    This theoretical study aims to carry out similarities between the philosophical thoughts of Walter Benjamin and sociological reflections of Manuel Jacinto Sarmento about childhood, recursive temporality, childish experiences and cultures. For this, we divided our exposition into four sessions that aim: to highlight the childhood as historical category in the modernity; to present Benjamin’s look at the approach; to highlight the prospect through the Sarmento’s optical; Finally, to approximate the concepts.     A novel scytalidium species : understand the cellulolytic system for biomass saccharification

    In order to overcome the bottlenecks related to lignocellulosic-derived sugars, the search for more efficient enzymatic cocktails, containing a broad-spectrum of specific activities, relies on an important feature. This paper describes new enzyme activities derived from the novel strain of the Scytalidium genus isolated from the Amazonas rainforest. The production of the enzymatic cocktail was induced by delignifiedhydrothermal bagasse (DHB), and yeast extract was used to improve secretion activities, resulting in a positive influence on total cellulase activity. The enzymatic cocktail produced by this novel strain contains specific activities for biomass degradation, including FPAse, xylanase and β-glucosidase. Moreover, it is capable of hydrolyzing 62% of the alkaline pretreated bagasse, surpassing in 14% the hydrolytic capability achieved by the commercial cocktail Celluclast.     Static Observers in Curved Spaces and Non-inertial Frames in Minkowski Spacetime

    Static observers in curved spacetimes may interpret their proper acceleration as the opposite of a local gravitational field (in the Newtonian sense). Based on this interpretation and motivated by the equivalence principle, we are led to investigate congruences of timelike curves in Minkowski spacetime whose acceleration field coincides with the acceleration field of static observers of curved spaces. The congruences give rise to non-inertial frames that are examined. Specifically we find, based on the locality principle, the embedding of simultaneity hypersurfaces adapted to the non-inertial frame in an explicit form for arbitrary acceleration fields. We also determine, from the Einstein equations, a covariant field equation that regulates the behavior of the proper acceleration of static observers in curved spacetimes. It corresponds to an exact relativistic version of the Newtonian gravitational field equation. In the specific case in which the level surfaces of the norm of the acceleration field of the static observers are maximally symmetric two-dimensional spaces, the energy-momentum tensor of the source is analyzed.Comment: 28 pages, 4 figures

    Selective disruption of Tcf7l2 in the pancreatic β cell impairs secretory function and lowers β cell mass.

    Type 2 diabetes (T2D) is characterized by β cell dysfunction and loss. Single nucleotide polymorphisms in the T-cell factor 7-like 2 (TCF7L2) gene, associated with T2D by genome-wide association studies, lead to impaired β cell function. While deletion of the homologous murine Tcf7l2 gene throughout the developing pancreas leads to impaired glucose tolerance, deletion in the β cell in adult mice reportedly has more modest effects. To inactivate Tcf7l2 highly selectively in β cells from the earliest expression of the Ins1 gene (∼E11.5) we have therefore used a Cre recombinase introduced at the Ins1 locus. Tcfl2(fl/fl)::Ins1Cre mice display impaired oral and intraperitoneal glucose tolerance by 8 and 16 weeks, respectively, and defective responses to the GLP-1 analogue liraglutide at 8 weeks. Tcfl2(fl/fl)::Ins1Cre islets displayed defective glucose- and GLP-1-stimulated insulin secretion and the expression of both the Ins2 (∼20%) and Glp1r (∼40%) genes were significantly reduced. Glucose- and GLP-1-induced intracellular free Ca(2+) increases, and connectivity between individual β cells, were both lowered by Tcf7l2 deletion in islets from mice maintained on a high (60%) fat diet. Finally, analysis by optical projection tomography revealed ∼30% decrease in β cell mass in pancreata from Tcfl2(fl/fl)::Ins1Cre mice. These data demonstrate that Tcf7l2 plays a cell autonomous role in the control of β cell function and mass, serving as an important regulator of gene expression and islet cell coordination. The possible relevance of these findings for the action of TCF7L2 polymorphisms associated with Type 2 diabetes in man is discussed

    Single Nucleotide Variants And Indels Identified From Whole-genome Re-sequencing Of Guzerat, Gyr, Girolando And Holstein Cattle Breeds

    Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq)Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES)Whole-genome re-sequencing, alignment and annotation analyses were undertaken for 12 sires representing four important cattle breeds in Brazil: Guzerat (multi-purpose), Gyr, Girolando and Holstein (dairy production). A total of approximately 4.3 billion reads from an Illumina HiSeq 2000 sequencer generated for each animal 10.7 to 16.4-fold genome coverage. A total of 27,441,279 single nucleotide variations (SNVs) and 3,828,041 insertions/deletions (InDels) were detected in the samples, of which 2,557,670 SNVs and 883,219 InDels were novel. The submission of these genetic variants to the dbSNP database significantly increased the number of known variants, particularly for the indicine genome. The concordance rate between genotypes obtained using the Bovine HD BeadChip array and the same variants identified by sequencing was about 99.05%. The annotation of variants identified numerous non-synonymous SNVs and frameshift InDels which could affect phenotypic variation. Functional enrichment analysis was performed and revealed that variants in the olfactory transduction pathway was over represented in all four cattle breeds, while the ECM-receptor interaction pathway was over represented in Girolando and Guzerat breeds, the ABC transporters pathway was over represented only in Holstein breed, and the metabolic pathways was over represented only in Gyr breed.     Study On The Introgression Of Beef Breeds In Canchim Cattle Using Single Nucleotide Polymorphism Markers

    Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES)Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq)The aim of this study was to evaluate the level of introgression of breeds in the Canchim (CA: 62.5% Charolais-37.5% Zebu) and MA genetic group (MA: 65.6% Charolais-34.4% Zebu) cattle using genomic information on Charolais (CH), Nelore (NE), and Indubrasil (IB) breeds. The number of animals used was 395 (CA and MA), 763 (NE), 338 (CH), and 37 (IB). The Bovine 50SNP BeadChip from Illumina panel was used to estimate the levels of introgression of breeds considering the Maximum likelihood, Bayesian, and Single Regression method. After genotype quality control, 32,308 SNPs were considered in the analysis. Furthermore, three thresholds to prune out SNPs in linkage disequilibrium higher than 0.10, 0.05, and 0.01 were considered, resulting in 15,286, 7,652, and 1,582 SNPs, respectively. For k = 2, the proportion of taurine and indicine varied from the expected proportion based on pedigree for all methods studied. For k = 3, the Regression method was able to differentiate the animals in three main clusters assigned to each purebred breed, showing more reasonable according to its biological viewpoint. Analyzing the data considering k = 2 seems to be more appropriate for Canchim-MA animals due to its biological interpretation. The usage of 32,308 SNPs in the analyses resulted in similar findings between the estimated and expected breed proportions. Using the Regression approach, a contribution of Indubrasil was observed in Canchim-MA when k = 3 was considered. Genetic parameter estimation could account for this breed composition information as a source of variation in order to improve the accuracy of genetic models. Our findings may help assemble appropriate reference populations for genomic prediction for Canchim-MA in order to improve prediction accuracy. Using the information on the level of introgression in each individual could also be useful in breeding or crossing design to improve individual heterosis in crossbred cattle. © 2017 Buzanskas et al.     Hydrogen-Helium Mixtures at High Pressure

    The properties of hydrogen-helium mixtures at high pressure are crucial to address important questions about the interior of Giant planets e.g. whether Jupiter has a rocky core and did it emerge via core accretion? Using path integral Monte Carlo simulations, we study the properties of these mixtures as a function of temperature, density and composition. The equation of state is calculated and compared to chemical models. We probe the accuracy of the ideal mixing approximation commonly used in such models.
