    Angular sensitivity of blowfly photoreceptors: intracellular measurements and wave-optical predictions

    The angular sensitivity of blowfly photoreceptors was measured in detail at wavelengths λ = 355, 494 and 588 nm. The measured curves often showed numerous sidebands, indicating the importance of diffraction by the facet lens. The shape of the angular sensitivity profile is dependent on wavelength. The main peak of the angular sensitivities at the shorter wavelengths was flattened. This phenomenon as well as the overall shape of the main peak can be quantitatively described by a wave-optical theory using realistic values for the optical parameters of the lens-photoreceptor system. At a constant response level of 6 mV (almost dark adapted), the visual acuity of the peripheral cells R1-6 is at longer wavelengths mainly diffraction limited, while at shorter wavelengths the visual acuity is limited by the waveguide properties of the rhabdomere. Closure of the pupil narrows the angular sensitivity profile at the shorter wavelengths. This effect can be fully described by assuming that the intracellular pupil progressively absorbs light from the higher order modes. In light-adapted cells R1-6 the visual acuity is mainly diffraction limited at all wavelengths.

    Expression microarray analysis of papillary thyroid carcinoma and benign thyroid tissue: emphasis on the follicular variant and potential markers of malignancy

    The most common sub-variant of papillary thyroid carcinoma (PTC) is the so-called follicular variant (FVPTC), which is a particularly problematic lesion and can be challenging from a diagnostic viewpoint even in resected lesions. Although fine needle aspiration cytology is very useful in the diagnosis of PTC, its accuracy and utility would be greatly facilitated by the development of specific markers for PTC and its common variants. We used the recently developed Applied Biosystems 1700 microarray system to interrogate a series of 11 benign thyroid lesions and conditions and 14 samples of PTC (six with classic morphology and eight with follicular variant morphology). TaqMan® reverse transcriptase-polymerase chain reaction was used to validate the expression portfolios of 50 selected transcripts. Our data corroborates potential biomarkers previously identified in the literature, such as LGALS3, S100A11, LYN, BAX, and cluster of differentiation 44 (CD44). However, we have also identified numerous transcripts never previously implicated in thyroid carcinogenesis, and many of which are not represented on other microarray platforms. Diminished expression of metallothioneins featured strongly among these and suggests a possible role for this family as tumour suppressors in PTC. Fifteen transcripts were significantly associated with FVPTC morphology. Surprisingly, these genes were associated with an extremely narrow repertoire of functions, including the major histocompatibility complex and cathepsin families

    Evaluating multi-year, multi-site data on the energy balance closure of eddy-covariance flux measurements at cropland sites in southwestern Germany

    The energy balance of eddy-covariance (EC) measurements is typically not closed, resulting in one of the main challenges in evaluating and interpreting EC flux data. Energy balance closure (EBC) is crucial for validating and improving regional and global climate models. To investigate the nature of the gap in EBC for agroecosystems, we analyzed EC measurements from two climatically contrasting regions (Kraichgau – KR – and Swabian Jura – SJ) in southwestern Germany. Data were taken at six fully equipped EC sites from 2010 to 2017. The gap in EBC was quantified by ordinary linear regression, relating the energy balance ratio (EBR), calculated as the quotient of turbulent fluxes and available energy, to the residual energy term. In order to examine potential reasons for differences in EBC, we compared the EBC under varying environmental conditions and investigated a wide range of possible controls. Overall, the variation in EBC was found to be higher during winter than summer. Moreover, we determined that the site had a statistically significant effect on EBC but no significant effect on either crop or region (KR vs SJ). The time-variable footprints of all EC stations were estimated based on data measured in 2015, complimented by micro-topographic analyses along the prevailing wind direction. The smallest mean annual energy balance gap was 17&thinsp;% in KR and 13&thinsp;% in SJ. Highest EBRs were mostly found for winds from the prevailing wind direction. The spread of EBRs distinctly narrowed under unstable atmospheric conditions, strong buoyancy, and high friction velocities. Smaller footprint areas led to better EBC due to increasing homogeneity. Flow distortions caused by the back head of the anemometer negatively affected EBC during corresponding wind conditions.</p

    Targeting deforestation rates in climate change policy: a "Preservation Pathway" approach

    We present a new methodological approach to incorporating deforestation within the international climate change negotiating regime. The approach, called "Preservation Pathway" combines the desire for forest preservation with the need to reduce emissions associated with forest loss by focusing on the relative rate of change of forest cover as the criteria by which countries gain access to trading preserved forest carbon stocks. This approach avoids the technically challenging task of quantifying historical or future deforestation emission baselines. Rather, it places emphasis on improving quantification of contemporary stocks and the relative decline in deforestation rates necessary to preserve those stocks. This approach places emphasis on the complete emissions trajectory necessary to attain an agreed-upon preserved forest and as such, meets both forest conservation and climate goals simultaneously
