    Colour variation in signal crayfish Pacifastacus leniusculus

    External colouration in animals depends on the interaction of several different factors including the genetics and epigenetics processes that underlie the colour expression, the mechanisms of colour perception, and the general mechanisms controlling colour evolution and function. Among all, camouflage from predators and conspicuousness are of particular interest since pose animal to choose between opposite adjustment in colouration. The external colouration of crustaceans is mainly due to the accumulation of carotenoids in the exoskeleton and the epidermal layer, and the trade-off between camouflage and communication had lead to a variety of responses, involving signal partitioning, spectral sensibility, changing colouration or signalling behaviour. Here, we used digital images to explore intrapopulation variability of the external colouration of Pacifastacus leniusculus among body regions within an individual and between sexes. We found that i) ventral colouration of claws are more saturated and brilliant than upperparts, ii) males express a more saturated and brightness colouration than females, especially on the lower portion of claws, iii) colour intensity and brightness increases with size differently in different body regions, and iv) brightness is more variable in males than in females. All the above patterns support the hypothesis that colour in this species could be the result of a compromise between camouflage from predators and conspicuousness for communication. The results of this study suggest that carotenoid might have something to do with intraspecific communication and perform more complex functions than that of a simple pigment

    Hemodynamic sensor in cardiac implantable electric devices: the endocardial accelaration technology

    There have been substantial progresses in the technology of cardiac implantable electric devices (CIEDs) during the past decades. One of the progresses is represented by the development of a hemodynamic sensor embedded at the tip of a pacing lead that measures myocardial contractility by the analysis of myocardial mechanical vibrations occurring during the cardiac cycle. This sensor, providing continuous hemodynamic monitoring, could play an important role in clinical practice because of several clinical applications in CIEDs recipients. The objectives of this work are to report how this sensor operates and to review the main findings about its clinical applications

    Susceptibility of different TMEM154 genotypes in three Italian sheep breeds infected by different SRLV genotypes.

    Small ruminant lentiviruses (SRLV) belong to the Retroviridae family and can cause various diseases. One of the most impacting diseases is visna-maedi, a complex disease characterized by long latencies and chronic progressive inflammatory events affecting the nervous system, lungs, mammary gland, and articular joints. A single nucleotide polymorphism (rs408593969, c.103G>A, missense mutation E35K) in the ovine transmembrane protein gene 154 (TMEM154) was identified as protective against small ruminant lentivirus infection in different herds worldwide. However, there is evidence in the scientific literature of a breed-specificity of this protective effect and, furthermore, there are still limited studies regarding the association between the animal genotype and the infecting virus genotype. Thus, the aim of this study was to further investigate the association between the animal genotype for the suggested protective mutation and the infecting virus genotype, in three different sheep breeds reared in northern Italy. The results obtained only partially confirmed the data available in the literature, as the protective effect was confirmed only for SRLV genotype A clusters, while other genotypes (namely B and E) infected AA and GA animals. Further studies with an experimental infection of specific virus genotypes in hosts with specific genotypes are required to confirm the larger number of cases the results obtained in this study

    Potentially Toxic Elements (PTEs) in Cultivated Soils from Lombardy (Northern Italy): Spatial Distribution, Origin, and Management Implications

    This work aims to quantify the contribution of agricultural practices to diffuse pollution in cultivated soils. Two samples (the first from the top layer and the second from a depth of about 100 cm) were retrieved from about 1000 soil profiles and analyzed for their potentially toxic elements (PTEs) content (Cd, Cu, Ni, Pb, Zn, Mn). In addition, we collected dedicated soil profiles for three specific types of land use and agronomic practice, namely vine-, cereal- and rice-growing areas. Baseline concentrations and distribution maps were produced. Statistical data treatment, coupled with the results of the dedicated soil profiles, enabled the identification of precise pollution sources and processes. In cultivated soils, PTEs contents prove to be generally well below the Italian regulatory limits, with the exception of Cu in vineyard soils, due to the long-lasting use of Cu-based pesticides. South of Milan and in other urban areas, we attribute the top soil enrichment in Cd, Cu, Pb, and Zn to industrial activities and urban settlement. Ni sporadically exceeds regulatory standards, but its association with Mn and its depth distribution point to a natural origin, mostly due to the occurrence of serpentine. The project demonstrates that although agriculture, especially in the form of vineyards and of intensive cultivation, certainly contributes to pollution, such contribution is exceeded by that deriving from industry and road traffic

    A Practical Application of Genomic Predictions for Mastitis Resistance in Italian Holstein Heifers

    SIMPLE SUMMARY: Selecting the best animals for farms has always been a fundamental aspect of animal breeding. To assist in the traditional selection performed on phenotypes and to further improve genetic selection as performed in recent years, genomic tools could be applied to select animals early in their productive careers. The aim of this study was to evaluate the application of a commercial genomic tool on animals selected from farms grouped by their average somatic cell count. The obtained results showed that farms with good animal management practices also rear animals with better genomic indexes, probably due to selection on other well-established indexes (e.g., productive traits). Selecting heifers based on their wellness genomic indexes would further improve both their economic value and their disease resistance and resilience. ABSTRACT: Heifers are a fundamental resource on farms, and their importance is reflected in both farm management and economy. Therefore, the selection of heifers to be reared on a farm should be carefully performed to select only the best animals. Genomic selection is available nowadays to evaluate animals in a fast and economic way. However, it is mainly used on the sire line and on performance traits. Ten farms were selected based on their 5-year records of average somatic cell count and evenly classified into high (>300,000 cells/mL) and low somatic cell count (<150,000 cells/mL). Genomic indexes (regarding both wellness and productive traits) were evaluated in 157 Italian Holstein heifers reared in the selected ten farms (90 from high-cells farms and 67 from low-cells ones). Linear mixed models were fitted to analyze the effects of the abovementioned genomic indexes on related phenotypes. Results have shown that farms classified into low somatic cell count had an overall better animal genomic pool compared to high somatic cell count ones. Additionally, the results shown in this study highlighted a difference in wellness genomic indexes in animals from farms with either a high or a low average somatic cell count. Applying genomic tools directly to heifer selection could improve economic aspects related to herd turnover

    The histone deacetylase inhibitor romidepsin synergizes with lenalidomide and enhances tumor cell death in T-cell lymphoma cell lines

    We investigated the cytotoxic interactions of romidepsin, a histone deacetylase inhibitor, and lenalidomide, an immunomodulatory agent, in a T-cell lymphoma preclinical model. Hut-78 and Karpas-299 cells were treated with romidepsin and lenalidomide alone and in combination. The interaction between romidepsin and lenalidomide was evaluated by the Chou–Talalay method, and cell viability and clonogenicity were also evaluated. Apoptosis, reactive oxygen species (ROS) levels, and cell cycle distribution were determined by flow cytometry. ER stress, caspase activation, and the AKT, MAPK/ERK, and STAT-3 pathways were analyzed by Western blot. Combination treatment with romidepsin and lenalidomide had a synergistic effect in Hut-78 cells and an additive effect in Karpas-299 cells at 24 hours and did not decrease the viability of normal peripheral blood mononuclear cells. This drug combination induced apoptosis, increased ROS production, and activated caspase-8, −9, −3 and PARP. Apoptosis was associated with increased hallmarks of ER stress and activation of UPR sensors and was mediated by dephosphorylation of the AKT, MAPK/ERK, and STAT3 pathways.The combination of romidepsin and lenalidomide shows promise as a possible treatment for T-cell lymphoma. This work provides a basis for further studies

    Doppler assessment of aortic stenosis: a 25-operator study demonstrating why reading the peak velocity is superior to velocity time integral

    Aims Measurements with superior reproducibility are useful clinically and research purposes. Previous reproducibility studies of Doppler assessment of aortic stenosis (AS) have compared only a pair of observers and have not explored the mechanism by which disagreement between operators occurs. Using custom-designed software which stored operators’ traces, we investigated the reproducibility of peak and velocity time integral (VTI) measurements across a much larger group of operators and explored the mechanisms by which disagreement arose. Methods and results Twenty-five observers reviewed continuous wave (CW) aortic valve (AV) and pulsed wave (PW) left ventricular outflow tract (LVOT) Doppler traces from 20 sequential cases of AS in random order. Each operator unknowingly measured each peak velocity and VTI twice. VTI tracings were stored for comparison. Measuring the peak is much more reproducible than VTI for both PW (coefficient of variation 10.1 vs. 18.0%; P < 0.001) and CW traces (coefficient of variation 4.0 vs. 10.2%; P < 0.001). VTI is inferior because the steep early and late parts of the envelope are difficult to trace reproducibly. Dimensionless index improves reproducibility because operators tended to consistently over-read or under-read on LVOT and AV traces from the same patient (coefficient of variation 9.3 vs. 17.1%; P < 0.001). Conclusion It is far more reproducible to measure the peak of a Doppler trace than the VTI, a strategy that reduces measurement variance by approximately six-fold. Peak measurements are superior to VTI because tracing the steep slopes in the early and late part of the VTI envelope is difficult to achieve reproducibly
