    Tropical cyanobacterial blooms: A review of prevalence, problem taxa, toxins and influencing environmental factors

    © 2015, Page Press Publications. All rights reserved. Toxic cyanobacterial blooms are a major issue in freshwater systems in many countries. The potentially toxic species and their ecological causes are likely to be different in tropical zones from those in temperate water bodies; however, studies on tropical toxic cyanobacterial blooms are sporadic and currently there is no global synthesis. In this review, we examined published information on tropical cyanobacterial bloom occurrence and toxin production to investigate patterns in their growth and distribution. Microcystis was the most frequently occurring bloom genus throughout tropical Asia, Africa and Central America, while Cylindrospermopsis and Anabaena blooms occurred in various locations in tropical Australia, America and Africa. Microcystis blooms were more prevalent during the wet season while Cylindrospermopsis blooms were more prevalent during the dry period. Microcystin was the most encountered toxin throughout the tropics. A meta-analysis of tropical cyanobacterial blooms showed that Microcystis blooms were more associated with higher total nitrogen concentrations, while Cylindrospermopsis blooms were more associated with higher maximum temperatures. Meta-analysis also showed a positive linear relationship between levels of microcystin and N:P (nitrate:phosphate) ratio. Tropical African Microcystis blooms were found to have the lowest microcystin levels in relation to biomass and N:P (nitrate:phosphate) compared to tropical Asian, Australian and American blooms. There was also no significant correlation between microcystin concentration and cell concentration for tropical African blooms as opposed to tropical Asian and American blooms. Our review illustrates that some cyanobacteria and toxins are more prevalent in tropical areas.     Global small RNA analysis in fast-growing Arabidopsis thaliana with elevated concentrations of ATP and sugars

    BACKGROUND: In higher eukaryotes, small RNAs play a role in regulating gene expression. Overexpression (OE) lines of Arabidopsis thaliana purple acid phosphatase 2 (AtPAP2) were shown to grow faster and exhibit higher ATP and sugar contents. Leaf microarray studies showed that many genes involved in microRNAs (miRNAs) and trans-acting siRNAs (tasiRNAs) biogenesis were significantly changed in the fast-growing lines. In this study, the sRNA profiles of the leaf and the root of 20-day-old plants were sequenced and the impacts of high energy status on sRNA expression were analyzed. RESULTS: 9-13 million reads from each library were mapped to genome. miRNAs, tasiRNAs and natural antisense transcripts-generated small interfering RNAs (natsiRNAs) were identified and compared between libraries. In the leaf of OE lines, 15 known miRNAs increased in abundance and 9 miRNAs decreased in abundance, whereas in the root of OE lines, 2 known miRNAs increased in abundance and 9 miRNAs decreased in abundance. miRNAs with increased abundance in the leaf and root samples of both OE lines (miR158b and miR172a/b) were predicted to target mRNAs coding for Dof zinc finger protein and Apetala 2 (AP2) proteins, respectively. Furthermore, a significant change in the miR173-tasiRNAs-PPR/TPR network was observed in the leaves of both OE lines. CONCLUSION: In this study, the impact of high energy content on the sRNA profiles of Arabidopsis is reported. While the abundance of many stress-induced miRNAs is unaltered, the abundance of some miRNAs related to plant growth and development (miR172 and miR319) is elevated in the fast-growing lines. An induction of miR173-tasiRNAs-PPR/TPR network was also observed in the OE lines. In contrast, only few cis- and trans-natsiRNAs are altered in the fast-growing lines.published_or_final_versio

    Review: revisiting the human cholinergic nucleus of the diagonal band of Broca

    Although the nucleus of the vertical limb of the diagonal band of Broca (nvlDBB) is the second largest cholinergic nucleus in the basal forebrain, after the nucleus basalis of Meynert (nbM), it has not generally been a focus for studies of neurodegenerative disorders. However, the nvlDBB does have an important projection to the hippocampus and discrete lesions of the rostral basal forebrain have been shown to disrupt retrieval memory function, a major deficit seen in patients with Lewy body disorders. One reason for its neglect is that the anatomical boundaries of the nvlDBB are ill defined and this area of the brain is not part of routine diagnostic sampling protocols. We have reviewed the history and anatomy of the nvlDBB and now propose guidelines for distinguishing nvlDBB from other neighbouring cholinergic cell groups for standardising future clinicopathological work. Thorough review of the literature regarding neurodegenerative conditions reveals inconsistent results in terms of cholinergic neuronal loss within the nvlDBB. This is likely to be due to the use of variable neuronal inclusion criteria and omission of cholinergic immunohistochemical markers. Extrapolating from those studies showing significant nvlDBB neuronal loss in Lewy body dementia, we propose an anatomical and functional connection between the cholinergic component of the nvlDBB (Ch2) and the CA2 subfield in the hippocampus which may be especially vulnerable in Lewy body disorders.     Cardiovascular health status between standard and nonstandard workers in Korea

    OBJECTIVES: The effect of employment insecurity on employee health is an important public health issue due to the recent effects of neoliberalism and the global financial crisis (2007-2008) on labor markets. This study aims to evaluate the differences in cardiovascular health status and the use of preventive screening services between standard and nonstandard workers. METHODS: Waged employees (N = 5,338) between the ages of 20 and 64 were grouped into standard (full-time, permanent) and nonstandard (part-time, temporary, or daily) employees. Data from the Fourth Korea National Health and Nutrition Examination Survey, 2007-2009, a nationwide representative survey, were examined, including cardiovascular health risk behaviors (tobacco, alcohol, physical inactivity), measured morbidities (blood pressure, blood glucose level, lipid profiles, body mass index), and the use of screening services for hypertension and diabetes mellitus. RESULTS: Female nonstandard employees tended to have higher blood pressure than did female standard employees (adjusted odds ratio, aOR 1.42, 95% confidence interval, CI 1.02 to 1.98). However, nonstandard employees (both men and women) were less likely to use preventive screening services for hypertension (aOR 0.72, 95% CI 0.54 to 0.94 in men; aOR 0.56, 95% CI 0.43 to 0.73 in women) and diabetes (aOR 0.58, 95% CI 0.43 to 0.79 in men; aOR 0.55, 95% CI 0.43 to 0.71 in women). CONCLUSION: Nonstandard work is associated with the underuse of screening services and poorer cardiovascular health in a specific population. Policies to reduce employment insecurity and encourage nonstandard employees to receive health screening services should be prioritized

    Combination of BMI1 and MAPK/ERK inhibitors is effective in medulloblastoma.

    BACKGROUND: Epigenetic changes play a key role in the pathogenesis of medulloblastoma (MB), the most common malignant paediatric brain tumour. METHODS: We explore the therapeutic potential of BMI1 and MAPK/ERK inhibition in BMI1 High;CHD7 Low MB cells and in a pre-clinical xenograft model. RESULTS: We identify a synergistic vulnerability of BMI1 High;CHD7 Low MB cells to a combination treatment with BMI1 and MAPK/ERK inhibitors. Mechanistically, CHD7-dependent binding of BMI1 to MAPK-regulated genes underpins the CHD7-BMI1-MAPK regulatory axis responsible of the anti-tumour effect of the inhibitors in vitro and in a pre-clinical mouse model. Increased ERK1 and ERK2 phosphorylation activity is found in BMI1 High;CHD7 Low G4 MB patients, raising the possibility that they could be amenable to a similar therapy. CONCLUSIONS: The molecular dissection of the CHD7-BMI1-MAPK regulatory axis in BMI1 High;CHD7 Low MB identifies this signature as a proxy to predict MAPK functional activation, which can be effectively drugged in preclinical models, and paves the way for further exploration of combined BMI1 and MAPK targeting in G4 MB patients

    Case Report: Approach to Dog Bite Eyelid Injury in Rabies Endemic Area

    Sarawak has recorded 24 rabies cases, since an outbreak was declared on July 1, 2017. We are presenting a case report dis-cussing the approach to dog bite eyelid laceration in Rabies endemic area. A 13-year-old boy came with full-thickness lid lacera-tion wound, after being attacked by a stray dog. The patient received a dose of intramuscular anti-tetanus injection, followed by immunoglobulin injection at the laceration site, rabies vaccine as well as completed a course of antibiotics. The laceration was allowed for primary closure, and it healed well with adequate apposition. This paper we outlined the importance of immediate accurate risk category stratification for early identification of those at risk in the primary care setting help minimize the risk of rabies infection and save a life
