Why are effort perceptions of limb discomfort markedly reduced and exercise performance increased when the respiratory muscles are unloaded? Our hypothesis is shown in Fig. 1

    We introduce a simple, practical approach with probabilistic information-theoretic security to mitigate one of quantum key distribution's major limitations: the short maximum transmission distance (~200 km) possible with present day technology. Our scheme uses classical secret sharing techniques to allow secure transmission over long distances through a network containing randomly-distributed compromised nodes. The protocol provides arbitrarily high confidence in the security of the protocol, with modest scaling of resource costs with improvement of the security parameter.     In vivo probing of nascent RNA structures reveals principles of cotranscriptional folding

    Defining the in vivo folding pathway of cellular RNAs is essential to understand how they reach their final native conformation. We here introduce a novel method, named Structural Probing of Elongating Transcripts (SPET-seq), that permits single-base resolution analysis of transcription intermediates' secondary structures on a transcriptome-wide scale, enabling base-resolution analysis of the RNA folding events. Our results suggest that cotranscriptional RNA folding in vivo is a mixture of cooperative folding events, in which local RNA secondary structure elements are formed as they get transcribed, and non-cooperative events, in which 5'-halves of long-range helices get sequestered into transient non-native interactions until their 3' counterparts have been transcribed. Together our work provides the first transcriptome-scale overview of RNA cotranscriptional folding in a living organism

    Effect of the MC1R gene on sexual dimorphism in melanin-based colorations.

    Variants of the melanocortin-1 receptor (MC1R) gene result in abrupt, naturally selected colour morphs. These genetic variants may differentially affect sexual dimorphism if one morph is naturally selected in the two sexes but another morph is naturally or sexually selected only in one of the two sexes (e.g. to confer camouflage in reproductive females or confer mating advantage in males). Therefore, the balance between natural and sexual selections can differ between MC1R variants, as suggest studies showing interspecific correlations between sexual dimorphism and the rate of nonsynonymous vs. synonymous amino acid substitutions at the MC1R. Surprisingly, how MC1R is related to within-species sexual dimorphism, and thereby to sex-specific selection, has not yet been investigated. We tackled this issue in the barn owl (Tyto alba), a species showing pronounced variation in the degree of reddish pheomelanin-based coloration and in the number and size of black feather spots. We found that a valine (V)-to-isoleucine (I) substitution at position 126 explains up to 30% of the variation in the three melanin-based colour traits and in feather melanin content. Interestingly, MC1R genotypes also differed in the degree of sexual colour dimorphism, with individuals homozygous for the II MC1R variant being 2 times redder and 2.5 times less sexually dimorphic than homozygous individuals for the VV MC1R variant. These findings support that MC1R interacts with the expression of sexual dimorphism and suggest that a gene with major phenotypic effects and weakly influenced by variation in body condition can participate in sex-specific selection processes

    Cooperation and Self-Regulation in a Model of Agents Playing Different Games

    A simple model for cooperation between "selfish" agents, which play an extended version of the Prisoner's Dilemma(PD) game, in which they use arbitrary payoffs, is presented and studied. A continuous variable, representing the probability of cooperation, pk(t)p_k(t) \in [0,1], is assigned to each agent kk at time tt. At each time step tt a pair of agents, chosen at random, interact by playing the game. The players update their pk(t)p_k(t) using a criteria based on the comparison of their utilities with the simplest estimate for expected income. The agents have no memory and use strategies not based on direct reciprocity nor 'tags'. Depending on the payoff matrix, the systems self-organizes - after a transient - into stationary states characterized by their average probability of cooperation pˉeq\bar{p}_{eq} and average equilibrium per-capita-income pˉeq,Uˉ\bar{p}_{eq},\bar{U}_\infty. It turns out that the model exhibit some results that contradict the intuition. In particular, some games which - {\it a priory}- seems to favor defection most, may produce a relatively high degree of cooperation.     Water quality study of the Muchea livestock truck wash

    Across Australia there is a lack of information on the quality of the water discharged from facilities that are used to wash livestock trucks. This water quality scoping study partially fills that information gap and provides a starting point for future planning, design and construction of livestock truck washes. The study was undertaken in 2011–12 at the Western Australian Muchea Livestock Centre, and aimed to gain insights into water quality associated with the truck wash facility at that site. These insights can help to inform the planning for further construction of truck wash facilities throughout the state, and whether disposing wastewater to Water Corporation’s sewerage system could be part of a new facility. We examined water quality at primary points of the wastewater treatment system to understand the impact of each part of the system and determine the most appropriate site for detailed temporal monitoring. This was followed by a two-day sampling program at one point in the treatment system. We concluded that the minimum infrastructure requirements to satisfy Water Corporation’s maximum allowable limits for disposal to the sewerage system include sieve bend screens (Hunter screens), an anaerobic or settling pond, and a holding pond to ensure sufficient safety margins if sewer disposal was ever delayed. We recommend that further consideration is given to estimating the capital and operational costs of a truck wash facility that meets these minimum requirements, compared to a closed system that retains all wastewater on-site, or other systems that recover nutrients from high-value products. We also recommend that a sampling program be conducted across all truck washes in WA to gain insight into possible geographical variations and the wider applicability of this study’s findings

    Entangling Two Bose-Einstein Condensates by Stimulated Bragg Scattering

    We propose an experiment for entangling two spatially separated Bose-Einstein condensates by Bragg scattering of light. When Bragg scattering in two condensates is stimulated by a common probe, the resulting quasiparticles in the two condensates get entangled due to quantum communication between the condensates via probe beam.     Molecular evolution of the proopiomelanocortin system in Barn owl species.

    Examination of genetic polymorphisms in outbred wild-living species provides insights into the evolution of complex systems. In higher vertebrates, the proopiomelanocortin (POMC) precursor gives rise to α-, β-, and γ-melanocyte-stimulating hormones (MSH), which are involved in numerous physiological aspects. Genetic defects in POMC are linked to metabolic disorders in humans and animals. In the present study, we undertook an evolutionary genetic approach complemented with biochemistry to investigate the functional consequences of genetic polymorphisms in the POMC system of free-living outbred barn owl species (family Tytonidae) at the molecular level. Our phylogenetic studies revealed a striking correlation between a loss-of-function H9P mutation in the β-MSH receptor-binding motif and an extension of a poly-serine stretch in γ3-MSH to ≥7 residues that arose in the barn owl group 6-8 MYA ago. We found that extension of the poly-serine stretches in the γ-MSH locus affects POMC precursor processing, increasing γ3-MSH production at the expense of γ2-MSH and resulting in an overall reduction of γ-MSH signaling, which may be part of a negative feedback mechanism. Extension of the γ3-MSH poly-serine stretches ≥7 further markedly increases peptide hormone stability in plasma, which is conserved in humans, and is likely relevant to its endocrine function. In sum, our phylogenetic analysis of POMC in wild living owls uncovered a H9P β-MSH mutation subsequent to serine extension in γ3-MSH to 7 residues, which was then followed by further serine extension.
