    Changes of plasma lipids in relation to the regular consumption of bilberries (Vaccinium myrtillus L.)

    In this work we studied the impact of regular consumption of bilberries on the lipid profile and triglycerides in the blood plasma. The research involved 18 women (average age 49.7) and 7 men (average age 52.8), who consumed 150 g of bilberries three times a week for 6 weeks. Based on these results, we can conclude that the average total cholesterol levels of women decreased from 5.65 mmol.l-1 at the beginning of research to 5.11 mmol.l-1. At end of the study, the average total cholesterol was 5.47 mmol.l-1. Similar results were found in the LDL cholesterol - the level of cholesterol of the second blood collection dropped from 4.06 mmol.l-1 to 3.70 mmol.l-1 and at the end of  the study it increased again to 4.00 mmol.l-1. We observed a slight increase in HDL choleserol (by 0.07 mmol.l-1) and reduction of triglycerides (by 0.16 mmol.l-1). The total cholesterol monitored men due to the regular consumption of bilberries decreased by 0.36 mmol.l-1, LDL cholesterol by 0.31 mmol.l-1, HDL cholesterol by 0.49 mmol.l-1 and triglycerides by 0,49 mmol.l-1. The improvement of triglycerides and lipid profile in blood plasma of monitored subjects can by evaluated positively. The observed data confirm the biological activity of bilberries and the effectiveness of their use in prevention and comprehensive treatment of cardiovascular diseases

    ANALYSIS OF BIOLOGICALLY ACTIVE SUBSTANCES IN BILBERRY (VACCINIUM MYRTILLUS L.) IN SELECTED NATURAL LOCALITIES OF SLOVAK REPUBLIC

    The aim of this study was to analyze the association between enteritis and pneumonia during first months of life and subsequent growth and milk production traits of these animals. A total of 1327 calves were followed in this study. Calves were divided into three groups: Group A) healthy calves; Group B) calves that suffered from the enteritis one or more times from the birth to 6 th month of age; and Group C) calves that suffered from birth to 6th month of age from the pneumonia. Subsequently all animals were observed at the following age stages: calves, heifers and cows. For all groups (age stages) the body weight and daily weight gain in calves (in the breeding period from birth to 6th month of age), the weight and daily weight gain weight in heifers (in the breeding period from 7 th to 25 th month of age) and production parameters of cows in the first lactation (milk in kg, fat in kg and protein in kg) was recorded. Calves in Groups B and C exhibited lower average daily gain (690.40 g, SD = 186.18, N.S. and 636.80 g, SD = 221.19, P < 0.05 respectively) than healthy calves (Group A, 703.91 g, SD = 223.48). Heifers in Group B exhibited increased the daily weight gain (830.01, SD = 73.57, N.S.) compared to healthy animals (Group A, 828.31, SD = 89.16). The general hypothesis regarding the negative effect of the enteritis (in breeding age animals that suffer from disease) on growth in the Simmental breed has not been confirmed. Heifers that had suffered with the pneumonia (Group C) showed significantly lower average daily gain (784.64, SD = 102.96, P < 0.05) than heifers in group A and B. In the evaluation of milk production in the first lactation, average production of 5800 kg per lactation (SD = 3.96) was recorded in Group A cows. Cows in Group B in the first lactation produced the 5977 kg of milk per lactation on average (SD = 4.31). Cows from Group C in first lactation produced lowest amount of milk during the first lactation period (5674 kg, SD = 3.87), but difference among A, B and C group was not significant

    Healthy eating index and different fruit dietary habits in Slovak adult female

    The healthy index is a tool for evaluation of nutrition recommendation aimed at prevention of chronic diseases. A lot of studies have been devoted to HEI of different aged groups of people but dates about Slovak population have been still missed. The goal of the study was to evaluate the Healthy Eating Index (HEI) in nutrition of adult female, determine their components in relation of parameters of anthropometry and body composition. Secondly, the research work was also aimed at the comparison of partial score HEI among groups of female with different fruit intake. Daily nutrition was evaluated by 24 hours dietary recalls. In set of female (average age 31.1 ±9.1 years) the average HEI index reached up 53.0 ±8.8 points in accord with medium degree of diet, it means dietary improvement has been highly recommended. In respect of all assayed components of index the best results achieved the variety of diet (8.8 ±1.3 points) and the worst the natrium intake (0.7 ±2.0 points). The average score for the individual components pointed to neccesary of increase in the grain intake and vegetable, on the other hand the intake of cholesterol, saturated fats and especially natrium should be decreased. The differences between groups with various intake of fruit (with recommended intake and insufficient intake) in the rest 9 components have not been proved as significant. The occurrence of risk values of body index, body fats determined by bioelectric impedance and android risk based on circumference of hips can not be considered between groups as significant. Average HEI has been in significance correlation with age (r = 0.240; p <0.05), circumference of hips (r = 0.2312; p <0.05) and body weight r = 0.1748; p <0.05). Future studies have been needed to evaluate diet according to the HEI in different groups of population in Slovakia

    The comparative study of medicinal plants utilization as herbal antibiotics by college students

    The medicinal plant utilization has become more and more popular and increasing number of consumers prefer alternative medicine to synthetic antibiotic. Research dealing with evaluation of medicinal plant usage as herbal antibiotics including the sample of 584 quizzed college students aged 19 - 25 years (337 women, 217 men) originated from Slovak Republic (n = 338), Czech Republic (n = 112) and Hungary (n = 134). According to university and the study programme the following groups were evaluated: Constantine the Philosopher University CPU (PEES - Pre-school and elementary education in Slovak language, PEEH - Pre-school and elementary education in Hungarian language, BI - Biology, RT - Regional Tourism), Mendel University in Brno MU (H - Horticulture), Slovak University of Agriculture SUA (H - Horticulture), University of Pécs UP (PE - Physical education), Comenius University CU (PE - Physical education). The study was aimed at the evaluation of the significance of the country and the study programme for the use of the most commonly used herbs: plantain, elderberry, stinging nettle, ginger and coneflower (Echinacea). Our results showed that the choice of preferred medicinal plants as herbal antibiotics during illness had not been clearly influenced by country or field of study programme. Plantain was the most frequently used herb by students of UP/PE (51.5%), CPU/PEES and CPU/PEEH (47.9%; 41.1%). Elderberry was the most popular herb among the students CPU/BI (52.9%), CPU/RT and SUA/H (37.8%). Stinging nettle was preferred as the most popular herb in groups of CPU/RT (46%). The significantly lower consumption of Echinacea was noticed in MU/H 4.5% in comparison with groups, CU/PE 26.4% (p < 0.05), CPU/PEEH 27.4% (p < 0.01), UP/PE 17.2% (p < 0.05) and CPU/RT 28% (p < 0.05). Regularly, all the year round the highest utilization of Echinacea was evident in CPU/BI 30.0%. The highest percentage formed respondent's utilized Echinacea only during illness. Otherwise, the differences between the frequencies of Echinacea usage cannot be considered as statistically significant. Generally, a significantly higher level of ginger usage was assayed within groups SUA/H 80.0% (p < 0.001), CPU/PEEH 66.3% (p < 0.001), UP/PE 36.6% (p < 0.001), CPU/BI 58.8% (p < 0.001), CPU/RT 56.0% (p < 0.001), MU/H 78.6% (p < 0.001) and CPU/PEES 77.1% (p < 0.001) in comparison with the rest of the groups. Daily the respondents from CU/PE 20.8% consumed ginger significantly more often than students belonging to CPU/BI 0.0% (p < 0.05) and MU/H 0.0% (p < 0.05). Respondents from CPU/PEEH consumed statistically significantly more ginger once a week in comparison with students belonged to MU/H 0.9% (p < 0.05). To sum up the research results, we can claim that state or study programme had no clear statistically significant evidence on the regular consumption of medicinal plants as herbal antibiotics. © 2019 Potravinarstvo Slovak Journal of Food Sciences

    Possible Implications of Bacteriospermia on the Sperm Quality, Oxidative Characteristics, and Seminal Cytokine Network in Normozoospermic Men

    This study focused on the identification of bacterial profiles of semen in normozoospermic men and their possible involvement in changes to the sperm structural integrity and functional activity. Furthermore, we studied possible fluctuations of selected cytokines, oxidative markers, and antibacterial proteins as a result of bacterial presence in the ejaculate. Sperm motility was assessed with computer-assisted sperm analysis, while sperm apoptosis, necrosis and acrosome integrity were examined with fluorescent methods. Reactive oxygen species (ROS) generation was quantified via luminometry, sperm DNA fragmentation was evaluated using the TUNEL protocol and chromatin-dispersion test, while the JC-1 assay was applied to evaluate the mitochondrial membrane potential. Cytokine levels were quantified with the biochip assay, whilst selected antibacterial proteins were quantified using the ELISA method. The predominant species identified by the matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry were Staphylococcus hominis, Staphylococcus capitis and Micrococcus luteus. The results revealed that the sperm quality decreased proportionally to the increasing bacterial load and occurrence of conditionally pathogenic bacteria, including Enterococcus faecalis, Staphylococcus aureus and Escherichia coli. Antimicrobial susceptibility tests revealed a substantial resistance of randomly selected bacterial strains to ampicillin, vancomycin, tobramycin, and tetracycline. Furthermore, an increased bacterial quantity in semen was accompanied by elevated levels of pro-inflammatory cytokines, including interleukin-1, interleukin-2, interleukin-6, tumor necrosis factor alpha as well as ROS overproduction and lipid peroxidation of the sperm membranes. Our results suggest that semen quality may be notably affected by the bacterial quantity as well as quality. It seems that bacteriospermia may be associated with inflammatory processes, oxidative stress, sperm structural deterioration, and a subsequent risk for the development of subfertility, even in normozoospermic males

    Ejaculatory Abstinence Affects the Sperm Quality in Normozoospermic Men—How Does the Seminal Bacteriome Respond?

    This study was designed to describe bacterial profiles of ejaculates collected following a long and short ejaculatory abstinence set in the context of changes in the conventional, oxidative, and immunological characteristics of semen. Two specimens were collected in succession from normozoospermic men (n = 51) following 2 days and 2 h, respectively. Semen samples were processed and analyzed according to the World Health Organization (WHO) 2021 guidelines. Afterwards, sperm DNA fragmentation, mitochondrial function, levels of reactive oxygen species (ROS), total antioxidant capacity, and oxidative damage to sperm lipids and proteins were evaluated in each specimen. Selected cytokine levels were quantified using the ELISA method. Bacterial identification by matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry revealed that samples collected following two days of abstinence presented with a higher bacterial load and diversity, and a greater prevalence of potentially uropathogenic bacteria including Escherichia coli, Staphylococcus aureus and Enterococcus faecalis. Only staphylococci and Escherichia coli remained present in specimens obtained after 2 h of abstinence. Whilst all samples accomplished the criteria set by WHO, a significantly higher motility (p p p p p p p p p p < 0.05) were observed in specimens collected after two days of abstinence. It may be summarized that shorter ejaculatory abstinence does not compromise sperm quality in normozoospermic men, while it contributes to a decreased occurrence of bacteria in semen which is accompanied by a lower probability of damage to spermatozoa by ROS or pro-inflammatory cytokines

    The effects of 12-week progressive strength training on strength, functional capacity, metabolic biomarkers, and serum hormone concentrations in healthy older women : morning versus evening training

    Previous findings suggest that performing strength training (ST) in the evening may provide greater benefit for young individuals. However, this may not be optimal for the older population. The purpose of this study was to compare the effects of a 12-week ST program performed in the morning vs. evening on strength, functional capacity, metabolic biomarker and basal hormone concentrations in older women. Thirty-one healthy older women (66 ± 4 years, 162 ± 4 cm, 75 ± 13 kg) completed the study. Participants trained in the morning (M) (07:30, n = 10), in the evening (E) (18:00, n = 10), or acted as a non-training control group (C) (n = 11). Both intervention groups performed whole-body strength training with 3 sets of 10–12 repetitions with 2–3 minutes rest between sets. All groups were measured before and after the 12-week period with; dynamic leg press and seated-row 6-repetition maximum (6-RM) and functional capacity tests (30-second chair stands and arm curl test, Timed Up and Go), as well as whole-body skeletal muscle mass (SMM) (kg) and fat mass (FM-kg, FM%) assessed by bioelectrical impedance (BIA). Basal blood samples (in the intervention groups only) taken before and after the intervention assessed low-density lipoprotein (LDL-C), high-density lipoprotein (HDL-C), blood glucose (GLU), triglycerides (TG), high-sensitive C-reactive protein (hsCRP) concentrations and total antioxidant status (TAS) after a 12 h fast. Hormone analysis included prolactin (PRL), progesterone (P) estradiol (ESTR), testosterone (T), follicle stimulating hormone (FSH), and luteinizing hormone (LH). While C showed no changes in any variable, both M and E significantly improved leg press (+ 46 ± 22% and + 21 ± 12%, respectively; p < 0.001) and seated-row (+ 48 ± 21% and + 42 ± 18%, respectively; p < 0.001) 6-RM, as well as all functional capacity outcomes (p < 0.01) due to training. M were the only group to increase muscle mass (+ 3 ± 2%, p < 0.01). Both M and E group significantly (p < 0.05) decreased GLU (–4 ± 6% and –8 ± 10%, respectively), whereas significantly greater decrease was observed in the E compared to the M group (p < 0.05). Only E group significantly decreased TG (–17 ± 25%, p < 0.01), whereas M group increased (+ 15%, p < 0.01). The difference in TG between the groups favored E compared to M group (p < 0.01). These results suggest that short-term “hypertrophic” ST alone mainly improves strength and functional capacity performance, but it influences metabolic and hormonal profile of healthy older women to a lesser extent. In this group of previously untrained older women, time-of-day did not have a major effect on outcome variables, but some evidence suggests that training in the morning may be more beneficial for muscle hypertrophy (i.e. only M significantly increased muscle mass and had larger effect size (M: g = 2 vs. E: g = 0.5).peerReviewe

    <i>Thymus zygis,</i> Valuable Antimicrobial (In Vitro and In Situ) and Antibiofilm Agent with Potential Antiproliferative Effects

    With the growing issues of food spoilage, microbial resistance, and high mortality caused by cancer, the aim of this study was to evaluate T. zygis essential oil (TZEO) as a potential solution for these challenges. Here, we first performed GC/MS analysis which showed that the tested TZEO belongs to the linalool chemotype since the abundance of linalool was found to be 38.0%. Antioxidant activity assays showed the superiority of TZEO in neutralizing the ABTS radical cation compared to the DPPH radical. The TZEO was able to neutralize 50% of ABTS•+ at the concentration of 53.03 ± 1.34 μg/mL. Antimicrobial assessment performed by employing disc diffusion and minimal inhibitory concentration assays revealed TZEO as a potent antimicrobial agent with the highest inhibition activity towards tested gram-negative strains. The most sensitive on the treatment with TZEO was Enterobacter aerogenes showing an MIC 50 value of 0.147 ± 0.006 mg/mL and a MIC 90 value of 0.158 ± 0.024 mg/mL. Additionally, an in situ analysis showed great effects of TZEO in inhibiting gram-negative E. coli, P. putida, and E. aerogenes growing on bananas and cucumbers. Treatment with the TZEO vapor phase in the concentration of 500 μg/mL was able to reduce the growth of these bacteria on the food models to the extent > 90%, except for E. coli growth on the cucumber, which was reduced to the extent of 83.87 ± 4.76%. Furthermore, a test on the antibiofilm activity of the tested essential oil revealed its biofilm prevention effects against Salmonella enterica which forms biofilms on plastic and stainless-steel surfaces. Performed tests on the TZEO effects towards cell viability showed no effects on the normal MRC-5 cell line. However, the results of MTT assay of TZEO effects on three cancer cell lines (MDA-MB-231, HCT-116, and K562) suggest that TZEO exerted the strongest effects on the inhibition of the viability of MDA-MB-231 cells, especially after long-term treatment in the highest concentration applied with reducing the viability of the cells to 57%. Additionally, results of NBT and Griess assays suggest that TZEO could be a convenient candidate for future testing for developing novel antitumor therapies
