    Is Body Fat a Predictor of Race Time in Female Long-Distance Inline Skaters?

    Purpose: The aim of this study was to evaluate predictor variables of race time in female ultra-endurance inliners in the longest inline race in Europe. Methods: We investigated the association between anthropometric and training characteristics and race time for 16 female ultraendurance inline skaters, at the longest inline marathon in Europe, the ‘Inline One-eleven’ over 111 km in Switzerland, using bi- and multivariate analysis. Results: The mean (SD) race time was 289.7 (54.6) min. The bivariate analysis showed that body height (r=0.61), length of leg (r=0.61), number of weekly inline skating training sessions (r=-0.51)and duration of each training unit (r=0.61) were significantly correlated with race time. Stepwise multiple regressions revealed that body height, duration of each training unit, and age were the best variables to predict race time. Conclusion: Race time in ultra-endurance inline races such as the ‘Inline One-eleven’ over 111 km might be predicted by the following equation (r2 = 0.65): Race time (min) = -691.62 + 521.71 (body height, m) + 0.58 (duration of each training unit, min) + 1.78 (age, yrs) for female ultra-endurance inline skaters

    Phosphorylated tyrosine-containing proteins in primary lung cancer correlates with proliferation and prognosis

    To determine the usefulness of tyrosine phosphorylation in evaluating biological characteristics, we attempted to evaluate the relationship between the amount of phosphorylated tyrosine-containing proteins and clinicopathological factors, cell proliferation and outcome in non-small cell lung cancer. To evaluate phosphorylated tyrosine-containing proteins we used 96 surgically resected materials of non-small cell lung cancer and normal peripheral lung, while immunohistochemical evaluation was performed. Cell proliferating ability was evaluated using the labelling index of proliferating cell nuclear antigen-positive nuclear staining cells. There were statistically significant differences between the expression levels of phosphorylated tyrosine-containing proteins of normal and cancerous tissues (P<0.0001). Evaluations based on clinicopathological factors apart from histopathological differentiation, showed no statistically significant differences of phosphorylated tyrosine-containing proteins expression. However, phosphorylated tyrosine-containing proteins correlated with cell proliferation activity evaluated (P(Low, High)<0.0001; P(Low, Int) <0.0001; P(Int, High)<0.0001). Furthermore, non-small cell lung cancer cases with high expression and intermediate expression of phosphorylated tyrosine-containing proteins had a significantly shorter disease-free postoperative survival than those with low expression of phosphorylated tyrosine-containing proteins using log-rank analysis (P(Low, Int) <0.0028; P(Low, High)=0.0002). Furthermore, phosphorylated tyrosine-containing proteins expression level statistically contributed to disease-free survival in Cox's proportional hazard model. Therefore, phosphorylated tyrosine-containing proteins in non-small cell lung cancer tissues seem to reflect its biological malignancy, and this evaluation may be valuable for constructing the most appropriate therapeutic strategy

    Retroviral expression of a kinase-defective IGF-I receptor suppresses growth and causes apoptosis of CHO and U87 cells in-vivo

    BACKGROUND: Phosphatidylinositol-3,4,5-triphosphate (PtdInsP3) signaling is elevated in many tumors due to loss of the tumor suppressor PTEN, and leads to constitutive activation of Akt, a kinase involved in cell survival. Reintroduction of PTEN in cells suppresses transformation and tumorigenicity. While this approach works in-vitro, it may prove difficult to achieve in-vivo. In this study, we investigated whether inhibition of growth factor signaling would have the same effect as re-expression of PTEN. METHODS: Dominant negative IGF-I receptors were expressed in CHO and U87 cells by retroviral infection. Cell proliferation, transformation and tumor formation in athymic nude mice were assessed. RESULTS: Inhibition of IGF-IR signaling in a CHO cell model system by expression of a kinase-defective IGF-IR impairs proliferation, transformation and tumor growth. Reduction in tumor growth is associated with an increase in apoptosis in-vivo. The dominant-negative IGF-IRs also prevented growth of U87 PTEN-negative glioblastoma cells when injected into nude mice. Injection of an IGF-IR blocking antibody αIR3 into mice harboring parental U87 tumors inhibits tumor growth and increases apoptosis. CONCLUSION: Inhibition of an upstream growth factor signal prevents tumor growth of the U87 PTEN-deficient glioma to the same extent as re-introduction of PTEN. This result suggests that growth factor receptor inhibition may be an effective alternative therapy for PTEN-deficient tumors

    Promoting fit bodies, healthy eating and physical activity among Indigenous Australian men: a study protocol

    Background: Overall the physical health of Indigenous men is among the worst in Australia. Research has indicated that modifiable lifestyle factors, such as poor nutrition and physical inactivity, appear to contribute strongly to these poor health conditions. To effectively develop and implement strategies to improve the health of Australia&rsquo;s Indigenous peoples, a greater understanding is needed of how Indigenous men perceive health, and how they view and care for their bodies. Further, a more systematic understanding of how sociocultural factors affect their health attitudes and behaviours is needed. This article presents the study protocol of a communitybased investigation into the factors surrounding the health and body image of Indigenous Australian men.Methods and design: The study will be conducted in a collaborative manner with Indigenous Australian men using a participatory action research framework. Men will be recruited from three locations around Australia (metropolitan, regional, and rural) and interviewed to understand their experiences and perspectives on a number of issues related to health and health behaviour. The information that is collected will be analysed using modified grounded theory and thematic analysis. The results will then be used to develop and implement community events in each location to provide feedback on the findings to the community, promote health enhancing strategies, and determine future action and collaboration.Discussion: This study will explore both risk and protective factors that affect the health of Indigenous Australian men. This knowledge will be disseminated to the wider Indigenous community and can be used to inform future health promotion strategies. The expected outcome of this study is therefore an increased understanding of health and health change in Indigenous Australian men, the development of strategies that promote healthy eating and positive patterns of physical activity and, in the longer term, more effective and culturally-appropriate interventions to improve health.<br /

    Demand for Zn2+ in Acid-Secreting Gastric Mucosa and Its Requirement for Intracellular Ca2+

    Recent work has suggested that Zn(2+) plays a critical role in regulating acidity within the secretory compartments of isolated gastric glands. Here, we investigate the content, distribution and demand for Zn(2+) in gastric mucosa under baseline conditions and its regulation during secretory stimulation.Content and distribution of zinc were evaluated in sections of whole gastric mucosa using X-ray fluorescence microscopy. Significant stores of Zn(2+) were identified in neural elements of the muscularis, glandular areas enriched in parietal cells, and apical regions of the surface epithelium. In in vivo studies, extraction of the low abundance isotope, (70)Zn(2+), from the circulation was demonstrated in samples of mucosal tissue 24 hours or 72 hours after infusion (250 µg/kg). In in vitro studies, uptake of (70)Zn(2+) from media was demonstrated in isolated rabbit gastric glands following exposure to concentrations as low as 10 nM. In additional studies, demand of individual gastric parietal cells for Zn(2+) was monitored using the fluorescent zinc reporter, fluozin-3, by measuring increases in free intracellular concentrations of Zn(2+) {[Zn(2+)](i)} during exposure to standard extracellular concentrations of Zn(2+) (10 µM) for standard intervals of time. Under resting conditions, demand for extracellular Zn(2+) increased with exposure to secretagogues (forskolin, carbachol/histamine) and under conditions associated with increased intracellular Ca(2+) {[Ca(2+)](i)}. Uptake of Zn(2+) was abolished following removal of extracellular Ca(2+) or depletion of intracellular Ca(2+) stores, suggesting that demand for extracellular Zn(2+) increases and depends on influx of extracellular Ca(2+).This study is the first to characterize the content and distribution of Zn(2+) in an organ of the gastrointestinal tract. Our findings offer the novel interpretation, that Ca(2+) integrates basolateral demand for Zn(2+) with stimulation of secretion of HCl into the lumen of the gastric gland. Similar connections may be detectable in other secretory cells and tissues

    Dietary And Socio-Economic Correlates Of Nutritional Status In A Rural Adult Kenyan Population

    There is a lack of data on trends in body composition among developing countries and the distribution of malnutrition among adults is poorly understood. Thus, the objective was to establish nutritional status, demographic make-up, dietary habits, and to determine if socioeconomic variables or dietary habits are associated with nutritional status in a rural adult Kenyan population. Five hundred Kenyan adults ages 18-55 years were recruited from the Kombewa division, located 40 km west of Kisumu, Kenya. The proportion of underweight and overweight adults is consistent with developing countries in nutrition transition. Using standard body mass index (BMI) classifications, 9.8% of the study population was underweight, 76.0% were normal weight, and 14.2% were overweight or obese. Significantly more females (20.7%) were overweight or obese than males (4.1%) (P=0.001). Iron deficiency was a significant issue, with 21.4% of females classified as anemic (Hb<120g/l). Among overweight females, anemia rates remained high (16.9%) and were greater than in underweight females (10.8%). While 20% of subjects were HIV positive, their status did not affect BMI (P=0.230). The most significant correlation between dietary data and BMI was french fry consumption (r = 0.167, P=0.001). Overweight or obese subjects reported eating french fries more frequently than subjects who had an underweight or normal range BMI (64.8% vs. 43.8%, P=0.002). Several other socioeconomic and dietary variables had weak correlations with BMI. Similar to data from developing countries in nutrition transition, it appears that the nutritional status of this rural Kenyan population has been affected by shifts in dietary patterns. Based on trends in other developing countries, it is likely that the incidence of overweight will increase in Kenya while micronutrient deficiencies and underweight remain concerns. Socioeconomic variables have been correlated with nutritional status in other developing countries and should be examined further in conjunction with food insecurity, so that culturally relevant and feasible interventions can be implemented
