    From symplectic cohomology to Lagrangian enumerative geometry

    We build a bridge between Floer theory on open symplectic manifolds and the enumerative geometry of holomorphic disks inside their Fano compactifications, by detecting elements in symplectic cohomology which are mirror to Landau-Ginzburg potentials. We also treat the higher Maslov index versions of LG potentials. We discover a relation between higher disk potentials and symplectic cohomology rings of anticanonical divisor complements (themselves related to closed-string Gromov-Witten invariants), and explore several other applications to the geometry of Liouville domains.Comment: 47 pages, 13 figures; v2: reference fixes, minor correction

    Reducing nitrogen footprints of consumer-level food loss and protein overconsumption in Japan, considering gender and age differences

    The agro-food system perturbs the nitrogen (N) cycle through its N loads to the environment. The present study focused on food-related consumer-level N loads in Japan from 1961–2015, with a particular focus on food loss and protein overconsumption. Gender and age differences were also analyzed. Consumer-level food loss was negligible until the 1970s, when it began to slowly increase, accounting for an average of 13.2% of the annual net supply during 2011–2015. Japanese people have consumed more protein than the World Health Organization’s recommended intake since 1961. Protein overconsumption increased until the mid-1990s, when it began to decrease, but it still accounted for an average of 32.3% of total annual protein consumption during 2011–2015. The national mean of food N footprints (total release of reactive N into the environment related to individual food consumption) in the same period was 18.3 kg N capita ^–1 yr ^–1 , of which food loss accounted for 4% and protein overconsumption for 37%. The food N footprint of each sex/age class varied from 16.0–21.6 kg N capita ^–1 yr ^–1 , males had a larger footprint in each age class. Seven scenarios to reduce the N footprints were evaluated; a scenario that included halving protein overconsumption, livestock meat consumption, and food loss was estimated to reduce the food N footprint by 31%. Thus, there is room for reducing consumer-induced N loads to the environment. Campaigns aimed at boosting healthy and environmentally friendly diets should consider the diverse consumption patterns of different sex and age classes

    Association between Hardness (Difficulty of Chewing) of the Habitual Diet and Premenstrual Symptoms in Young Japanese Women

    Recent evidence suggests that voluntary rhythmic movements such as chewing may increase blood serotonin and subsequently brain serotonin, which in turn acts to alleviate premenstrual symptoms. In this observational cross-sectional study, we tested the hypothesis that hardness (difficulty of chewing) of the habitual diet (i.e. dietary hardness) is associated with decreased premenstrual symptoms. Subjects were 640 female Japanese dietetic students aged 18–22 years. Dietary hardness was assessed as an estimate of masticatory muscle activity for the habitual diet (i.e. the difficulty of chewing the food). The consumption of a total of 107 foods was estimated by means of a self-administered, comprehensive diet history questionnaire, and masticatory muscle activity during the ingestion of these foods was estimated according to published equations. Menstrual cycle symptoms were assessed using the retrospective version of the Moos Menstrual Distress Questionnaire, from which total score and subscale scores (i.e. pain, concentration, behavioral change, autonomic reactions, water retention, and negative affect) in the premenstrual phase were calculated and expressed as percentages relative to those in the intermenstrual phase. Dietary hardness was not associated with total score in the premenstrual phase (P for trend = 0.48). Further, no association was seen for any subscale score in the premenstrual phase (P for trend = 0.18–0.91). In conclusion, this preliminary study failed to substantiate a hypothesized inverse relationship between hardness of the habitual diet and premenstrual symptoms. Considering the plausibility of the putative mechanism, however, further investigation using more relevant measures of chewing and premenstrual symptoms is warranted

    Transcriptional Repression of Cdc25B by IER5 Inhibits the Proliferation of Leukemic Progenitor Cells through NF-YB and p300 in Acute Myeloid Leukemia

    The immediately-early response gene 5 (IER5) has been reported to be induced by γ-ray irradiation and to play a role in the induction of cell death caused by radiation. We previously identified IER5 as one of the 2,3,4-tribromo-3-methyl-1-phenylphospholane 1-oxide (TMPP)-induced transcriptional responses in AML cells, using microarrays that encompassed the entire human genome. However, the biochemical pathway and mechanisms of IER5 function in regulation of the cell cycle remain unclear. In this study, we investigated the involvement of IER5 in the cell cycle and in cell proliferation of acute myeloid leukemia (AML) cells. We found that the over-expression of IER5 in AML cell lines and in AML-derived ALDHhi (High Aldehyde Dehydrogenase activity)/CD34+ cells inhibited their proliferation compared to control cells, through induction of G2/M cell cycle arrest and a decrease in Cdc25B expression. Moreover, the over-expression of IER5 reduced colony formation of AML-derived ALDHhi/CD34+ cells due to a decrease in Cdc25B expression. In addition, over-expression of Cdc25B restored TMPP inhibitory effects on colony formation in IER5-suppressed AML-derived ALDHhi/CD34+ cells. Furthermore, the IER5 reduced Cdc25B mRNA expression through direct binding to Cdc25B promoter and mediated its transcriptional attenuation through NF-YB and p300 transcriptinal factors. In summary, we found that transcriptional repression mediated by IER5 regulates Cdc25B expression levels via the release of NF-YB and p300 in AML-derived ALDHhi/CD34+ cells, resulting in inhibition of AML progenitor cell proliferation through modulation of cell cycle. Thus, the induction of IER5 expression represents an attractive target for AML therapy

    Strategies of community health activities to improve the quality of inhabitants' life

    ヘルスプロモーションの理念にそって,住民組織,行政,研究グループが協働して,住民の生活の質の向上をめざした地域保健活動の展開を計画した。まず,休眠状態にあった岡山県M町の健康づくり推進協議会(以下M町協議会と略す)を活性化するため,協議会の委員18人を対象に聴き取り調査を行い,次に,M町協議会でのディスカッションを深めた。1.M町協議会構成団体のうち住民組織は9組織で,このうち6組織は居住地域内の住民の推薦や輪番で代表を選出しており,他の3組織は任意であった。8組織の事務局は行政機関内にあり,活動経費の公費補助率は70%以上であった。2.委員の描く理想の町のイメージとして,福祉の充実した町で老後も安心できるという内容を含んだ回答が多かった。それを実現する為に,住民一人一人が自己管理意識を持つと同時に,協議会が支援的な活動を身近なところで展開することの必要性が挙げられた。3.M町協議会活動への委員の期待は大きく,協議会内での委員間の意思疎通も次第に深められた。事前に聴き取り調査を行ったこと等が効果的に影響したと考えられた。Along with the idea of health promotion, we planned the community health activities to improve the quality of inhabitants' life in cooperation with inhabitants, community organizations, local government and health professionals. In order to revitalize an inactive committee for health promotion of M town hearing surveys were conducted on the members of the committee. 1. Leaders of community organizations occupied 9 of 19 members of the committee. Leaders of 6 organizations out of 9 ones were decided by recommendation or rotation. Eight secretariats of those organizations were located in the governmental office. Seventy percent or more of activity costs of these organizations depends on assistance of local government. 2. Members of the committee imaged the town, which was healthy and filled with mind of welfare as an ideal town. They pointed out that it was required for the committee to perform support activities to realize an ideal town as well as for every inhabitant to have the consciousness of self-management. 3. Activities of the committee were much expected by members of the committee. Communications between members deepened gradually by repeating the meetings. Hearing survey on members also seemed to be much effective for those progress

    The renin–angiotensin system promotes arrhythmogenic substrates and lethal arrhythmias in mice with non-ischaemic cardiomyopathy

    [Aims]The progression of pathological left ventricular remodelling leads to cardiac dysfunction and contributes to the occurrence of malignant arrhythmias and sudden cardiac death. The underlying molecular mechanisms remain unclear, however. Our aim was to examine the role of the renin–angiotensin system (RAS) in the mechanism underlying arrhythmogenic cardiac remodelling using a transgenic mouse expressing a cardiac-specific dominant-negative form of neuron-restrictive silencer factor (dnNRSF-Tg). This mouse model exhibits progressive cardiac dysfunction leading to lethal arrhythmias. [Methods and results]Subcutaneous administration of aliskiren, a direct renin inhibitor, significantly suppressed the progression of pathological cardiac remodelling and improved survival among dnNRSF-Tg mice while reducing arrhythmogenicity. Genetic deletion of the angiotensin type 1a receptor (AT1aR) similarly suppressed cardiac remodelling and sudden death. In optical mapping analyses, spontaneous ventricular tachycardia (VT) and fibrillation (VF) initiated by breakthrough-type excitations originating from focal activation sites and maintained by functional re-entry were observed in dnNRSF-Tg hearts. Under constant pacing, dnNRSF-Tg hearts exhibited markedly slowed conduction velocity, which likely contributes to the arrhythmogenic substrate. Aliskiren treatment increased conduction velocity and reduced the incidence of sustained VT. These effects were associated with suppression of cardiac fibrosis and restoration of connexin 43 expression in dnNRSF-Tg ventricles. [Conclusion]Renin inhibition or genetic deletion of AT1aR suppresses pathological cardiac remodelling that leads to the generation of substrates maintaining VT/VF and reduces the occurrence of sudden death in dnNRSF-Tg mice. These findings demonstrate the significant contribution of RAS activation to the progression of arrhythmogenic substrates

    GALAXY CRUISE: Deep Insights into Interacting Galaxies in the Local Universe

    We present the first results from GALAXY CRUISE, a community (or citizen) science project based on data from the Hyper Suprime-Cam Subaru Strategic Program (HSC-SSP). The current paradigm of galaxy evolution suggests that galaxies grow hierarchically via mergers, but our observational understanding of the role of mergers is still limited. The data from HSC-SSP are ideally suited to improve our understanding with improved identifications of interacting galaxies thanks to the superb depth and image quality of HSC-SSP. We have launched a community science project, GALAXY CRUISE, in 2019 and collected over 2 million independent classifications of 20,686 galaxies at z < 0.2. We first characterize the accuracy of the participants' classifications and demonstrate that it surpasses previous studies based on shallower imaging data. We then investigate various aspects of interacting galaxies in detail. We show that there is a clear sign of enhanced activities of super massive black holes and star formation in interacting galaxies compared to those in isolated galaxies. The enhancement seems particularly strong for galaxies undergoing violent merger. We also show that the mass growth rate inferred from our results is roughly consistent with the observed evolution of the stellar mass function. The 2nd season of GALAXY CRUISE is currently under way and we conclude with future prospects. We make the morphological classification catalog used in this paper publicly available at the GALAXY CRUISE website, which will be particularly useful for machine-learning applications.Comment: 23 pages, 22 figures, PASJ in press.     Trapping of CDC42 C-terminal variants in the Golgi drives pyrin inflammasome hyperactivation

    CDC42-C末端異常症に於ける炎症病態を解明 --ゴルジ体への異常蓄積がパイリンインフラマソーム形成を過剰促進--. 京都大学プレスリリース. 2022-05-02.Mutations in the C-terminal region of the CDC42 gene cause severe neonatal-onset autoinflammation. Effectiveness of IL-1β–blocking therapy indicates that the pathology involves abnormal inflammasome activation; however, the mechanism underlying autoinflammation remains to be elucidated. Using induced-pluripotent stem cells established from patients carrying CDC42[R186C], we found that patient-derived cells secreted larger amounts of IL-1β in response to pyrin-activating stimuli. Aberrant palmitoylation and localization of CDC42[R186C] protein to the Golgi apparatus promoted pyrin inflammasome assembly downstream of pyrin dephosphorylation. Aberrant subcellular localization was the common pathological feature shared by CDC42 C-terminal variants with inflammatory phenotypes, including CDC42[*192C*24] that also localizes to the Golgi apparatus. Furthermore, the level of pyrin inflammasome overactivation paralleled that of mutant protein accumulation in the Golgi apparatus, but not that of the mutant GTPase activity. These results reveal an unexpected association between CDC42 subcellular localization and pyrin inflammasome activation that could pave the way for elucidating the mechanism of pyrin inflammasome formation
