    Maintenance of Genome Stability by Ubiquitination of DNA Repair Proteins in Mammalian Development and Disease

    To maintain genome DNA, DNA repair machinery has been developed in cellular life cycle. Multiple DNA repair pathways such as base excision repair, nucleotide excision repair, DNA cross link damage repair, DNA single strand break repair and DNA double strand break repair including nonhomologous end joining and homologous recombination are regulated by protein signal cascade. Because of limited gene number, protein posttranslational modification signal has advantage to control cell dynamics during development and senescence. This chapter focuses on how DNA repair proteins molecular modification including phosphorylation and ubiquitination contribute to genome stability pathway during mammalian development and disease

    Some Properties of O-Methyltransferase from Bamboo Shoot

    この論文は国立情報学研究所の学術雑誌公開支援事業により電子化されました。The incubation of CA, 5-HFA and TCA with sliced bamboo shoot in the presence of methionine-^CH_3 gave radioactive FA and SA. On the other hand, the incubation of those compounds with sliced gingko shoot yielded only FA from CA and did not SA from 5-HFA and TCA. The enzyme preparation obtained from bamboo shoot was fractionated by (NH_4)_2SO_4-and iso-electric point precipitations and column chromatographies on DEAE-cellulose. Variation of the ratios (SA/FA) which were obtained by assay of O-methyltransferase are indicative of possible presence of two enzymes in the bamboo enzyme preparation. The results are discussed in relation to the biochemical differences of the methoxyl patterns in lignins between angiosperms and gymnosperms

    Activation Pattern of Langerhans Cells in the Afferent and Efferent Phases of Contact Hypersensitivity

    Langerhans cells are MHC class II (Ia) positive antigen- presenting cells that play a crucial role in the induction of contact hypersensitivity (CHS). The topical application of a hapten modifies the cell surface moieties of Langerhans cells, and activates Langerhans cells to increase their size and Ia intensity. The haptenated and activated Langerhans cells emigrate from the epidermis and thus the in situ density of Langerhans cells usually decreases during 24-48h after the hapten application in CHS. To determine whether the early activation pattern of Langerhans cells is different between the afferent phase and the efferent phase of CHS, we compared the density and morphologic changes of Langerhans cells in CHS to trinitrochlorobenzene using nonsensitized and sensitized mice. We found that the application of a hapten induces more significant enlargement of Langerhans cell size in the afferent phase than in the efferent phase, whereas the reduction of Langerhans cell density is more marked in the efferent than in the afferent phase of CHS. Moreover, topical immunosuppressive drugs inhibit the in situ activation of Langerhans cells
