The future of molecular dynamics simulations in drug discovery
Molecular dynamics simulations can now track rapid processes—those occurring in less than about a millisecond—at atomic resolution for many biologically relevant systems. These simulations appear poised to exert a significant impact on how new drugs are found, perhaps even transforming the very process of drug discovery. We predict here future results we can expect from, and enhancements we need to make in, molecular dynamics simulations over the coming 25 years, and in so doing set out several Grand Challenges for the field. In the context of the problems now facing the pharmaceutical industry, we ask how we can best address drug discovery needs of the next quarter century using molecular dynamics simulations, and we suggest some possible approaches
Indications for liver transplantation in the cyclosporine era
One hundred seventy orthotopic liver transplants were performed under conventional immunosuppression with azathioprine and steroids with 1- and 5-year survivals of 32.9% and 20.0%, respectively. Since the introduction of cyclosporine-prednisone therapy in March 1980, 313 primary orthotopic liver transplants have been performed. Actuarial survivals at 1 and 5 years have improved to 69.7% and 62.8%, respectively. Biliary atresia is now the most common indication for liver replacement. In adults, primary biliary cirrhosis and sclerosing cholangitis have become more common indications for transplantation, and alcoholic cirrhosis and primary liver malignancy as indications have declined. Early enthusiasm for liver transplantation in patients with hepatic cancer has been tempered by the finding that recurrence is both common and rapid. An increasing number of patients with inborn errors of metabolism originating in the liver are receiving transplants, including patients with Wilson's disease, tyrosinemia, alpha-1-antitrypsin deficiency, glycogen storage disease, familial hypercholesterolemia, and hemochromatosis. Survival in this group of patients has been excellent (74.4% at 1 and 5 years). A hemophiliac who received a transplant for postnecrotic cirrhosis has survived and may have been cured of his hemophilia. About 20% of patients require retransplantation for rejection, technical failure, or primary graft failure. Only 4 of the patients receiving retransplants under conventional immunosuppression survived beyond 6 months, and all died within 14 months of retransplantation. Sixty-eight patients have received retransplants under cyclosporine-prednisone. Thirty-one patients are surviving, all for at least 1 year. Six of the 12 patients requiring a third transplant are alive 2 to 3 years after the primary operation. An aggressive approach to retransplantation in the patient with a failed graft is justified
The thermodynamics of prediction
A system responding to a stochastic driving signal can be interpreted as
computing, by means of its dynamics, an implicit model of the environmental
variables. The system's state retains information about past environmental
fluctuations, and a fraction of this information is predictive of future ones.
The remaining nonpredictive information reflects model complexity that does not
improve predictive power, and thus represents the ineffectiveness of the model.
We expose the fundamental equivalence between this model inefficiency and
thermodynamic inefficiency, measured by dissipation. Our results hold
arbitrarily far from thermodynamic equilibrium and are applicable to a wide
range of systems, including biomolecular machines. They highlight a profound
connection between the effective use of information and efficient thermodynamic
operation: any system constructed to keep memory about its environment and to
operate with maximal energetic efficiency has to be predictive.Comment: 5 pages, 1 figur
(Super)twistors and (super)strings
The Lagrangian formulation of the D=4 bosonic string and superstring in terms
of the (super)twistors is considered. The (super)twistor form of the equations
of motion is derived and the kappa-symmetry transformation for the
supertwistors is given. It is shown that the covariant kappa-symmetry gauge

Optical Spectroscopy of Bright Fermi LAT Blazars
We report on HET and Palomar 5 m spectroscopy of recently identified
-ray blazars in the {\it Fermi} LAT Bright Source List. These data
provide identifications for 10 newly discovered -ray flat spectrum
radio quasars (FSRQ) and six new BL Lacs plus improved spectroscopy for six
additional BL Lacs. We substantially improve the identification completeness of
the bright LAT blazars and give new redshifts and constraints, new

Parallel algorithm with spectral convergence for nonlinear integro-differential equations
We discuss a numerical algorithm for solving nonlinear integro-differential
equations, and illustrate our findings for the particular case of Volterra type
equations. The algorithm combines a perturbation approach meant to render a
linearized version of the problem and a spectral method where unknown functions
are expanded in terms of Chebyshev polynomials (El-gendi's method). This
approach is shown to be suitable for the calculation of two-point Green
functions required in next to leading order studies of time-dependent quantum

A New History and Discussion of 180-Day Exclusivity
This is the third in a series of articles on 180-day exclusivity. The first article traced the history of 180-day exclusivity from 1984 through its amendment in 2003 and court cases in 2004. A second article, published by two of the authors in 2007, updated the earlier piece through the end of 2006 but was arranged by issue rather than in a chronology. This article, which includes a third author, provides a comprehensive resource on 180-day exclusivity for old abbreviated new drug applications (ANDAs) (but less detail in some places where the 2007 article may be referenced) and focuses more discussion on the new provisions as well as some policy and legal issues related to 180-day exclusivity
All Who Wander: On the Prevalence and Characteristics of Multi-community Engagement
Although analyzing user behavior within individual communities is an active
and rich research domain, people usually interact with multiple communities
both on- and off-line. How do users act in such multi-community environments?
Although there are a host of intriguing aspects to this question, it has
received much less attention in the research community in comparison to the
intra-community case. In this paper, we examine three aspects of
multi-community engagement: the sequence of communities that users post to, the
language that users employ in those communities, and the feedback that users
receive, using longitudinal posting behavior on Reddit as our main data source,
and DBLP for auxiliary experiments. We also demonstrate the effectiveness of
features drawn from these aspects in predicting users' future level of
activity.
One might expect that a user's trajectory mimics the "settling-down" process
in real life: an initial exploration of sub-communities before settling down
into a few niches. However, we find that the users in our data continually post
in new communities; moreover, as time goes on, they post increasingly evenly
among a more diverse set of smaller communities. Interestingly, it seems that
users that eventually leave the community are "destined" to do so from the very
beginning, in the sense of showing significantly different "wandering" patterns
very early on in their trajectories; this finding has potentially important
design implications for community maintainers. Our multi-community perspective
also allows us to investigate the "situation vs. personality" debate from
