Epigenetic Regulation of Intronic Transgenes in Arabidopsis
Defense mechanisms of plant genomes can epigenetically inactivate repetitive sequences and exogenous transgenes. Loss of mutant phenotypes in intronic T-DNA insertion lines by interaction with another T-DNA locus, termed T-DNA suppression, has been observed in Arabidopsis thaliana, although the molecular basis of establishment and maintenance of T-DNA suppression is poorly understood. Here we show that maintenance of T-DNA suppression requires heterochromatinisation of T-DNA sequences and the nuclear proteins, INCREASED IN BONSAI METHYLATION 2 (IBM2) and ENHANCED DOWNY MILDEW 2 (EDM2), which prevent ectopic 3′ end processing of mRNA in atypically long introns containing T-DNA sequences. Initiation of T-DNA suppression is mediated by the canonical RdDM pathway after hybridisation of two T-DNA strains, accompanied by DNA hypermethylation of T-DNA sequences in the F1 generation. Our results reveal the presence of a genome surveillance mechanism through genome hybridisation that masks repetitive DNAs intruding into transcription units
El marketing relacional y la generación de valor en el cliente de la empresa Viettel Perú S.A.C., Lima Metropolitana 2020
Esta investigación tuvo como objetivo general analizar de qué manera
el marketing relacional contribuye en la generación de valor de los
clientes de la empresa Viettel Perú S.A.C., Lima Metropolitana 2020.
Este estudio fue de enfoque cuantitativo, tipo básica, nivel correlacional
y de diseño no experimental de corte transversal. La población la
conformaron 389045 personas que son la totalidad de la población
comprendida por los distritos Jesús María, Pueblo Libre, Magdalena del
Mar, San Miguel y Lince, según datos del INEI; del cual se utilizó un
muestreo estratificado con afijación proporcional, el cual con la muestra
de 228 se obtuvo un número de encuestados determinado para cada
distrito. La técnica que se empleó fue la encuesta, y el instrumento fue
el cuestionario, del cual se obtuvo un estadístico global de fiabilidad de
alfa de Cronbach de 0,970, lo que establece una fiabilidad muy alta, a
su vez se obtuvo una validación de contenido por juicio de expertos de
98%, siendo una excelente calificación. Finalmente, se concluyó que,
existe una relación entre el marketing relacional y la generación de valor
del cliente, ya que se obtuvo coeficiente de Rho de Spearman de 0,378,
correspondiente a una correlación positiva media. El marketing
relacional ayuda a incrementar la generación de valor en la empresa

Absence of Tc-Pinning Phenomenon Under High Pressure in High-Entropy REO0.5F0.5BiS2 Layered Superconductor
Recently, robustness of superconductivity (transition temperature, Tc) under
high pressures has been observed in high-entropy alloy (HEA), bcc-type
Ti-Zr-Hf-Nb-Ta, and HEA-type compounds (Ag,In,Sn,Pb,Bi)Te with a NaCl-type
structure. Since those materials have three-dimensional crystal structure,
investigation on the pressure dependence of Tc of low-dimensional materials is
needed to understand the phenomena. Here, we investigated the superconducting
properties and the crystal structure of BiS2-based layered system
REO0.5F0.5BiS2. Although the robustness of Tc was induced in MTe with
increasing M-site configurational entropy, the increase in RE-site
configurational entropy does not induce robustness of Tc under high pressures
in REO0.5F0.5BiS2. The crystal structure of HEA-type REO0.5F0.5BiS2 was
confirmed as monoclinic P21/m, which is the same space group as the
zero-entropy counterpart LaO0.5F0.5BiS2. The results suggest that an increase
in configurational entropy at blocking layers do not affect crystal structure
and superconducting properties under high pressures in the BiS2-based layered

Epigenetic regulation of spurious transcription initiation in Arabidopsis
In plants, epigenetic regulation is critical for silencing transposons and maintaining proper gene expression. However, its impact on the genome-wide transcription initiation landscape remains elusive. By conducting a genome-wide analysis of transcription start sites (TSSs) using cap analysis of gene expression (CAGE) sequencing, we show that thousands of TSSs are exclusively activated in various epigenetic mutants of Arabidopsis thaliana and referred to as cryptic TSSs. Many have not been identified in previous studies, of which up to 65% are contributed by transposons. They possess similar genetic features to regular TSSs and their activation is strongly associated with the ectopic recruitment of RNAPII machinery. The activation of cryptic TSSs significantly alters transcription of nearby TSSs, including those of genes important for development and stress responses. Our study, therefore, sheds light on the role of epigenetic regulation in maintaining proper gene functions in plants by suppressing transcription from cryptic TSSs
The chromosome-scale genome assembly of the yellowtail clownfish Amphiprion clarkii provides insights into the melanic pigmentation of anemonefish
Anemonefish are an emerging group of model organisms for studying genetic, ecological, evolutionary, and developmental traits of coral reef fish. The yellowtail clownfish Amphiprion clarkii possesses species-specific characteristics such as inter-species co-habitation, high intra-species color variation, no anemone specificity, and a broad geographic distribution, that can increase our understanding of anemonefish evolutionary history, behavioral strategies, fish-anemone symbiosis, and color pattern evolution. Despite its position as an emerging model species, the genome of A. clarkii is yet to be published. Using PacBio long-read sequencing and Hi-C chromatin capture technology, we generated a high-quality chromosome-scale genome assembly initially comprised of 1, 840 contigs with an N50 of 1, 203, 211 bp. These contigs were successfully anchored into 24 chromosomes of 843, 582, 782 bp and annotated with 25, 050 protein-coding genes encompassing 97.0% of conserved actinopterygian genes, making the quality and completeness of this genome the highest among all published anemonefish genomes to date. Transcriptomic analysis identified tissue-specific gene expression patterns, with the brain and optic lobe having the largest number of expressed genes. Further analyses revealed higher copy numbers of erbb3b (a gene involved in melanocyte development) in A. clarkii compared with other anemonefish, thus suggesting a possible link between erbb3b and the natural melanism polymorphism observed in A. clarkii. The publication of this high-quality genome, along with A. clarkii's many unique traits, position this species as an ideal model organism for addressing scientific questions across a range of disciplines
Endoplasmic Reticulum Associated Degradation of Spinocerebellar Ataxia-Related CD10 Cysteine Mutant
Spinocerebellar ataxia (SCA) is one of the most severe neurodegenerative diseases and is often associated with misfolded protein aggregates derived from the genetic mutation of related genes. Recently, mutations in CD10 such as C143Y have been identified as SCA type 43. CD10, also known as neprilysin or neuroendopeptidase, digests functional neuropeptides, such as amyloid beta, in the extracellular region. In this study, we explored the cellular behavior of CD10 C143Y to gain an insight into the functional relationship of the mutation and SCA pathology. We found that wild-type CD10 is expressed on the plasma membrane and exhibits endopeptidase activity in a cultured cell line. CD10 C143Y, however, forms a disulfide bond-mediated oligomer that does not appear by the wild-type CD10. Furthermore, the CD10 C143Y mutant was retained in the endoplasmic reticulum (ER) by the molecular chaperone BiP and was degraded through the ER-associated degradation (ERAD) process, in which representative ERAD factors including EDEM1, SEL1L, and Hrd1 participate in the degradation. Suppression of CD10 C143Y ERAD recovers intracellular transport but not enzymatic activity. Our results indicate that the C143Y mutation in CD10 negatively a ects protein maturation and results in ER retention and following ERAD. These findings provide beneficial insight into SCA type 43 pathology
