    LIF measurements of scalar mixing in turbulent shear layers

    The structure of shear layer flows at high Reynolds numbers remains a very interesting problem. Straight mixing layers have been studied and yielded information on the probability density function (pdf) of a passive scalar across the layer. Konrad and Koochesfahani & Dimotakis measured the pdf of the mixture fraction for mixing layers of moderate Reynolds numbers, each about 25,000 (Re based on velocity difference and visual thickness). Their measurements showed a 'non-marching' pdf (central hump which is invariant from edge to edge across the layer), a result which is linked to the visualizations of the spanwise Kelvin-Helmholtz (K-H) instability mode, which is the primary instability for plane shear layer flows. A secondary instability mode, the Taylor-Gortler (T-G) instability, which is associated with streamwise vortical structures, has also been observed in shear layers. Image reconstruction by Jimenez et al. and volume renderings by Karasso & Mungal at low Re numbers have demonstrated that the K-H and the T-G instability modes occur simultaneously in a non-mutually destructive way, evidence that supports the quasi two-dimensional aspect of these flows and the non-marching character of the pdf at low Reynolds numbers. At higher Re numbers though, the interaction of these two instability modes is still unclear and may affect the mixing process. In this study, we perform measurements of the concentration pdf of plane mixing layers for different operating conditions. At a speed ratio of r = U(sub 1)/U(sub 2) = 4:1, we examine three Reynolds number cases: Re = 14,000, Re = 31,000, and Re = 62,000. Some other Re number cases' results, not presented in detail, are invoked to explain the behavior of the pdf of the concentration field. A case of r = 2.6:1 at Re = 20,000 is also considered. The planar laser-induced fluorescence technique is used to yield quantitative measurements. The different Re are obtained by changing the velocity magnitudes of the two streams. The question of resolution of these measurements is addressed. In order to investigate the effects of the initial conditions on the development and the structure of the mixing layer, the boundary layer on the high-speed side of the splitter plate is tripped. The average concentration and the average mixed fluid concentration are also calculated to further understand the changes in the shear layer for the different cases examined

    Quality of internal audit reports

    The purpose of this paper is to explore the completeness and quality of audit reports as perceived by internal audit’s primary customer – the audit committee. Data was collected using a structured questionnaire that was sent to audit committee chairpersons of banks registered with the South African Reserve Bank. Respondents were asked to provide their perceptions of the quality of the internal audit reports they routinely received. The results highlight that not all internal audit functions present clear and appropriately focused reports. Whilst the audit committee chairpersons recognise that the internal audit reports do have value, there is also significant potential for improvement.http://reference.sabinet.co.za/sa_epublication/sajaaram201

    Viral trans-factor independent replication of human papillomavirus genomes

    <p>Abstract</p> <p>Background</p> <p>Papillomaviruses (PVs) establish a persistent infection in the proliferating basal cells of the epithelium. The viral genome is replicated and maintained as a low-copy nuclear plasmid in basal keratinocytes. Bovine and human papillomaviruses (BPV and HPV) are known to utilize two viral proteins; E1, a DNA helicase, and E2, a transcription factor, which have been considered essential for viral DNA replication. However, growing evidence suggests that E1 and E2 are not entirely essential for stable replication of HPV.</p> <p>Results</p> <p>Here we report that multiple HPV16 mutants, lacking either or both E1 and E2 open reading frame (ORFs) and the long control region (LCR), still support extrachromosomal replication. Our data clearly indicate that HPV16 has a mode of replication, independent of viral trans-factors, E1 and E2, which is achieved by origin activity located outside of the LCR.</p

    Plasmid-derived human papillomavirus type 11 (HPV11) DNA has been shown to replicate episomally and semiconservatively following transfection of primary human foreskin keratinocytes. HPV11 DNA was excised from the bacterial plasmid, religated to form circular molecules, and cotransfected along with pSV2Neo. Transfectants were selected and shown to contain replicated episomal HPV DNA. Once selected, HPV11 DNA persists in cells through at least two additional passages

    FEDSM2005-77101 Determination of Total Shear Stress and Polymer Stress Profiles in Drag Reduced Boundary Layer Flows with Polymer Injection

    Abstract Two methods of recovering the entire total shear stress profile from incomplete velocity data in turbulent boundary layers are presented and validated for both DNS simulations and experimental measurements. The first method, an exponential-polynomial curve fit, recovers the whole total shear stress profile well by using the data from the outer part of the boundary layer (y/δ &gt; 0.3). However, this curve fit is sensitive to the quality of the data. The second method, a new (1-y) weighted straight line fit, which is very simple and accurate, has been applied to current experiments of drag reduction in zero pressure gradient turbulent boundary layers with polymer injection. The total shear stress profile obtained from this fit is used to estimate the contribution of the polymer stress to the total shear stress
