    A Dynamical Study of Galaxies in the Hickson Compact Groups

    In order to investigate dynamical properties of spiral galaxies in the Hickson compact groups (HCGs), we present rotation curves of 30 galaxies in 20 HCGs. We found as follows. 1) There is not significant relation between dynamical peculiarity and morphological peculiarity in HCG spirals. 2) There is no significant relation between the dynamical properties and the frequency distribution of nuclear activities in HCG spirals. 3) There are no significant correlations between the dynamical properties of HCG spirals and any group properties (i.e., the size, the velocity dispersion, the galaxy number density, and the crossing time). 4) Asymmetric and peculiar rotation curves are more frequently seen in the HCG spirals than in field spirals and in cluster ones. However, this tendency is more obviously seen in late-type HCG spirals. These results suggest that the dynamical properties of HCG spirals do not strongly correlate with the morphology, the nuclear activity, and the group properties.     The Nuclear Activity of the Galaxies in the Hickson Compact Groups

    In order to investigate the nuclear activity of galaxies residing in compact groups of galaxies, we present results of our optical spectroscopic program made at Okayama Astrophysical Observatory. We have performed optical spectroscopy of 69 galaxies which belong to 31 Hickson Compact Groups (HCGs) of Galaxies. Among them, three galaxies have discordant redshifts. Further, spectral quality is too poor to classify other three galaxies. Therefore, we describe our results for the remaining 63 galaxies. Our main results are summarized below. (1) We have found in our sample; 28 AGN, 16 HII nuclei, and 19 normal galaxies which show no emission line. We used this HCG sample for statistical analyses. (2) Comparing the frequency distributions of activity types between the HCGs and the field galaxies whose data are taken from Ho, Filippenko, & Sargent (382 field galaxies), we find that the frequency of HII nuclei in the HCGs is significantly less than that in the field. However, this difference may be due to selection bias that our HCG sample contains more early-type galaxies than the field, because it is known that HII nuclei are rarer in early-type galaxies than in later ones. (3) Applying correction this morphological bias to the HCG sample, we find that there is no statistically significant difference in the frequency of occurrence of emission-line galaxies between the HCGs and the field. This implies that the dense galaxy environment in the HCGs does not affect triggering both the AGN activity and the nuclear starburst.     Hemolytic C-Type Lectin CEL-III from Sea Cucumber Expressed in Transgenic Mosquitoes Impairs Malaria Parasite Development

    The midgut environment of anopheline mosquitoes plays an important role in the development of the malaria parasite. Using genetic manipulation of anopheline mosquitoes to change the environment in the mosquito midgut may inhibit development of the malaria parasite, thus blocking malaria transmission. Here we generate transgenic Anopheles stephensi mosquitoes that express the C-type lectin CEL-III from the sea cucumber, Cucumaria echinata, in a midgut-specific manner. CEL-III has strong and rapid hemolytic activity toward human and rat erythrocytes in the presence of serum. Importantly, CEL-III binds to ookinetes, leading to strong inhibition of ookinete formation in vitro with an IC50 of 15 nM. Thus, CEL-III exhibits not only hemolytic activity but also cytotoxicity toward ookinetes. In these transgenic mosquitoes, sporogonic development of Plasmodium berghei is severely impaired. Moderate, but significant inhibition was found against Plasmodium falciparum. To our knowledge, this is the first demonstration of stably engineered anophelines that affect the Plasmodium transmission dynamics of human malaria. Although our laboratory-based research does not have immediate applications to block natural malaria transmission, these findings have significant implications for the generation of refractory mosquitoes to all species of human Plasmodium and elucidation of mosquito–parasite interactions

    New Surgical Procedure for Pancreas Head

    In this study, we demonstrate two new methods for pancreaticoduodenectomy (PD). One method is the mini‐laparotomic PD by Shuriken‐shaped umbilicoplasty with the real‐time moving window‘s method. The other method is the new pancreaticojejunostomy (PJ) by punctured stent slide guiding method (PSSGM). This procedure could be performed by complete mini‐laparotomy under direct vision, and the final major wound is only 2 cm of round navel. PSSGM prevents the difference of caliber between pancreatic anastomosis and the inside out of jejunal mucosa in theory. Ten cases of mini‐lap PD were successfully performed under new PJ anastomosis. The pancreatic leakage (PL) was only one case of ISGPF grade A, and its frequency was 9% (1/11). Our mini‐lap PD by Shuriken‐shaped umbilicoplasty might be a useful way for overcoming the obstacles about safety, complication risk, cosmetic demand, and medical cost compared to laparoscopic PD. Also, our new device of PJ reconstruction by PSSGM might be an easy and useful device for the prevention of PL

    OBJECTIVE: To assess whether the phosphorylated Fas-associated death domain protein (FADD) at 194 serine (p-FADD) is valuable as a marker of biochemical recurrence in hormone-naive patients who had undergone radical prostatectomy. MATERIALS AND METHODS: We used radical prostatectomy specimens from 106 patients. None of the patients had received neoadjuvant or adjuvant therapy. The percentage of positive p-FADD cells (nuclear staining) was immunohistochemically evaluated. The correlation between FADD phosphorylation and the clinicopathologic parameters was assessed. The correlation between the biochemical recurrence-free rate and the p-FADD expression level was analyzed using the Kaplan-Meier method. RESULTS: Overall, 39 patients developed biochemical recurrence. We investigated the expression of p-FADD in 106 patients with prostate cancer using immunohistochemistry. We compared our findings with the clinicopathologic parameters, including the follow-up data. Patients with a greater positive p-FADD rate had a significantly lower biochemical recurrence rate than those with a lower positive p-FADD rate (P < .001). A significant inverse correlation was found between the positive p-FADD rate and the Gleason score. CONCLUSION: A low expression of p-FADD could be a predictor of biochemical recurrence in hormone-naive patients who have undergone radical prostatectomy.博士(医学)・乙第1313号・平成25年5月29

    Effects of 3-styrylchromones on metabolic profiles and cell death in oral squamous cell carcinoma cells

    Abstract4H-1-benzopyran-4-ones (chromones) are important naturally-distributing compounds. As compared with flavones, isoflavones and 2-styrylchromones, there are only few papers of 3-styrylchromones that have been published. We have previously reported that among fifteen 3-styrylchromone derivatives, three new synthetic compounds that have OCH3 group at the C-6 position of chromone ring, (E)-3-(4-hydroxystyryl)-6-methoxy-4H-chromen-4-one (compound 11), (E)-6-methoxy-3-(4-methoxystyryl)-4H-chromen-4-one (compound 4), (E)-6-methoxy-3-(3,4,5-trimethoxystyryl)-4H-chromen-4-one (compound 6) showed much higher cytotoxicities against four epithelial human oral squamous cell carcinoma (OSCC) lines than human normal oral mesenchymal cells. In order to further confirm the tumor specificities of these compounds, we compared their cytotoxicities against both human epithelial malignant and non-malignant cells, and then investigated their effects on fine cell structures and metabolic profiles and cell death in human OSCC cell line HSC-2. Cytotoxicities of compounds 4, 6, 11 were assayed with MTT method. Fine cell structures were observed under transmission electron microscope. Cellular metabolites were extracted with methanol and subjected to CE-TOFMS analysis. Compounds 4, 6, 11 showed much weaker cytotoxicity against human oral keratinocyte and primary human gingival epithelial cells, as compared with HSC-2, confirming their tumor-specificity, whereas doxorubicin and 5-FU were highly cytotoxic to these normal epithelial cells, giving unexpectedly lower tumor-specificity. The most cytotoxic compound 11, induced the mitochondrial vacuolization, autophagy suppression followed by apoptosis induction, and changes in the metabolites involved in amino acid and glycerophospholipid metabolisms. Chemical modification of lead compound 11 may be a potential choice for designing new type of anticancer drugs
