    TRECVid 2011 Experiments at Dublin City University

    This year the iAd-DCU team participated in three of the assigned TRECVid 2011 tasks; Semantic Indexing (SIN), Interactive Known-Item Search (KIS) and Multimedia Event Detection (MED). For the SIN task we presented three full runs using global features, local features and fusion of global, local features and relationships between concepts respectively. The evaluation results show that local features achieve better performance, with marginal gains found when introducing global features and relationships between concepts. With regard to our KIS submission, similar to our 2010 KIS experiments, we have implemented an iPad interface to a KIS video search tool. The aim of this year’s experimentation was to evaluate different display methodologies for KIS interaction. For this work, we integrate a clustering element for keyframes, which operates over MPEG-7 features using k-means clustering. In addition, we employ concept detection, not simply for search, but as a means of choosing most representative keyframes for ranked items. For our experiments we compare the baseline non-clustering system to a clustering system on a topic by topic basis. Finally, for the first time this year the iAd group at DCU has been involved in the MED Task. Two techniques are compared, employing low-level features directly and using concepts as intermediate representations. Evaluation results show promising initial results when performing event detection using concepts as intermediate representations

    Multi-chord fiber-coupled interferometer with a long coherence length laser

    This paper describes a 561 nm laser heterodyne interferometer that provides time-resolved measurements of line-integrated plasma electron density within the range of 10^15-10^18 cm^(-2). Such plasmas are produced by railguns on the Plasma Liner Experiment (PLX), which aims to produce \mu s-, cm-, and Mbar-scale plasmas through the merging of thirty plasma jets in a spherically convergent geometry. A long coherence length, 320 mW laser allows for a strong, sub-fringe phase-shift signal without the need for closely-matched probe and reference path lengths. Thus only one reference path is required for all eight probe paths, and an individual probe chord can be altered without altering the reference or other probe path lengths. Fiber-optic decoupling of the probe chord optics on the vacuum chamber from the rest of the system allows the probe paths to be easily altered to focus on different spatial regions of the plasma. We demonstrate that sub-fringe resolution capability allows the interferometer to operate down to line-integrated densities of order 10^15 cm^(-2).Comment: submitted to Rev. Sci. Instrum. (2011

    Pre-amplification methods for tracking low-grade Plasmodium falciparum populations during scaled-up interventions in Southern Zambia

    Background: Malaria is receding in many endemic countries with intervention scale -up against the disease. However, this resilient scourge may persist in low-grade submicroscopic infections among semi-immune members of the population, and be poised for possible resurgence, creating challenges for detection and assessment of intervention impact. Parasite genotyping methods, such as the molecular barcode, can identify specific malaria parasite types being transmitted and allow tracking and evaluation of parasite population structure changes as interventions are applied. This current study demonstrates application of pre-amplification methods for successful detection and genotyping of residual Plasmodium falciparum infections during a dramatic malarial decline. Methods: The study was a prospective cross-sectional design and based on a 2,000 sq km vicinity of Macha Mission Hospital in southern Zambia. Willing and predominantly asymptomatic residents of all ages were screened for malaria by microscopy during the 2005 and 2008 transmission seasons, with simultaneous collection of dried blood spots (DBS) on filter paper, and extraction of Plasmodium falciparum DNA was performed. Plasmodium falciparum infections were genotyped using a 24 SNP-based molecular barcode assay using real-time PCR. Submicroscopic parasitaemia samples were subjected to pre-amplification using TaqMan PreAmp Master Mix following the manufacturer’s instructions before SNP barcode analysis. Results: There was a dramatic decline of malaria between 2005 and 2008, and the geometric mean parasite density (95% CI) fell from 704/μL (390–1,271) in 2005 to 39/μL (23–68) in 2008, culminating in a large proportion of submicroscopic infections of which 90% failed to yield ample DNA for standard molecular characterization among 2008 samples. Pre-amplification enabled successful detection and genotyping of 74% of these low-grade reservoir infections, overall, compared to 54% that were detectable before pre-amplification (p <0.0005, n = 84). Furthermore, nine samples negative for parasites by microscopy and standard quantitative PCR amplification were positive after pre-amplification. Conclusions: Pre-amplification allows analysis of an otherwise undetectable parasite population and may be instrumental for parasites identification, tracking and assessing the impact of interventions on parasite populations during malaria control and elimination programmes when parasitaemia is expected to decline to submicroscopic levels
