    Creative Indicators in Brief Psychotherapy

    Observation of unexpected results in Brief Psychotherapy examined in clinical supervision has instigated us to examine some of the aspects involved in the accomplished counsellings from the point of view of the creating production. We have considered the existence of a creative atmosphere that permeates the affectionate relationship between pacient and therapist, which seems to be responsible for genuine and original products of the clinical situation. From Winnicott’s conceptions of creativity, we present some considerations on the creative power of the therapeutical entailment, as well as an initial elaboration on the use of Creative Indicators as prognoses of positive outcomes on Brief Psychotherapy

    Creativity: An Integrating View

    This article searching a global view of the creative phenomenon through creative experience, as a study object, once the customary studies of creativity are presented limited to the understanding of aspects of its manifestation. Interviews have been conduted with people from different professional areas who describe their experiences with creativity, and utilizing as a theorical guide, D.W.Winnicott’s theory of emotional development, which represents na original approach to the creative phenomenon. The research introduces three new elements for the understanding of creativity: the presence of an appropriation feeling which indicates a commitment with the creative process, the existence of na internal order that rules all actions related to the experience and the existence of a space and time that inherent to the creative activity

    The Rorschach Method presents stimuli for the accomplishment of a cogniti-ve structuration task that appeases especially the imagination. Therefore, the generation ofanswers depends on the objective and subjective aspects that involve attention, perception,logical analysis, decision making processes, in other words: Creativity. From this point ofview, we selected some results from the Rorschach protocols, so as to direct an initial deba-te regarding the proposal of creative indicators offered by this method. The perspective ofan analysis in which we can verify the presence and quality of the Creative Capacity on theindividual may represent an interesting enrichment to the interpretation possibilities of thepsychological profile offered by the Rorschach Psychodiagnosis

    A global phylogenomic analysis of the shiitake genus Lentinula

    Lentinula is a broadly distributed group of fungi that contains the cultivated shiitake mushroom, L. edodes. We sequenced 24 genomes representing eight described species and several unnamed lineages of Lentinula from 15 countries on four continents. Lentinula comprises four major clades that arose in the Oligocene, three in the Americas and one in Asia–Australasia. To expand sampling of shiitake mushrooms, we assembled 60 genomes of L. edodes from China that were previously published as raw Illumina reads and added them to our dataset. Lentinula edodes sensu lato (s. lat.) contains three lineages that may warrant recognition as species, one including a single isolate from Nepal that is the sister group to the rest of L. edodes s. lat., a second with 20 cultivars and 12 wild isolates from China, Japan, Korea, and the Russian Far East, and a third with 28 wild isolates from China, Thailand, and Vietnam. Two additional lineages in China have arisen by hybridization among the second and third groups. Genes encoding cysteine sulfoxide lyase (lecsl) and γ-glutamyl transpeptidase (leggt), which are implicated in biosynthesis of the organosulfur flavor compound lenthionine, have diversified in Lentinula. Paralogs of both genes that are unique to Lentinula (lecsl 3 and leggt 5b) are coordinately up-regulated in fruiting bodies of L. edodes. The pangenome of L. edodes s. lat. contains 20,308 groups of orthologous genes, but only 6,438 orthogroups (32%) are shared among all strains, whereas 3,444 orthogroups (17%) are found only in wild populations, which should be targeted for conservation

In particular, studies of gene expression dynamics and regulation landscape in COVID-19 infected individuals are limited. Here, we report on a thorough analysis of whole blood RNA-seq data from 465 genotyped samples from the Japan COVID-19 Task Force, including 359 severe and 106 non-severe COVID-19 cases. We discover 1169 putative causal expression quantitative trait loci (eQTLs) including 34 possible colocalizations with biobank fine-mapping results of hematopoietic traits in a Japanese population, 1549 putative causal splice QTLs (sQTLs; e.g. two independent sQTLs at TOR1AIP1), as well as biologically interpretable trans-eQTL examples (e.g., REST and STING1), all fine-mapped at single variant resolution. We perform differential gene expression analysis to elucidate 198 genes with increased expression in severe COVID-19 cases and enriched for innate immune-related functions. Finally, we evaluate the limited but non-zero effect of COVID-19 phenotype on eQTL discovery, and highlight the presence of COVID-19 severity-interaction eQTLs (ieQTLs; e.g., CLEC4C and MYBL2). Our study provides a comprehensive catalog of whole blood regulatory variants in Japanese, as well as a reference for transcriptional landscapes in response to COVID-19 infection

    DOCK2 is involved in the host genetics and biology of severe COVID-19

Here we conducted a genome-wide association study (GWAS) involving 2, 393 cases of COVID-19 in a cohort of Japanese individuals collected during the initial waves of the pandemic, with 3, 289 unaffected controls. We identified a variant on chromosome 5 at 5q35 (rs60200309-A), close to the dedicator of cytokinesis 2 gene (DOCK2), which was associated with severe COVID-19 in patients less than 65 years of age. This risk allele was prevalent in East Asian individuals but rare in Europeans, highlighting the value of genome-wide association studies in non-European populations. RNA-sequencing analysis of 473 bulk peripheral blood samples identified decreased expression of DOCK2 associated with the risk allele in these younger patients. DOCK2 expression was suppressed in patients with severe cases of COVID-19. Single-cell RNA-sequencing analysis (n = 61 individuals) identified cell-type-specific downregulation of DOCK2 and a COVID-19-specific decreasing effect of the risk allele on DOCK2 expression in non-classical monocytes. Immunohistochemistry of lung specimens from patients with severe COVID-19 pneumonia showed suppressed DOCK2 expression. Moreover, inhibition of DOCK2 function with CPYPP increased the severity of pneumonia in a Syrian hamster model of SARS-CoV-2 infection, characterized by weight loss, lung oedema, enhanced viral loads, impaired macrophage recruitment and dysregulated type I interferon responses. We conclude that DOCK2 has an important role in the host immune response to SARS-CoV-2 infection and the development of severe COVID-19, and could be further explored as a potential biomarker and/or therapeutic target
