An Experimental Investigation of Applying Mica2 Motes in Pavement Condition Monitoring
Pavement maintenance is vital for travel safety, thus detecting dangerous road conditions in a real-time fashion is desirable. Using an off-the-shelf wireless sensor network to detect such conditions at a low cost poses many challenges. In order to meet these challenges, a Mica2 Mote sensor network is adopted in this study to process and transmit data collected from three external analog sensors. Consequentially, several hardware and software interfaces are developed to complete a pavement monitoring system that uses temperature and moisture presence to detect hazardous road conditions. Surge Time Synchronization is explored in this specific application to enable the wireless sensor network to operate in a low power consumption mode. A fairly simplistic pattern classification algorithm is embedded into the motes to create the smart wireless sensing application. A series of outdoor tests are conducted in this study paying special attention to the survivability of fragile analog sensors in harsh roadway conditions. In this regard, a novel solution called the ``Sensor-Road Button''(SRB) is developed and validated experimentally. This is one of several exercises made in this study to enable the application of sensor technologies in intelligent transportation systems (ITS). The size of the wireless sensor network in this study is relatively small, utilizing a total of five motes in order to fully exploit the transmitting range of each mote. Long testing periods (i.e., uninterrupted 12-hour time frames for each period of data collection) add an additional advantage, allowing for the evaluation of the selected wireless sensor network for long-term monitoring using the low power consumption mode under Surge Time Synchronization. Many performance metrics of the adopted small-size, large-interval Mica2 Motes wireless sensor network are revealed in this study through a series of data processing efforts. Results are presented to examine (i) inter-node connectivity and transmitting range, (ii) battery life, (iii) the length of the initial network connection time as affected by methods of setting up tests under practical conditions, (iv) error rate and analysis of different error types (showing the importance of the subsequent data cleansing step), and (v) other network routing properties including the parent time histories for each mote. The results and analysis form a database for future efforts to better understand, appreciate, and improve the performance of Mica2 Motes. Dramatic Repositioning of c-Myb to Different Promoters during the Cell Cycle Observed by Combining Cell Sorting with Chromatin Immunoprecipitation
The c-Myb transcription factor is a critical regulator of proliferation and stem cell differentiation, and mutated alleles of c-Myb are oncogenic, but little is known about changes in c-Myb activity during the cell cycle. To map the association of c-Myb with specific target genes during the cell cycle, we developed a novel Fix-Sort-ChIP approach, in which asynchronously growing cells were fixed with formaldehyde, stained with Hoechst 33342 and separated into different cell cycle fractions by flow sorting, then processed for chromatin immunoprecipitation (ChIP) assays. We found that c-Myb actively repositions, binding to some genes only in specific cell cycle phases. In addition, the specificity of c-Myb is dramatically different in small subpopulations of cells, for example cells in the G2/M phase of the cell cycle, than in the bulk population. The repositioning of c-Myb during the cell cycle is not due to changes in its expression and also occurs with ectopically expressed, epitope-tagged versions of c-Myb. The repositioning occurs in established cell lines, in primary human CD34+ hematopoietic progenitors and in primary human acute myeloid leukemia cells. The combination of fixation, sorting and ChIP analysis sheds new light on the dynamic nature of gene regulation during the cell cycle and provides a new type of tool for the analysis of gene regulation in small subsets of cells, such as cells in a specific phase of the cell cycle

In this chapter, I show that the reasons for the success of Cornaro\u2019s work in early modern England lie in the fact that two very different communities of practice
saw the work\u2019s conclusions as grounded upon a particular configuration of evidence that resonated with them: one spiritual, where it was used as part of an attempt to forge a via media between Puritans and Anglicans; the other medical, where it served as a case study from which more general conclusions about how to prolong life might be extrapolated. The unique context in which the first English translation of the Discorsi was conceived, produced, and published\u2014involving some of the most prominent intellectual figures of the time, such as Francis Bacon, Nicholas Ferrar, and George Herbert\u2014make this an important case study, useful for the reconstruction of a significant chapter of the history of dieting and hygiene, and the history of conceptions of evidence and their relationship to different communities of practice
Just for the fun of it: making playgrounds accessible to all children
There seems to be growing interest worldwide in the concept of purpose-built public playground facilities that are intended specifically to provide play experiences for all children, regardless of their abilities. As this article will reveal, the complex principle of inclusion underpins the concept of shared play in inclusive playgrounds. However, despite generally being identified as important, inclusion, particularly for children with impairments within play environments, seems to be often poorly understood and sometimes overlooked in existing literature about children’s play in playgrounds. This article draws attention to the current situation surrounding public playground accessibility and teases out two discernible ways of approaching playground provision for people with impairments. The first is technically driven and mandated by guidelines and legislation. The other is child-centred or community-focused, in which participation in play in a playground is supported as a desirable social activity within a connected, inclusive community. Furthermore, this article will provide an outline of legislation and policies in Australia that are relevant to inclusive playground provision, along with some approaches that have been adopted in some other countries (notably, the United States and the United Kingdom) to provide playground access for children with impairments
