    Intersecting D-brane states derived from the KP theory

    A general scheme to find tachyon boundary states is developed within the framework of the theory of KP hierarchy. The method is applied to calculate correlation function of intersecting D-branes and rederived the results of our previous works as special examples.     Quantitative Mapping of Cocaine-Induced ΔFosB Expression in the Striatum of Male and Female Rats

    ΔFosB plays a critical role in drug-induced long-term changes in the brain. In the current study, we evaluated locomotor activity in male and female rats treated with saline or cocaine for 2 weeks and quantitatively mapped ΔFosB expression in the dorsal striatum and nucleus accumbens of each animal by using an anti-FosB antibody that recognizes ΔFosB isoforms preferentially. Behavioral analysis showed that while there was little difference between males and females that sensitized to cocaine, nonsensitizing rats showed a large sex difference. Nonsensitizing males showed low behavioral activation in response to cocaine on the first day of treatment, and their activity remained low. In contrast, nonsensitizing females showed high activation on the first day of treatment and their activity remained high. Western blot and immunohistochemical analyses indicated that basal levels of ΔFosB were higher in the nucleus accumbens than the dorsal striatum, but that the effect of cocaine on ΔFosB was greater in the dorsal striatum. Immunostaining showed that the effect of cocaine in both the dorsal striatum and nucleus accumbens was primarily to increase the intensity of ΔFosB immunoreactivity in individual neurons, rather than to increase the number of cells that express ΔFosB. Detailed mapping of ΔFosB-labeled nuclei showed that basal ΔFosB levels were highest in the medial portion of the dorsal striatum and dorsomedial accumbens, particularly adjacent to the lateral ventricle, whereas the cocaine-induced increase in ΔFosB was most pronounced in the lateral dorsal striatum, where basal ΔFosB expression was lowest. Sex differences in ΔFosB expression were small and independent of cocaine treatment.     Modeling gene expression regulatory networks with the sparse vector autoregressive model

    <p>Abstract</p> <p>Background</p> <p>To understand the molecular mechanisms underlying important biological processes, a detailed description of the gene products networks involved is required. In order to define and understand such molecular networks, some statistical methods are proposed in the literature to estimate gene regulatory networks from time-series microarray data. However, several problems still need to be overcome. Firstly, information flow need to be inferred, in addition to the correlation between genes. Secondly, we usually try to identify large networks from a large number of genes (parameters) originating from a smaller number of microarray experiments (samples). Due to this situation, which is rather frequent in Bioinformatics, it is difficult to perform statistical tests using methods that model large gene-gene networks. In addition, most of the models are based on dimension reduction using clustering techniques, therefore, the resulting network is not a gene-gene network but a module-module network. Here, we present the Sparse Vector Autoregressive model as a solution to these problems.</p> <p>Results</p> <p>We have applied the Sparse Vector Autoregressive model to estimate gene regulatory networks based on gene expression profiles obtained from time-series microarray experiments. Through extensive simulations, by applying the SVAR method to artificial regulatory networks, we show that SVAR can infer true positive edges even under conditions in which the number of samples is smaller than the number of genes. Moreover, it is possible to control for false positives, a significant advantage when compared to other methods described in the literature, which are based on ranks or score functions. By applying SVAR to actual HeLa cell cycle gene expression data, we were able to identify well known transcription factor targets.</p> <p>Conclusion</p> <p>The proposed SVAR method is able to model gene regulatory networks in frequent situations in which the number of samples is lower than the number of genes, making it possible to naturally infer partial Granger causalities without any <it>a priori </it>information. In addition, we present a statistical test to control the false discovery rate, which was not previously possible using other gene regulatory network models.</p

    Unbalanced relationships: insights into the interaction between gut microbiota, geohelminths, and schistosomiasis

    Hosts and their microbiota and parasites have co-evolved in an adaptative relationship since ancient times. The interaction between parasites and intestinal bacteria in terms of the hosts’ health is currently a subject of great research interest. Therapeutic interventions can include manipulations of the structure of the intestinal microbiota, which have immunological interactions important for modulating the host’s immune system and for reducing inflammation. Most helminths are intestinal parasites; the intestinal environment provides complex interactions with other microorganisms in which internal and external factors can influence the composition of the intestinal microbiota. Moreover, helminths and intestinal microorganisms can modulate the host’s immune system either beneficially or harmfully. The immune response can be reduced due to co-infection, and bacteria from the intestinal microbiota can translocate to other organs. In this way, the treatment can be compromised, which, together with drug resistance by the parasites makes healing even more difficult. Thus, this work aimed to understand interactions between the microbiota and parasitic diseases caused by the most important geohelminths and schistosomiasis and the consequences of these associations

    Lepton Flavor Violating Process in Bi-maximal texture of Neutrino Mixings

    We investigate the lepton flavor violation in the framework of the MSSM with right-handed neutrinos taking the large mixing angle MSW solution in the quasi-degenerate and the inverse-hierarchical neutrino masses. We predict the branching ratio of μe+γ\mu \to e+\gamma and τμ+γ\tau \to \mu+\gamma processes assuming the degenerate right-handed Majorana neutrino masses. We find that the branching ratio in the quasi-degenerate neutrino mass spectrum is 100 times smaller than the ones in the inverse-hierarchical and the hierarchical neutrino spectra. We emphasize that the magnitude of Ue3U_{e3} is one of important ingredients to predict BR(μe+γ\mu \to e +\gamma ). The effect of the deviation from the complete-degenerate right-handed Majorana neutrino masses are also estimated. Furtheremore, we examine the S_{3\sL}\times S_{3\sR} model, which gives the quasi-degenerate neutrino masses, and the Shafi-Tavartkiladze model, which gives the inverse-hierarchical neutrino masses.     Leptogenesis in Neutrino Textures with Two Zeros

    The leptogenesis is studied in the neutrino textures with two zeros, which reduce the number of independent phases of the CP violation. The phenomenological favored neutrino textures with two zeros are decomposed into the Dirac neutrino mass matrix and the right-handed Majorana one in the see-saw mechanism. Putting the condition to suppress the μeγ\mu \to e\gamma decay enough, the texture zeros of the Dirac neutrino mass matrix are fixed in the framework of the MSSM with right-handed neutrinos. These textures have only one CP violatig phase. The magnitude of each entry of the Dirac mass matrix is determined in order to explain the baryon asymmetry of the universe by solving the Boltzman equations.     Prevention of type 2 diabetes in a primary healthcare setting: Three-year results of lifestyle intervention in Japanese subjects with impaired glucose tolerance

    <p>Abstract</p> <p>Background</p> <p>A randomized control trial was performed to test whether a lifestyle intervention program, carried out in a primary healthcare setting using existing resources, can reduce the incidence of type 2 diabetes in Japanese with impaired glucose tolerance (IGT). The results of 3 years' intervention are summarized.</p> <p>Methods</p> <p>Through health checkups in communities and workplaces, 304 middle-aged IGT subjects with a mean body mass index (BMI) of 24.5 kg/m<sup>2 </sup>were recruited and randomized to the intervention group or control group. The lifestyle intervention was carried out for 3 years by public health nurses using the curriculum and educational materials provided by the study group.</p> <p>Results</p> <p>After 1 year, the intervention had significantly improved body weight (-1.5 ± 0.7 vs. -0.7 ± 2.5 kg in the control; p = 0.023) and daily non-exercise leisure time energy expenditure (25 ± 113 vs. -3 ± 98 kcal; p = 0.045). Insulin sensitivity assessed by the Matsuda index was improved by the intervention during the 3 years. The 3-year cumulative incidence tended to be lower in the intervention group (14.8% vs.8.2%, log-rank test: p = 0.097). In a sub-analysis for the subjects with a BMI > 22.5 kg/m<sup>2</sup>, a significant reduction in the cumulative incidence was found (p = 0.027).</p> <p>Conclusions</p> <p>The present lifestyle intervention program using existing healthcare resources is beneficial in preventing diabetes in Japanese with IGT.     Inhibition of dendritic cell migration by transforming growth factor-β1 increases tumor-draining lymph node metastasis

    <p>Abstract</p> <p>Background</p> <p>Transforming growth factor (TGF)-β is known to be produced by progressor tumors and to immobilize dendritic cells (DCs) within those tumors. Moreover, although TGF-β1 has been shown to promote tumor progression, there is still no direct, in vivo evidence as to whether TGF-β1 is able to directly induce distant metastasis.</p> <p>Methods</p> <p>To address that issue and investigate the mechanism by which TGF-β1 suppresses DC activity, we subdermally inoculated mouse ears with squamous cell carcinoma cells stably expressing TGF-β1 or empty vector (mock).</p> <p>Results</p> <p>The numbers of DCs within lymph nodes draining the resultant TGF-β1-expressing tumors was significantly lower than within nodes draining tumors not expressing TGF-β1. We then injected fluorescently labeled bone marrow-derived dendritic cells into the tumors, and subsequent analysis confirmed that the tumors were the source of the DCs within the tumor-draining lymph nodes, and that there were significantly fewer immature DCs within the nodes draining TGF-β1-expressing tumors than within nodes draining tumors not expressing TGF-β1. In addition, 14 days after tumor cell inoculation, lymph node metastasis occurred more frequently in mice inoculated with TGF-β1 transfectants than in those inoculated with the mock transfectants.</p> <p>Conclusions</p> <p>These findings provide new evidence that tumor-derived TGF-β1 inhibits migration of DCs from tumors to their draining lymph nodes, and this immunosuppressive effect of TGF-β1 increases the likelihood of metastasis in the affected nodes.</p
