Phänotypische Merkmale für die Selektion heimischer Leguminosen auf Methioninreichtum in der Pflanzenzüchtung
Ein Problem der Fütterung von Schweinen und Geflügel mit 100 % Futtermitteln aus Ökologischen Landbau sind die unzureichenden Methionin-(Met)-Gehalte im Protein europäischer Körnerleguminosen. Ziel der hier präsentierten Arbeiten ist es, neue Leguminosen Kultivare mit hohem Met-Gehalt im Samenprotein zu identifizieren. Neben der direkten Analyse der Aminosäuregehalte eines Zuchtsortiments der Pflanzenarten wird dazu die Übertragbarkeit der Methoden eines bei Soja (Glycine max) erfolgreich angewandten Verfahrens zur phänotypischen Selektion Met-reicher Pflanzen nach Imsande (2001) an Erbsen (Pisum sativum (L.)), Ackerbohnen (Vicia faba (L.)) and Lupinen (Lupinus angustifolius (L.)) erprobt. Als Indikatoren für Pflanzen mit hohem Metgehalt dienten der Chlorophyllgehalt der Blätter und das Wurzelwachstum von Keimlingen in einer Ethionin Lösung (0,75 mM). Erste Ergebnisse im Labormaßstab an unter Met-Zugabe aufgezogenen Pflanzen zeigten bei L. angustifolius and V. faba eine positive Korrelation zwischen der Met-Versorgung und den Chlorophyllgehalten. Met-reiche Pflanzen zeigten im Vergleich zu den unbehandelten Pflanzen um bis zu 59 % (L. angustifolius) bzw. bis zu 34 % (V. faba) erhöhte Chlorophyllgehalte in den Blättern. In einem zweiten Ansatz wurde der phytotoxische Effekt von Ethionin, einem chemischen Derivat zu Met, zum Screening auf Met-reiche Pflanzen genutzt. Der phytotoxische Effekt wird durch ansteigende Met-Konzentrationen gemindert. In einer 1 mM Met Lösung inkubierte Samen bildeten in Ethionin Lösung bis zu 33 % (P. sativum) bzw. bis zu 18 % (V. faba) längere Wurzeln aus. Die Erfolg versprechenden Ansätze zur Selektion Met-reicher Pflanzen mit direkt mit Met behandelten Pflanzen werden derzeit an Feldpopulationen erprobt
Variational Convergence and Discrete Minimal Surfaces
This work is concerned with the convergence behavior of the solutions to parametric variational problems. An emphasis is put on sequences of variational problems that arise as discretizations of either infinite-dimensional optimization problems or infinite-dimensional operator problems. Finally, the results are applied to discretizations of the Douglas-Plateau problem and of a boundary value problem in nonlinear elasticity
Impact of methionine nutrition on the leaf proteome of Lupinus angustifolius L. and Vicia faba L.
Grain legumes possess a gernerally favourable seed protein amino acid composition but low contents of sulfur amino acids decrease their nutrient value. To enhance the methionie content of local grain legumes by plant breeding, phenotypical attributes for high methionine contents are required for the selection process
CoBarS: Fast reweighted sampling for polygon spaces in any dimension
We present the first algorithm for sampling random configurations of closed
-gons with any fixed edgelengths in any dimension
which is proved to sample correctly from standard probability measures on these
spaces. We generate open -gons as weighted sets of edge vectors on the unit
sphere and close them by taking a M\"obius transformation of the sphere which
moves the center of mass of the edges to the origin. Using previous results of
the authors, such a M\"obius transformation can be found in time. The
resulting closed polygons are distributed according to a pushforward measure.
The main contribution of the present paper is the explicit calculation of
reweighting factors which transform this pushforward measure to any one of a
family of standard measures on closed polygon space, including the symplectic
volume for polygons in . For fixed dimension, these reweighting
factors may be computed in time. Experimental results show that our
algorithm is efficient and accurate in practice, and an open-source reference

Computing the conformal barycenter
The conformal barycenter of a point cloud on the sphere at infinity of the
Poincar\'e ball model of hyperbolic space is a hyperbolic analogue of the
geometric median of a point cloud in Euclidean space. It was defined by Douady
and Earle as part of a construction of a conformally natural way to extend
homeomorphisms of the circle to homeomorphisms of the disk, and it plays a
central role in Millson and Kapovich's model of the configuration space of
cyclic linkages with fixed edgelengths.
In this paper we consider the problem of computing the conformal barycenter.
Abikoff and Ye have given an iterative algorithm for measures on
which is guaranteed to converge. We analyze Riemannian versions of Newton's
method computed in the intrinsic geometry of the Poincare ball model. We give
Newton-Kantorovich (NK) conditions under which we show that Newton's method
with fixed step size is guaranteed to converge quadratically to the conformal
barycenter for measures on any (including infinite-dimensional
spheres). For measures given by atoms on a finite dimensional sphere which
obey the NK conditions, we give an explicit linear bound on the computation
time required to approximate the conformal barycenter to fixed error. We prove
that our NK conditions hold for all but exponentially few atom measures.
For all measures with a unique conformal barycenter we show that a regularized
Newton's method with line search will always converge (eventually
superlinearly) to the conformal barycenter. Though we do not have hard time
bounds for this algorithm, experiments show that it is extremely efficient in
practice and in particular much faster than the Abikoff-Ye iteration.Comment: 29 pages, 8 figure
Selektion auf Methioninreichtum bei heimischen Leguminosen (Vicia faba, Pisum sativum und Lupinus angustifolius)
Im Projekt wurden phänotyische Selektionskriterien zur Identifizierung von Leguminosenpflanzen (Vicia faba, Pisum sativum und Lupinus angustifolius) mit methioninreichem Samen bzw. Samenprotein eingesetzt (nach Imsande 2001). Die Selektion erfolgte an Mx-Generationen EMS behandelter Pflanzen anhand des Chlorophyllgehaltes der Blätter und der Keimwurzellänge in einer Ethioninlösung (Et-Test). Der Methioningehalt der Samen sollte anhand der Schwefelgehalte eingestuft werden. Der im Projekt alternativ entwickelte bakterielle Schnelltest zur Messung der Methioningehalte im Samenprotein erreichte noch keine ausreichende Genauigkeit.
Zwischen den Chlorophyllgehalten der Blätter und den Methionin- oder Schwefelgehalten in den Samen der Pflanzen konnte kein belastbarer Zusammenhang ermittelt werden.
Im Et-Test konnten bei L. angustifolius im Mittel signifikant höhere relative Methioningehalte (g/16g N) in den Samen der selektierten Pflanzen gegenüber den Kontrollpflanzen festgestellt werden. Bei den übrigen untersuchten Pflanzenarten gelang dies nicht. Bei P. sativum lagen bei den im Et-Test als potentiell methioninreich selektierten Pflanzen bei hohen absoluten Methioningehalten (g/kg) gleichzeitig auch oft hohe S-Gehalte in den Samen vor. Die Methodik des Et-Tests wurde im Projekt erfolgreich entwickelt, jedoch ist die vorliegende Probenzahl für eine abschließende Bewertung der Methodik zu gering.
Bei den Untersuchungen zur regulatorischen Funktion vom freiem, zellulären Methionin auf die Protein- und Genexpression der Pflanzen konnte eine Reduktion der allgemeinen Proteinexpression in Gegenwart von 1mM Methionin festgestellt werden. Ein Zusammenhang zwischen Methioningehalt und Chlorophyllbiosynthese konnte nicht bestätigt werden.
Anhand der Aminosäurezusammensetzung in den Samen eines Leguminosensortiments (je Art 27-50 Sorten) konnte die aus der Literatur bekannte negative Korrelation zwischen Rohproteingehalt und relativem Methioningehalt bestätigt werden. Die Methioningehalte zeigten mit 12-22% (g/16gN) bzw. 16-24% (g/kg) positiver Abweichung vom Mittelwert eine hohe Variabilität des Merkmals.
Die Züchtung von methioninreichen Körnerleguminosen erscheint bereits auf Basis der natürlichen Variabilität der Gehalte mit dem bestehenden Sortiment durchaus erfolgversprechend. Empfehlenswert wäre dazu eine breitangelegte Selektion von Einzelpflanzen aus bestehenden Sorten und Linien nach exakt bestimmten Aminosäuregehalten (z. B. NIR) im Samen
