    The evolution of inverted magnetic fields through the inner heliosphere

    Local inversions are often observed in the heliospheric magnetic field (HMF), but their origins and evolution are not yet fully understood.Parker Solar Probe has recently observed rapid, AlfvĂ©nic, HMF inversions in the inner heliosphere, known as ‘switchbacks’, which have been interpreted as the possible remnants of coronal jets. It has also been suggested that inverted HMF may be produced by near-Sun interchange reconnection; a key process in mechanisms proposed for slow solar wind release. These cases suggest that the source of inverted HMF is near the Sun, and it follows that these inversions would gradually decay and straighten as they propagate out through the heliosphere. Alternatively, HMF inversions could form during solar wind transit, through phenomena such velocity shears, draping over ejecta, or waves and turbulence. Such processes are expected to lead to a qualitatively radial evolution of inverted HMF structures. Using Helios measurements spanning 0.3–1 AU, we examine the occurrence rate of inverted HMF, as well as other magnetic field morphologies, as a function of radial distance r, and find that it continually increases. This trend may be explained by inverted HMF observed between 0.3–1 AU being primarily driven by one or more of the above in-transit processes, rather than created at the Sun. We make suggestions as to the relative importance of these different processes based on the evolution of the magnetic field properties associated with inverted HMF. We also explore alternative explanations outside of our suggested driving processes which may lead to the observed trend

    Utilising co-design to improve outpatient neurological care in a rural setting

    It has been identified that the physiotherapy needs of patients with central neurological conditions are specific and that this cohort are generally under-serviced in rural and remote areas in Australia. A quality improvement project was undertaken to improve patient experience in outpatient physiotherapy services in Tasmania, facilitating increased self-efficacy and quality of life, in patients with central chronic neurological conditions.. An experience-based co-design approach was utilised, involving past and current patients as well as physiotherapy staff in the project design, data collection, analysis and evaluation phases. The results suggest that timely access to care and goal achievement are common areas of need across both patient and staff cohorts. Patients also identified that shared-decision making is important for improving patient experience and staff were generally unclear on what services were available. The findings from this study demonstrate the importance of including patients and staff in the health service improvement process.     Overcoming barriers to single-cell RNA sequencing adoption in low- and middle-income countries

    The advent of single-cell resolution sequencing and spatial transcriptomics has enabled the delivery of cellular and molecular atlases of tissues and organs, providing new insights into tissue health and disease. However, if the full potential of these technologies is to be equitably realised, ancestrally inclusivity is paramount. Such a goal requires greater inclusion of both researchers and donors in low- and middle-income countries (LMICs). In this perspective, we describe the current landscape of ancestral inclusivity in genomic and single-cell transcriptomic studies. We discuss the collaborative efforts needed to scale the barriers to establishing, expanding, and adopting single-cell sequencing research in LMICs and to enable globally impactful outcomes of these technologies

    A novel bismuth-based metal-organic framework for high volumetric methane and carbon dioxide adsorption

    Solvothermal reaction of H4L (L = biphenyl-3,3’,5,5’-tetracarboxylate) and Bi(NO3)3·(H2O)5 in a mixture of DMF/MeCN/H2O in the presence of piperazine and nitric acid at 100 oC for 10 h affords the solvated metal-organic polymer [Bi2(L)1.5(H2O)2]·(DMF)3.5·(H2O)3 (NOTT-220-solv). A single crystal X-ray structure determination confirms that it crystallises in space group P2/c and has a neutral and non-interpenetrated structure comprising binuclear {Bi2} centres bridged by tetracarboxylate ligands. NOTT-220-solv shows a 3,6-connected network having a new framework topology with a {4·62}2{42·65·88}{62·8} point symbol. The desolvated material NOTT-220a shows exceptionally high adsorption uptakes for CH4 and CO2 on a volumetric basis at moderate pressures and temperatures with a CO2 uptake of 553 gL-1 (20 bar, 293 K) with a saturation uptake of 688 gL-1 (1 bar, 195 K). The corresponding CH4 uptake of 165 V(STP)/V (20 bar, 293 K) and 189 V(STP/V) (35 bar, 293 K) is within the top three MOF materials under the same conditions, surpassed only by PCN-14 and Ni-MOF-74 (230 and 190 V(STP)/V 35 Bar, 298 K). The maximum CH4 uptake for NOTT-220a was recorded at 20 bar and 195 K to be 287 V(STP)/V, while H2 uptake of NOTT-220a at 20 bar, 77 K is 42 gL-1. These gas uptakes have been modelled by Grand Canonical Monte Carlo (GCMC) and Density Functional Theory (DFT) calculations, which confirm the experimental data and give insights into the nature of the binding sites of CH4 and CO2 in this porous hybrid material

    The Relationship between Intelligence and Anxiety: An Association with Subcortical White Matter Metabolism

    We have demonstrated in a previous study that a high degree of worry in patients with generalized anxiety disorder (GAD) correlates positively with intelligence and that a low degree of worry in healthy subjects correlates positively with intelligence. We have also shown that both worry and intelligence exhibit an inverse correlation with certain metabolites in the subcortical white matter. Here we re-examine the relationships among generalized anxiety, worry, intelligence, and subcortical white matter metabolism in an extended sample. Results from the original study were combined with results from a second study to create a sample comprised of 26 patients with GAD and 18 healthy volunteers. Subjects were evaluated using the Penn State Worry Questionnaire, the Wechsler Brief intelligence quotient (IQ) assessment, and proton magnetic resonance spectroscopic imaging (1H-MRSI) to measure subcortical white matter metabolism of choline and related compounds (CHO). Patients with GAD exhibited higher IQ’s and lower metabolite concentrations of CHO in the subcortical white matter in comparison to healthy volunteers. When data from GAD patients and healthy controls were combined, relatively low CHO predicted both relatively higher IQ and worry scores. Relatively high anxiety in patients with GAD predicted high IQ whereas relatively low anxiety in controls also predicted high IQ. That is, the relationship between anxiety and intelligence was positive in GAD patients but inverse in healthy volunteers. The collective data suggest that both worry and intelligence are characterized by depletion of metabolic substrate in the subcortical white matter and that intelligence may have co-evolved with worry in humans

    Predicting the Effects of Reservoir Water Level Management on the Reproductive Output of a Riparian Songbird

    Dams and reservoirs alter natural water flow regimes with adverse effects on natural ecosystems. Quantifying and reducing these effects are important as global demands for energy and water, and the number of dams and reservoir, increase. However, costs and logistic constraints typically preclude experimental assessment of reservoir effects on the environment. We developed a stochastic individual-based model (IBM), parameterized using empirical data, to estimate the annual productivity of yellow warblers that breed in riparian habitat within the footprint of the Arrow Lakes Reservoir in British Columbia, Canada. The IBM incorporated information on breeding phenology, nest site selection, brood parasitism, daily nest survival, re-nesting probabilities and post-fledging survival. We used the IBM to estimate the effect of four different water management scenarios on annual productivity. We found that the IBM accurately estimated average nest success (0.39 ± 0.10 SD), the proportion of females that produced at least one fledgling during a breeding season (0.56 ± 0.11), and annual fledging success (2.06 ± 0.43) under current conditions. The IBM estimated that reservoir operations currently reduce the annual productivity of this population by 37%, from an average of 1.62 to 1.06 independent young/female. Delaying when reservoir water levels reach 435m asl (the minimum elevation occupied by yellow warblers) by approximately 2 weeks was predicted to increase annual productivity to 1.44 independent young/female. The standardized effect on annual productivity of reducing the maximum elevation of the reservoir so that yellow warbler habitat is not inundated (Cohen’s d = 1.52) or delaying when water is stored (Cohen’s d = 0.83) was primarily driven by inundation effects on post-fledging survival. Reservoir operation effects on breeding birds will be species specific, but this IBM can easily be modified to allow the environmental impacts on the entire breeding bird community to be incorporated into water management decisions

    Testing the price and affordability of healthy and current (unhealthy) diets and the potential impacts of policy change in Australia

    Abstract Background: Price and affordability of foods are important determinants of health. Targeted food pricing policies may help improve population diets. However, methods producing comparable data to inform relevant policy decisions are lacking in Australia and globally. The objective was to develop and pilot standardised methods to assess the price, relative price and affordability of healthy (recommended) and current (unhealthy) diets and test impacts of a potential policy change. Methods: Methods followed the optimal approach proposed by INFORMAS using recent Australian dietary intake data and guidelines. Draft healthy and current (unhealthy) diet baskets were developed for five household structures. Food prices were collected in stores in a high and low SES location in Brisbane, Australia. Diet prices were calculated and compared with household incomes, and with potential changes to the Australian Taxation System. Wilcoxen-signed rank tests were used to compare differences in price. Results: The draft tools and protocols were deemed acceptable at household level, but methods could be refined. All households spend more on current (unhealthy) diets than required to purchase healthy (recommended) diets, with the majority (53-64 %) of the food budget being spent on 'discretionary' choices, including take-away foods and alcohol. A healthy diet presently costs between 20-31 % of disposable income of low income households, but would become unaffordable for these families under proposed changes to expand the GST to apply to all foods in Australia. Conclusions: Results confirmed that diet pricing methods providing meaningful, comparable data to inform potential fiscal and health policy actions can be developed, but draft tools should be refined. Results suggest that healthy diets can be more affordable than current (unhealthy) diets in Australia, but other factors may be as important as price in determining food choices

    Single nucleus and spatial transcriptomic profiling of human healthy hamstring tendon

    The molecular and cellular basis of health in human tendons remains poorly understood. Amongst human tendons, the hamstrings are the least likely to be injured or degenerate, providing a prototypic healthy tendon reference. The aim of this study was to define the transcriptome and location of all cell types in healthy hamstring tendon. We profiled the transcriptomes of 10,533 nuclei from 4 healthy donors using single-nucleus RNA sequencing (snRNA-seq) and identified 12 distinct cell types. We confirmed the presence of two fibroblast cell types, endothelial cells, mural cells, and immune cells, and revealed the presence of cell types previously unreported for tendon sites, including different skeletal muscle cell types, satellite cells, adipocytes, and nerve cells, which are undefined nervous system cells. Location of these cell types within tendon was defined using spatial transcriptomics and imaging, and transcriptional networks and cell-cell interactions were identified. We demonstrate that fibroblasts have a high number of potential cell-cell interactions, are present throughout the whole tendon tissue, and play an important role in the production and organisation of extracellular matrix, thus confirming their role as key regulators of hamstring tendon tissue homeostasis. Overall, our findings highlight the highly complex cellular networks underpinning tendon function and underpins the importance of fibroblasts as key regulators of hamstring tendon tissue homeostasis
