    Fumaric acid production by fermentation

    The potential of fumaric acid as a raw material in the polymer industry and the increment of cost of petroleum-based fumaric acid raises interest in fermentation processes for production of this compound from renewable resources. Although the chemical process yields 112% w/w fumaric acid from maleic anhydride and the fermentation process yields only 85% w/w from glucose, the latter raw material is three times cheaper. Besides, the fermentation fixes CO2. Production of fumaric acid by Rhizopus species and the involved metabolic pathways are reviewed. Submerged fermentation systems coupled with product recovery techniques seem to have achieved economically attractive yields and productivities. Future prospects for improvement of fumaric acid production include metabolic engineering approaches to achieve low pH fermentations

    A procedure for the estimation over time of metabolic fluxes in scenarios where measurements are uncertain and/or insufficient

    <p>Abstract</p> <p>Background</p> <p>An indirect approach is usually used to estimate the metabolic fluxes of an organism: couple the available measurements with known biological constraints (e.g. stoichiometry). Typically this estimation is done under a static point of view. Therefore, the fluxes so obtained are only valid while the environmental conditions and the cell state remain stable. However, estimating the evolution over time of the metabolic fluxes is valuable to investigate the dynamic behaviour of an organism and also to monitor industrial processes. Although Metabolic Flux Analysis can be successively applied with this aim, this approach has two drawbacks: i) sometimes it cannot be used because there is a lack of measurable fluxes, and ii) the uncertainty of experimental measurements cannot be considered. The Flux Balance Analysis could be used instead, but the assumption of optimal behaviour of the organism brings other difficulties.</p> <p>Results</p> <p>We propose a procedure to estimate the evolution of the metabolic fluxes that is structured as follows: 1) measure the concentrations of extracellular species and biomass, 2) convert this data to measured fluxes and 3) estimate the non-measured fluxes using the Flux Spectrum Approach, a variant of Metabolic Flux Analysis that overcomes the difficulties mentioned above without assuming optimal behaviour. We apply the procedure to a real problem taken from the literature: estimate the metabolic fluxes during a cultivation of CHO cells in batch mode. We show that it provides a reliable and rich estimation of the non-measured fluxes, thanks to considering measurements uncertainty and reversibility constraints. We also demonstrate that this procedure can estimate the non-measured fluxes even when there is a lack of measurable species. In addition, it offers a new method to deal with inconsistency.</p> <p>Conclusion</p> <p>This work introduces a procedure to estimate time-varying metabolic fluxes that copes with the insufficiency of measured species and with its intrinsic uncertainty. The procedure can be used as an off-line analysis of previously collected data, providing an insight into the dynamic behaviour of the organism. It can be also profitable to the on-line monitoring of a running process, mitigating the traditional lack of reliable on-line sensors in industrial environments.</p

    Identification of a mitotic recombination hotspot on chromosome III of the asexual fungus Aspergillus niger and its possible correlation elevated basal transcription

    Genetic recombination is an important tool in strain breeding in many organisms. We studied the possibilities of mitotic recombination in strain breeding of the asexual fungus Aspergillus niger. By identifying genes that complemented mapped auxotrophic mutations, the physical map was compared to the genetic map of chromosome III using the genome sequence. In a program to construct a chromosome III-specific marker strain by selecting mitotic crossing-over in diploids, a mitotic recombination hotspot was identified. Analysis of the mitotic recombination hotspot revealed some physical features, elevated basal transcription and a possible correlation with purine stretches

    Formation of gullies on Mars by debris flows triggered by CO_2 sublimation

    Martian gully landforms resemble terrestrial debris flows formed by the action of liquid water and have thus been interpreted as evidence for potential habitable environments on Mars within the past few millennia. However, ongoing gully formation has been detected under surface conditions much too cold for liquid water, but at times in the martian year when a thin layer of seasonal CO_2 frost is present and defrosting above the regolith. These observations suggest that the CO_2 condensation–sublimation cycle could play a role in gully formation. Here we use a thermo-physical numerical model of the martian regolith underlying a CO_2 ice layer and atmosphere to show that the pores beneath the ice layer can be filled with CO_2 ice and subjected to extreme pressure variations during the defrosting season. The subsequent gas fluxes can destabilize the regolith material and induce gas-lubricated debris flows with geomorphic characteristics similar to martian gullies. Moreover, we find that subsurface CO_2 ice condensation, sublimation and pressurization occurs at conditions found at latitudes and slope orientations where gullies are observed. We conclude that martian gullies can result from geologic dry ice processes that have no terrestrial analogues and do not require liquid water. Such dry ice processes may have helped shape the evolution of landforms elsewhere on the martian surface

In der ersten Stufe wird die Lactose der Molke in Milchsäure vergoren und in der zweiten Stufe mit enzymatisch abgebauter Stärke aus Weizenmehl als Substrat in der umgesetzten Molke, Saccharomyces cerevisiae in fed batch Betrieb kultiviert. Die Zusammensetzung der Biomasse wird näher untersucht
