    Chimeric antigen receptor T-cell therapy : design improvements and therapeutic strategies in cancer treatment

    Objectives: To summarize important findings from research on chimeric antigen receptor (CAR) T-cell immunotherapy in cancer. We discuss CAR design, cell products, toxicity management, heterogenous solid tumors and allogeneic transfer. Methods: A review of literature was conducted. The available literature was selected on original research, state-of-the art design, relevance to the objective and journal impact factor. Results: First-generation CARs provide patient T cells with tumor-specific antigen recognition. Second- and third-generation CARs incorporate costimulatory domains for enhanced T-cell persistence and antitumor activity. Fourth-generation CAR T cells (TRUCKs) include a cytokine production cassette, and hold promise in the treatment of heterogenous solid tumors. Transduced cell phenotype and subset composition are important factors. Suicide genes and safety switches are designed to decrease potential toxicity. Multi-specific CAR T cells can address heterogenous tumors. Allogeneic, off-the-shelf CAR T cells might reduce the production delay. Conclusion: CAR T cells have revolutionized the immunotherapeutic treatment of cancer: exciting results in refractory and relapsed B-cell malignancies have been published. Neurologic complications, solid tumor management and allogeneic constructs require further research. In conclusion, further design adjustments will enable CAR T cells to decisively reshape the field of cancer immunotherapy

    From Philanthropists to Juvenile Judges: Women dealing with Juvenile Delinquency. International Debates and Practices (1890-1960)

    Child protection is one of the first domains in which women got involved socially, politically, and professionally. From the end of the nineteenth century onwards, there was growing debate on the international level about the role they could play in the juvenile justice system and the specialized courts dealing with delinquent children that emerged all around Europe. From probation officers to juvenile judges, from women doctors to female police officers, women got engaged in child protection in multiple ways. Yet each of these functions or duties was the subject of fierce discussion and negotiation concerning the specificity – and even legitimacy – of women’s formal participation in this field of social (re)action. How did women come to the forefront as privileged auxiliaries of the justice or police system in juvenile delinquency matters? How did their involvement and commitment, qualified as “natural” in some discourses, actually translate into practice? Did their integration really take place smoothly? And, finally, how was their work received, evaluated or criticised? This paper will analyse the debates and ideas brought forward by major international agencies (League of Nations, International council of Women) on the one hand, and the ways in which some of these ideas were put into practice at the local level on the other. In doing so, the Belgian case, for which we have the most varied and rich source at our disposal, will be at the centre of our attention

    The justification of the combination of vaseline and oxygen has been subject for discussion in many hospitals. Due to the lack of evidence based data in literature, we have provided recommendations from a pharmacist's perspective. The use of petroleum-based products should be avoided when handling patients under oxygen therapy. Whenever a skin moisturizer is needed for lubrication or rehydration of dry nasal passages, the lips or nose when breathing oxygen, consider the use of oil-in water creams or water-based products

    Chronic respiratory infection with Burkholderia cenocepacia (Bc) in patients with cystic fibrosis (CF) is associated with accelerated decline in lung function and increased mortality. It is therefore important to attempt to eradicate new isolates, especially in children. However, there are no standardized guidelines to eradicate Bc. We report a case of successful eradication of new isolates of Bc in a 2-year-old child with CF using a combination of IV, nebulized antibiotics and sinus surgery.SCOPUS: ar.jinfo:eu-repo/semantics/publishe
