Spatio-temporal patterns in fin whale <i>Balaenoptera physalus</i> habitat use in the northern Gulf of St. Lawrence
Significant ecosystem changes in the Gulf of St. Lawrence (GSL), Canada, have had far-reaching effects at all trophic levels. The abundance of fin whales Balaenoptera physalus has declined significantly in the northern GSL over the past decade. This study aimed to test the hypothesis that the observed decline was correlated to changing environmental conditions. Cetacean sightings data from 292 surveys, resulting in 2986 fin whale encounters from 2007 to 2013, were used to fit 2 separate generalised additive models in terms of (1) bathymetric and oceanographic variables (the proxy model) and (2) modelled krill biomass (the prey model). The concept of ‘handling time’ was introduced to correct for time off search effort, applicable to other studies relying on opportunistically sampled data. While a positive correlation between krill biomass and fin whale numbers was found, the performance of the proxy model (24.2% deviance explained) was overall better than the prey model (11.8%). Annual predictive maps derived from the final proxy model highlighted 2 key areas with recurrently high relative fin whale abundance and a significant overlap with shipping lanes. While both models provided evidence for an annual decline in relative fin whale abundance, static bathymetric features were the most important predictors of habitat use, and no correlation between dynamic variables and the decline was found. High resolution prey data and a better understanding of the feeding ecology of fin whales are proposed to further investigate the predator-prey relationship and decline of fin whales in the GSL
Temperature elevations can induce switches to homoclinic action potentials that alter neural encoding and synchronization
Almost seventy years after the discovery of the mechanisms of action potential generation, some aspects of their computational consequences are still not fully understood. Based on mathematical modeling, we here explore a type of action potential dynamics - arising from a saddle-node homoclinic orbit bifurcation - that so far has received little attention. We show that this type of dynamics is to be expected by specific changes in common physiological parameters, like an elevation of temperature. Moreover, we demonstrate that it favours synchronization patterns in networks - a feature that becomes particularly prominent when system parameters change such that homoclinic spiking is induced. Supported by in-vitro hallmarks for homoclinic spikes in the rodent brain, we hypothesize that the prevalence of homoclinic spikes in the brain may be underestimated and provide a missing link between the impact of biophysical parameters on abrupt transitions between asynchronous and synchronous states of electrical activity in the brain

Granulocyte colony-stimulating factor blockade enables dexamethasone to inhibit lipopolysaccharide-induced murine lung neutrophils
Glucocorticoids promote neutrophilic inflammation, the mechanisms of which are poorly characterized. Using a lipopolysaccharide (LPS)-induced acute murine lung injury model, we determined the role of granulocyte colony-stimulating factor (G-CSF) in mouse lung neutrophil numbers in the absence and presence of dexamethasone, a potent glucocorticoid. G-CSF was blocked using a neutralizing antibody. Airway neutrophil numbers, cytokine levels, and lung injury parameters were measured. Glucocorticoid treatment maintained LPS-induced airway G-CSF while suppressing TNF and IL-6. The addition of anti-G-CSF antibodies enabled dexamethasone to decrease airway G-CSF, neutrophils, and lung injury scores. In LPS-challenged murine lungs, structural cells and infiltrating leukocytes produced G-CSF. In vitro using BEAS 2B bronchial epithelial cells, A549 lung epithelial cells, human monocyte-derived macrophages, and human neutrophils, we found that dexamethasone and proinflammatory cytokines synergistically induced G-CSF. Blocking G-CSF production in BEAS 2B cells using shRNAs diminished the ability of BEAS 2B cells to protect neutrophils from undergoing spontaneous apoptosis. These data support that G-CSF plays a role in upregulation of airway neutrophil numbers by dexamethasone in the LPS-induced acute lung injury model

Genome-wide association study identifies peanut allergy-specific loci and evidence of epigenetic mediation in US children
Food allergy (FA) affects 2%-10% of US children and is a growing clinical and public health problem. Here we conduct the first genome-wide association study of well-defined FA, including specific subtypes (peanut, milk and egg) in 2,759 US participants (1,315 children and 1,444 parents) from the Chicago Food Allergy Study, and identify peanut allergy (PA)-specific loci in the HLA-DR and -DQ gene region at 6p21.32, tagged by rs7192 (P=5.5 × 10 -8) and rs9275596 (P=6.8 × 10 -10), in 2,197 participants of European ancestry. We replicate these associations in an independent sample of European ancestry. These associations are further supported by meta-analyses across the discovery and replication samples.
