    The Organizational Effects of Oxytocin on the Central Expression of Estrogen Receptor Alpha and Oxytocin in Adulthood

    Background: Previous studies have demonstrated that neonatal manipulation of oxytocin (OT) has effects on the expression of estrogen receptor alpha (ER alpha) and the central production of oxytocin observed in juveniles ( at weaning, 21 days of age). The goal of this study was to determine whether the effects of neonatal manipulation of OT last into adulthood, and if the effects differ from those observed during the early postnatal period. On the first day of life, prairie voles ( Microtus ochrogaster) received one of three doses of OT (High, 3 mu g; Med, 0.3 mu g; Low, 0.03 mu g), an OT antagonist, or isotonic saline. Another group was handled, but not injected. Then as adults, brains were collected, sectioned, and stained for ERa or OT using immunocytochemistry. Results: In females, treatment with OT increased the expression of ERa immunoreactivity in the ventral lateral septum (0.03 mu g) and the ventromedial nucleus of the hypothalamus and central amygdala ( 0.3 g). In males, OT antagonist increased ERa expression in the bed nucleus of the stria terminalis. There was no apparent effect of OT on the number of cells producing OT in the paraventricular nucleus of the hypothalamus. Conclusion: The current results suggest that neonatal manipulation of OT has long-term organizational effects on the expression of ERa in both males and females. The lack of effect on OT neurons in the paraventricular nucleus suggests that some developmental effects of OT previously observed in weanlings do not persist into adulthood.     Isotopic fractionation of water during snow formation: Experimental evidence of kinetic effect

    Deuterium excess(d-excess=δD-8・δ^(18)O), which is calculated using two water isotope ratios(δD and δ^(18)O), is an indicator of kinetic isotope fractionation. The d-excess value reflects the evaporation process from the ocean or ice-crystal growth. Consequently, d-excess records preserved in ice cores may provide a climatic history of ocean surface conditions at the vapor source area. J. Jouzel and L. Merlivat(J. Geophys. Res., 89, 11749, 1984) proposed an isotope model to analyze information from ice cores. That model includes kinetic fractionation during snow formation, depending on the degree of the supersaturation ratio of vapor. However, no experiment was conducted under the controlled supersaturation ratio. Experiments described herein measured the isotopic ratios of the vapor and artificial snow produced under a controlled supersaturation ratio to confirm the kinetic isotope effect experimentally. Results indicate a higher d-excess value for ice crystals at a higher vapor supersaturation ratio and provide experimental evidence for the kinetic effect during snow formation

    Generation of Stratified Squamous Epithelial Progenitor Cells from Mouse Induced Pluripotent Stem Cells

    Background: Application of induced pluripotent stem (iPS) cells in regenerative medicine will bypass ethical issues associated with use of embryonic stem cells. In addition, patient-specific IPS cells can be useful to elucidate the pathophysiology of genetic disorders, drug screening, and tailor-made medicine. However, in order to apply iPS cells to mitotic tissue, induction of tissue stem cells that give rise to progeny of the target organ is required. Methodology/Principal Findings: We induced stratified epithelial cells from mouse iPS cells by co-culture with PA6 feeder cells (SDIA-method) with use of BMP4. Clusters of cells positive for the differentiation markers KRT1 or KRT12 were observed in KRT14-positive colonies. We successfully cloned KRT14 and p63 double-positive stratified epithelial progenitor cells from iPS-derived epithelial cells, which formed stratified epithelial sheets consisting of five- to six-polarized epithelial cells in vitro. When these clonal cells were cultured on denuded mouse corneas, a robust stratified epithelial layer was observed with physiological cell polarity including high levels of E-cadherin, p63 and K15 expression in the basal layer and ZO-1 in the superficial layer, recapitulating the apico-basal polarity of the epithelium in vivo.     Rare earth intermetallic compounds RX and RX_2 ( R=Gd, Tb, Dy, Ho and Er : X=Ag and Au) are antiferromagnetic compounds with the CsCl-type crystal structure and the MoSi_2-type one, respectively.     Inhibitory Effects of Glycyrrhetinic Acid on DNA Polymerase and Inflammatory Activities

    We investigated the inhibitory effect of three glycyrrhizin derivatives, such as Glycyrrhizin (compound 1), dipotassium glycyrrhizate (compound 2) and glycyrrhetinic acid (compound 3), on the activity of mammalian pols. Among these derivatives, compound 3 was the strongest inhibitor of mammalian pols α, β, κ, and λ, which belong to the B, A, Y, and X families of pols, respectively, whereas compounds 1 and 2 showed moderate inhibition. Among the these derivatives tested, compound 3 displayed strongest suppression of the production of tumor necrosis factor-α (TNF-α) induced by lipopolysaccharide (LPS) in a cell-culture system using mouse macrophages RAW264.7 and peritoneal macrophages derived from mice. Moreover, compound 3 was found to inhibit the action of nuclear factor-κB (NF-κB) in engineered human embryonic kidney (HEK) 293 cells. In addition, compound 3 caused greater reduction of 12-O-tetradecanoylphorbol-13-acetate-(TPA-) induced acute inflammation in mouse ear than compounds 1 and 2. In conclusion, this study has identified compound 3, which is the aglycone of compounds 1 and 2, as a promising anti-inflammatory candidate based on mammalian pol inhibition
