    Exploring the practical knowledge of eccentric resistance training in high-performance strength and conditioning practitioners

    Habitual use of eccentric exercise has been recognised to increase strength and power; however, the current body of knowledge has limited potential to understand the application of such resistance training in athletic populations. In order to develop appropriate applied research, that relates to elite athletic populations, it is vital to appreciate the practical knowledge of strength and conditioning practitioners operating in high-performance environments. This study summarised the questionnaire responses from 100 strength and conditioning practitioners operating in performance sport relating to questions such as the training effects to various eccentric resistance training regimes, the rationale for the use of these techniques and the knowledge supporting its application. The combination of closed and open-ended questions enabled a thematic analysis to be conducted. There was evidence that practitioners employed a variety of eccentric training methodologies; however, there was interest in gaining greater understanding of the training dose to bring about the optimal adaptive changes, and importantly how this might translate to sport-specific performance. In addition, practitioners would welcome recommendations associated with eccentric training, whilst concurrently minimising the issues of excessive fatigue, muscle damage and soreness. The training effects of interest included neural, architectural and morphological adaptations and, importantly, translation to performance of sports-specific skills. Collectively, these responses called for more practically relevant research to be conducted within the high-performance environment, alongside more opportunities for professional development through learning and knowledge-sharing opportunities. The outcomes summarised in this work should inform future applied research projects and educational content relating to eccentric training

    Drug transport mechanism of P-glycoprotein monitored by single molecule fluorescence resonance energy transfer

    In this work we monitor the catalytic mechanism of P-glycoprotein (Pgp) using single-molecule fluorescence resonance energy transfer (FRET). Pgp, a member of the ATP binding cassette family of transport proteins, is found in the plasma membrane of animal cells where it is involved in the ATP hydrolysis driven export of hydrophobic molecules. When expressed in the plasma membrane of cancer cells, the transport activity of Pgp can lead to the failure of chemotherapy by excluding the mostly hydrophobic drugs from the interior of the cell. Despite ongoing effort, the catalytic mechanism by which Pgp couples MgATP binding and hydrolysis to translocation of drug molecules across the lipid bilayer is poorly understood. Using site directed mutagenesis, we have introduced cysteine residues for fluorescence labeling into different regions of the nucleotide binding domains (NBDs) of Pgp. Double-labeled single Pgp molecules showed fluctuating FRET efficiencies during drug stimulated ATP hydrolysis suggesting that the NBDs undergo significant movements during catalysis. Duty cycle-optimized alternating laser excitation (DCO-ALEX) is applied to minimize FRET artifacts and to select the appropriate molecules. The data show that Pgp is a highly dynamic enzyme that appears to fluctuate between at least two major conformations during steady state turnover.Comment: 10 pages, 7 figure

    AN EXAMINATION OF MANAGERIAL STRATEGIES FOR INCREASING INFORMATION TECHNOLOGY PENETRATION IN ORGANIZATIONS

    This paper describes an empirical study comparing two competing perspectives for explaining information technology (IT) penetration in organizations. IT penetration is defined as the extent to which IT is embedded within an organization\u27s strategic, managerial, and operational work systems. With the first perspective, IT penetration is hypothesized to be related to the implementation of a comprehensive set of management processes for the information systems (IS) function. With the second perspective, IT penetration is hypothesized to be related to the effectiveness of IT-related interactions among an organization\u27s managers, To strengthen the study\u27s research design, the hypotheses are examined separately across two samples of organizations. Senior ]S executives completed the study\u27s research instrument. Respondents in the first sample represented 132 large organizations across a variety of industries, while the respondents in the second sample represented 44 business units within a large, high teChnology firm. IT penetration and IT-related managerial interactions were measured at a subunit level, while IS management processes were measured at an organizational level.     The “Welcome to european stories” Grundtvig partnership aims to contribute to formal and informal learning of adult persons in the four European countries through the "Storytelling" educational tool. This project complies as an educational tool and as an educational strategy and motivation in the adult education. Stories are a way to build bridges between different cultures; they are carriers of cultural and historical knowledge. Storytelling techniques are an educational strategy and a pedagogical and motivational tool in order to improve adult or senior learning and in order to integrate them into the life. In this connection, we would like to use the potential of storytelling as an educational tool for the formal or informal adult education. We think that the power of storytelling will enhance the adults and senior´s life. It is a strong tool for empowerment and social inclusion. It has a great potential to attract and motivate adults or seniors to learn, to connect with others, to express themselves and integrate in the society. The partnership is addressed to adults who left the school early (or many migrants who never had the possibility to receive a school education), to the ageing population, to senior citizens and to other related organisations with adult education
