    Quantitative phase microscopy for the study of electromotility in living cells

These intrinsic changes can be used as an indicator for cell electric activity, but, to our knowledge, the intrinsic signal of electric activity has never been detected in single vertebrate cells. We describe here our development of a quantitative phase microscopy technique that is capable of detecting the intrinsic changes induced by electric activity in a human cell line. Chapter 1 provides introductory material regarding cellular electrophysiology and a review of the literature on the intrinsic signal of cell electric activity. This chapter also briefly introduces the quantitative phase microscope. In Chapter 2, we discuss our pilot studies and introduce the electromotility of prestinexpressing HEK293 cells as a test system. We describe our design of an effective optical detection scheme based on quantitative phase imaging and frequency domain detection which provides full-field, high resolution, high sensitivity, quantitative detection of electrically induced optical signals in cells. In Chapter 3, we demonstrate an improved quantitative phase microscope based on low-coherence interferometry with enhanced sensitivity and lower noise. We successfully acquired images of the intrinsic optical signal from electrically stimulated single HEK293 cells. In Chapter 4, we characterized the electrochemical properties and dynamic properties of the intrinsic optical signal. We argue that the signal is generated through the electromechanical coupling mechanism called membrane electromotility (MEM). Using the MEM signal as an indicator of membrane electric activity, we imaged the propagation of an applied potential in a network of cells in Chapter 5. Our research shows that high resolution quantitative phase imaging is a powerful tool that can provide significant insight into the underlying mechanism of cellular intrinsic optical signal of electric activity.     Service-Learning in Language for Specific Purposes: A Case of Korean Language Practicum

    While service-learning has been greatly acknowledged as an effective instructional tool in education for the past two decades, it has not been extensively applied to the Korean classroom and Language for Specific Purposes (LSP) settings as of yet. The practical challenges of community-involved service-learning for the less commonly taught language (LCTL) programs such as Korean are discussed. Moreover, community engagement through the campus community combined with the language teaching practicum is suggested for service-learning as an alternative to address the challenges. This study explores the curriculum design of a service-learning for a small language group and offers practical ideas on how to expose the students to campus communities, thereby providing them with cross-cultural insights and language practicum competence that extend beyond classroom language learning. The outcome supports that the campus community engagement effectively provides a hands-on opportunity for the students to practice not only their academics but also their leadership skills through the experience of designing their own service details in collaboration with their campus community partners

    Metabolic Syndrome Prediction Using Machine Learning Models with Genetic and Clinical Information from a Nonobese Healthy Population

    The prevalence of metabolic syndrome (MS) in the nonobese population is not low. However, the identification and risk mitigation of MS are not easy in this population. We aimed to develop an MS prediction model using genetic and clinical factors of nonobese Koreans through machine learning methods. A prediction model for MS was designed for a nonobese population using clinical and genetic polymorphism information with five machine learning algorithms, including naïve Bayes classification (NB). The analysis was performed in two stages (training and test sets). Model A was designed with only clinical information (age, sex, body mass index, smoking status, alcohol consumption status, and exercise status), and for model B, genetic information (for 10 polymorphisms) was added to model A. Of the 7,502 nonobese participants, 647 (8.6%) had MS. In the test set analysis, for the maximum sensitivity criterion, NB showed the highest sensitivity: 0.38 for model A and 0.42 for model B. The specificity of NB was 0.79 for model A and 0.80 for model B. In a comparison of the performances of models A and B by NB, model B (area under the receiver operating characteristic curve [AUC] = 0.69, clinical and genetic information input) showed better performance than model A (AUC = 0.65, clinical information only input). We designed a prediction model for MS in a nonobese population using clinical and genetic information. With this model, we might convince nonobese MS individuals to undergo health checks and adopt behaviors associated with a preventive lifestyle

    The Impact of Overactive Bladder on Health-Related Quality of Life, Sexual Life and Psychological Health in Korea

    Purpose We aimed to estimate the prevalence of overactive bladder (OAB) in Korea, to assess the variation in prevalence by sex and age, and to measure the impact of OAB on quality of life. Methods A population-based, cross-sectional telephone survey was conducted between April and June 2010 with a questionnaire regarding the prevalence of OAB, demographics, and the impact of OAB on quality of life. A geographically stratified random sample of men and women aged ≥30 years was selected. Results The overall prevalence of OAB was 22.9% (male, 19%; female, 26.8%). Of a total of 458 participants with OAB, 37.6% and 19.9% reported moderate or severe impact on their daily life and sexual life (5.6% and 3.5%, respectively, in participants without OAB). Anxiety and depression were reported by 22.7% and 39.3% of participants with OAB, respectively (9.7% and 22.8%, respectively, in participants without OAB). Only 19.7% of participants with OAB had consulted a doctor for their voiding symptoms, but 50.7% of respondents with OAB were willing to visit a hospital for the management of their OAB symptoms. Conclusions This study confirmed that OAB symptoms are highly prevalent in Korea, and many sufferers appear to have actively sought medical help. OAB has severe effects on daily and sexual life as well as psychological health

    Effects of a radiation dose reduction strategy for computed tomography in severely injured trauma patients in the emergency department: an observational study

    <p>Abstract</p> <p>Background</p> <p>Severely injured trauma patients are exposed to clinically significant radiation doses from computed tomography (CT) imaging in the emergency department. Moreover, this radiation exposure is associated with an increased risk of cancer. The purpose of this study was to determine some effects of a radiation dose reduction strategy for CT in severely injured trauma patients in the emergency department.</p> <p>Methods</p> <p>We implemented the radiation dose reduction strategy in May 2009. A prospective observational study design was used to collect data from patients who met the inclusion criteria during this one year study (intervention group) from May 2009 to April 2010. The prospective data were compared with data collected retrospectively for one year prior to the implementation of the radiation dose reduction strategy (control group). By comparison of the cumulative effective dose and the number of CT examinations in the two groups, we evaluated effects of a radiation dose reduction strategy. All the patients met the institutional adult trauma team activation criteria. The radiation doses calculated by the CT scanner were converted to effective doses by multiplication by a conversion coefficient.</p> <p>Results</p> <p>A total of 118 patients were included in this study. Among them, 33 were admitted before May 2009 (control group), and 85 were admitted after May 2009 (intervention group). There were no significant differences between the two groups regarding baseline characteristics, such as injury severity and mortality. Additionally, there was no difference between the two groups in the mean number of total CT examinations per patient (4.8 vs. 4.5, respectively; p = 0.227). However, the mean effective dose of the total CT examinations per patient significantly decreased from 78.71 mSv to 29.50 mSv (p < 0.001).</p> <p>Conclusions</p> <p>The radiation dose reduction strategy for CT in severely injured trauma patients effectively decreased the cumulative effective dose of the total CT examinations in the emergency department. But not effectively decreased the number of CT examinations.</p

    Struvite Crystallization of Anaerobic Digestive Fluid of Swine Manure Containing Highly Concentrated Nitrogen

    In this study, the optimal operation factors for struvite crystallization for removing and recovering nitrogen and phosphorus from anaerobic digestive fluid of swine manure containing highly concentrated nitrogen was determined. Every experiment for the struvite crystallization reaction was conducted by placing 1,000 mL of digestion fluid in a 2,000 mL Erlenmeyer flask at various temperatures, pH, and mixing speed. Except for special circumstances, the digestion fluid was centrifuged (10,000 rpm, 10 min) and then the supernatant was used for the experiment at room temperature and 100 rpm. The optimal mole ratio of PO43−:Mg2+ was 1:1.5, and the pH effect ranging from 9 to 11 was similar, when mixed for 1 hour. Under this condition, the removal efficiency of NH4+-N and PO43−-P was 40% and 88.6%, respectively. X-shaped crystal was observed by light and scanning electron microscopy.     Effect of RNA Interference-Mediated Suppression of p75 on the Viability of Rat Notochordal Cells

    Study DesignIn vitro cell culture model.PurposeTo investigate the effects of RNA interference (RNAi) on p75 expression and viability of rat notochordal cells treated with serum deprivation.Overview of LiteratureRNAi enables the inhibition of specific genes by sequence-specific gene silencing using a double-stranded RNA.MethodsNotochordal cells were isolated, cultured, and placed in 10% (control) or 0% (apoptosis-promoting) fetal bovine serum (FBS) for 48 hours. The expression of p75, apoptosis, and cell proliferation were determined. To suppress p75 expression, a small interfering RNA (siRNA) was synthesized against p75 (p75 siRNA) and transfected into cells. The suppression of p75 mRNA expression was investigated using the reverse transcription-polymerase chain reaction. The degree of p75 suppression was semiquantitatively analyzed using densitometry. The effect of p75 siRNA on apoptosis and proliferation of cells was determined. Solutions of an unrelated siRNA and transfection agent alone served as controls.ResultsSerum deprivation significantly increased apoptosis by 40.3%, decreased proliferation of notochordal cells by 45.3% (both, p<0.001), and upregulated p75 expression. The p75 siRNA suppressed p75 expression in cells cultured in 0% FBS. The rate of suppression by p75 siRNA of p75 mRNA was 72.9% (p<0.001). Suppression of p75 expression by p75 siRNA inhibited apoptosis by 7% and increased proliferation by 14% in cells cultured in 0% FBS (both, p<0.05).ConclusionssiRNA-mediated suppression of p75 inhibited apoptosis and increased proliferation of notochordal cells under conditions of serum deprivation, suggesting that RNAi might serve as a novel therapeutic approach for disc degeneration caused by insufficient viability of disc cells through the suppression of the expression of harmful genes
