    Persaingan yang semakin ketat menuntut para pelaku bisnis untuk memaksimalkan kinerjanya melalui kualitas produk, harga, dan kualitas pelayanan untuk dapat bertahan dalam persaingan bisnis dan mampu menciptakan kepuasan pelanggan. Tujuan penelitian ini adalah untuk mengetahui pengaruh kualitas produk, harga, dan kualitas pelayanan terhadap kepuasan pengguna Cafe dan Resto Cabana Manado. Jenis penelitian yang digunakan adalah penelitian asosiatif dan tekhnik analisis menggunakan regresi linear berganda. Hasil penelitian menunjukan bahwa kualitas produk, harga dan kualitas pelayanan berpengaruh positif terhadap kepuasan pengguna Cafe dan Resto Cabana Manado. Kualitas produk berpengaruh namun tidak signifikan terhadap kepuasan pengguna. Harga dan kualitas pelayanan berpengaruh signifikan terhadap kepuasan pengguna. Manajemen Cafe dan Resto Cabana Manado sebaiknya meningkatkan kualitas produk agar kepuasaan pengguna dapat selalu terpenuhi

    Protein Expression, Characterization and Activity Comparisons of Wild Type and Mutant DUSP5 Proteins

    Background The mitogen-activated protein kinases (MAPKs) pathway is critical for cellular signaling, and proteins such as phosphatases that regulate this pathway are important for normal tissue development. Based on our previous work on dual specificity phosphatase-5 (DUSP5), and its role in embryonic vascular development and disease, we hypothesized that mutations in DUSP5 will affect its function. Results In this study, we tested this hypothesis by generating full-length glutathione-S-transferase-tagged DUSP5 and serine 147 proline mutant (S147P) proteins from bacteria. Light scattering analysis, circular dichroism, enzymatic assays and molecular modeling approaches have been performed to extensively characterize the protein form and function. We demonstrate that both proteins are active and, interestingly, the S147P protein is hypoactive as compared to the DUSP5 WT protein in two distinct biochemical substrate assays. Furthermore, due to the novel positioning of the S147P mutation, we utilize computational modeling to reconstruct full-length DUSP5 and S147P to predict a possible mechanism for the reduced activity of S147P. Conclusion Taken together, this is the first evidence of the generation and characterization of an active, full-length, mutant DUSP5 protein which will facilitate future structure-function and drug development-based studies

    The major purpose of the Papua New Guinea Agricultural Systems Project is to produce information on small holder (subsistence) agriculture at provincial and national levels (Allen et al 1995). Information was collected by field observation, interviews with villagers and reference to published and unpublished documents. Methods are described by Bourke et al. (1993). This Working Paper contains a written summary of the information on the Agricultural Systems in this Province, maps of the location of agriculture systems, a complete listing of all information in the database in coded form, and lists of villages with National Population Census codes, indexed by agricultural systems. This information is available as a map-linked database (GIS) suitable for use on a personal computer in ESRI and MapInfo formats. An Agricultural System is identified when a set of similar agricultural crops and practices occur within a defined area. Six criteria are used to distinguish one system from another: 1. Fallow type (the vegetation which is cleared from a garden site before cultivation). 2. Fallow period (the length of time a garden site is left unused between cultivations). 3. Cultivation intensity (the number of consecutive crops planted before fallow). 4. The staple, or most important, crops. 5. Garden and crop segregation (the extent to which crops are planted in separate gardens; in separate areas within a garden; or are planted sequentially). 6. Soil fertility maintenance techniques (other than natural regrowth fallows). Where one or more of these factors differs significantly and the differences can be mapped, then a separate system is distinguished. Where variation occurs, but is not able to be mapped at 1:500 000 scale because the areas in which the variation occurs are too small or are widely dispersed within the larger system, a subsystem is identified. Subsystems within an Agricultural System are allocated a separate record in the database, identified by the Agricultural System number and a subsystem number. Sago is a widespread staple food in lowland Papua New Guinea. Sago is produced from palms which are not grown in gardens. Most of the criteria above cannot be applied. In this case, systems are differentiated on the basis of the staple crops only.     Manajemen Perusahaan otomotif saat ini sedang berupaya untuk meningkatkan penjualan kendaraan bermotor kepada para konsumen untuk itu potensi pasar dan pembelian menjadi sangat penting bagi Perusahaan. Melalui informasi yang positif dari konsumen, iklan yang menarik perhatian dengan kreatifitas dari Perusahaan, harga yang dapat dijangkau dengan kualitas dari produk yang tinggi tidak menutup kemungkinan akan menarik keinginan membeli dari konsumen. Tujuan penelitian ini untuk mengetahui pengaruh Word of Mouth, Daya Tarik Iklan, Persepsi Harga dan Kualitas Produk terhadap Minat Beli motor Suzuki Satria Fu di PT. Sinar Galesong Mandiri. Metode yang digunakan adalah asosiatif. Sampel yang digunakan sebanyak 97 responden. Hasil penelitian menunjukkan bahwa secara simultan Word of Mouth, Daya Tarik Iklan, Persepsi Harga dan Kualitas Produk mempunyai pengaruh positif dan signifikan terhadap Minat Beli konsumen. Secara parsial Word of Mouth, Daya Tarik Iklan dan Kualitas Produk mempunyai pengaruh yang positif terhadap Minat Beli akan tetapi Persepi Harga mempunyai pengaruh yang negatif terhadap Minat Beli konsumen. Manajemen PT. Sinar Galesong Mandiri sebaiknya memperhatikan penetapan harga pada produk karena hasil penelitian menunjukkan harga bagi responden masih kurang kompetitif.     Variability of dinoflagellates and their associated toxins in relation with environmental drivers in Ambon Bay, eastern Indonesia

    The aim of the present work was to unravel which environmental drivers govern the dynamics of toxic dinoflagellate abundance as well as their associated paralytic shellfish toxins (PSTs), diarrhetic shellfish toxins (DSTs) and pectenotoxin-2 (PTX2) in Ambon Bay, Eastern Indonesia. Weather, biological and physicochemical parameters were investigated weekly over a 7-month period. Both PSTs and PTX2 were detected at low levels, yet they persisted throughout the research. Meanwhile, DSTs were absent. A strong correlation was found between total particulate PST and Gymnodinium catenatum cell abundance, implying that this species was the main producer of this toxin. PTX2 was positively correlated with Dinophysis miles cell abundance. Vertical mixing, tidal elevation and irradiance attenuation were the main environmental factors that regulated both toxins and cell abundances, while nutrients showed only weak correlations. The present study indicates that dinoflagellate toxins form a potential environmental, economic and health risk in this Eastern Indonesian bay
