Flow Analysis through Collector Well Laterals: A Case Study from Sonoma County Water Agency, California
The Sonoma County Water Agency (SWCA) uses six radial collector wells along the Russian River west of Santa Rosa, to provide water for several municipalities and water districts in north-western California. Three collector wells (1, 2, and 6) are located in the Wohler area, and three collector wells (3, 4, and 5) are located in the Mirabel area. The objective of this paper is to highlight the performance of the three collector wells located in the Mirabel area since their construction. The 2015 investigation showed a lower performance of Collectors 3 and 4 compared to their original performances after construction in 1975, while the performance of Collector 5 was relatively stable since 1982. The potential change in capacity could be due to the increase in encrustation observed during the visual inspection of laterals in all three collector wells. Overall, the three collectors are still within the optimal design parameters (screen entrance velocity \u3c 0.305 m min-1 and axial flow velocity of lateral screens \u3c 1.524 m s-1)

(The following is an edited transcript of the proceedings of the section on Employment Discrimination Law at the AALS Annual Meeting, New Orleans, Louisiana, January 9, 1999.)
DOUGLAS D. SCHERER*: Good morning. The program description asks the question, Is there a disconnect between existing EEO jurisprudence and the realities of the workplace and workforce of the Twenty-First Century? Societal disapproval of employment discrimination is reflected in federal EEO laws that have been enacted during the last thirty-six years and in court interpretations of these laws. The goals of these laws are fairly clear. It is less clear how well these goals have been achieved. Of more importance for this morning\u27s session, serious questions can be raised concerning the extent to which these laws connect to the forms of discrimination that are emerging and that are becoming visible as the workforce and workplace change. These issues will be addressed this morning by our superb panel of speakers
A systems view of epithelial–mesenchymal transition signaling states
Epithelial–mesenchymal transition (EMT) is an important contributor to the invasion and metastasis of epithelial-derived cancers. While considerable effort has focused in the regulators involved in the transition process, we have focused on consequences of EMT to prosurvival signaling. Changes in distinct metastable and ‘epigentically-fixed’ EMT states were measured by correlation of protein, phosphoprotein, phosphopeptide and RNA transcript abundance. The assembly of 1167 modulated components into functional systems or machines simplified biological understanding and increased prediction confidence highlighting four functional groups: cell adhesion and migration, metabolism, transcription nodes and proliferation/survival networks. A coordinate metabolic reduction in a cluster of 17 free-radical stress pathway components was observed and correlated with reduced glycolytic and increased oxidative phosphorylation enzyme capacity, consistent with reduced cell cycling and reduced need for macromolecular biosynthesis in the mesenchymal state. An attenuation of EGFR autophosphorylation and a switch from autocrine to paracrine-competent EGFR signaling was implicated in the enablement of tumor cell chemotaxis. A similar attenuation of IGF1R, MET and RON signaling with EMT was observed. In contrast, EMT increased prosurvival autocrine IL11/IL6-JAK2-STAT signaling, autocrine fibronectin-integrin α5β1 activation, autocrine Axl/Tyro3/PDGFR/FGFR RTK signaling and autocrine TGFβR signaling. A relatively uniform loss of polarity and cell–cell junction linkages to actin cytoskeleton and intermediate filaments was measured at a systems level. A more heterogeneous gain of ECM remodeling and associated with invasion and migration was observed. Correlation to stem cell, EMT, invasion and metastasis datasets revealed the greatest similarity with normal and cancerous breast stem cell populations, CD49f(hi)/EpCAM(-/lo) and CD44(hi)/CD24(lo), respectively. Climacteric and non-climacteric fruits have traditionally been viewed as representing two distinct programmes of
ripening associated with differential respiration and ethylene hormone effects. In climacteric fruits, such as tomato
and banana, the ripening process is marked by increased respiration and is induced and co-ordinated by ethylene,
while in non-climacteric fruits, such as strawberry and grape, it is controlled by an ethylene-independent process
with little change in respiration rate. The two contrasting mechanisms, however, both lead to texture, colour, and
flavour changes that probably reflect some common programmes of regulatory control. It has been shown that
a SEPALLATA(SEP)4-like gene is necessary for normal ripening in tomato. It has been demonstrated here that
silencing a fruit-related SEP1/2-like (FaMADS9) gene in strawberry leads to the inhibition of normal development and
ripening in the petal, achene, and receptacle tissues. In addition, analysis of transcriptome profiles reveals pleiotropic
effects of FaMADS9 on fruit development and ripening-related gene expression. It is concluded that SEP
genes play a central role in the developmental regulation of ripening in both climacteric and non-climacteric fruits.
These findings provide important information to extend the molecular control of ripening in a non-climacteric fruit
beyond the limited genetic and cultural options currently available
Novel Chikungunya Vaccine Candidate with an IRES-Based Attenuation and Host Range Alteration Mechanism
Chikungunya virus (CHIKV) is a reemerging mosquito-borne pathogen that has recently caused devastating urban epidemics of severe and sometimes chronic arthralgia. As with most other mosquito-borne viral diseases, control relies on reducing mosquito populations and their contact with people, which has been ineffective in most locations. Therefore, vaccines remain the best strategy to prevent most vector-borne diseases. Ideally, vaccines for diseases of resource-limited countries should combine low cost and single dose efficacy, yet induce rapid and long-lived immunity with negligible risk of serious adverse reactions. To develop such a vaccine to protect against chikungunya fever, we employed a rational attenuation mechanism that also prevents the infection of mosquito vectors. The internal ribosome entry site (IRES) from encephalomyocarditis virus replaced the subgenomic promoter in a cDNA CHIKV clone, thus altering the levels and host-specific mechanism of structural protein gene expression. Testing in both normal outbred and interferon response-defective mice indicated that the new vaccine candidate is highly attenuated, immunogenic and efficacious after a single dose. Furthermore, it is incapable of replicating in mosquito cells or infecting mosquitoes in vivo. This IRES-based attenuation platform technology may be useful for the predictable attenuation of any alphavirus
