Periodic organization of the contractile apparatus in smooth muscle revealed by the motion of dense bodies in single cells
To study the organization of the contractile apparatus in smooth muscle and its behavior during shortening, the movement of dense bodies in contracting saponin skinned, isolated cells was analyzed from digital images collected at fixed time intervals. These cells were optically lucent so that punctate structures, identified immunocytochemically as dense bodies, were visible in them with the phase contrast microscope. Methods were adapted and developed to track the bodies and to study their relative motion. Analysis of their tracks or trajectories indicated that the bodies did not move passively as cells shortened and that nearby bodies often had similar patterns of motion. Analysis of the relative motion of the bodies indicated that some bodies were structurally linked to one another or constrained so that the distance between them remained relatively constant during contraction. Such bodies tended to fall into laterally oriented, semirigid groups found at approximately 6-microns intervals along the cell axis. Other dense bodies moved rapidly toward one another axially during contraction. Such bodies were often members of separate semirigid groups. This suggests that the semirigid groups of dense bodies in smooth muscle cells may provide a framework for the attachment of the contractile structures to the cytoskeleton and the cell surface and indicates that smooth muscle may be more well-ordered than previously thought. The methods described here for the analysis of the motion of intracellular structures should be directly applicable to the study of motion in other cell types
Isomorphs in model molecular liquids
Isomorphs are curves in the phase diagram along which a number of static and
dynamic quantities are invariant in reduced units. A liquid has good isomorphs
if and only if it is strongly correlating, i.e., the equilibrium
virial/potential energy fluctuations are more than 90% correlated in the NVT
ensemble. This paper generalizes isomorphs to liquids composed of rigid
molecules and study the isomorphs of two systems of small rigid molecules, the
asymmetric dumbbell model and the Lewis-Wahnstrom OTP model. In particular, for
both systems we find that the isochoric heat capacity, the excess entropy, the
reduced molecular center-of-mass self part of the intermediate scattering
function, the reduced molecular center-of-mass radial distribution function to
a good approximation are invariant along an isomorph. In agreement with theory,
we also find that an instantaneous change of temperature and density from an
equilibrated state point to another isomorphic state point leads to no
relaxation. The isomorphs of the Lewis-Wahnstrom OTP model were found to be
more approximative than those of the asymmetric dumbbell model, which is
consistent with the OTP model being less strongly correlating. For both models
we find "master isomorphs", i.e., isomorphs have identical shape in the

Dynamics-based centrality for general directed networks
Determining the relative importance of nodes in directed networks is
important in, for example, ranking websites, publications, and sports teams,
and for understanding signal flows in systems biology. A prevailing centrality
measure in this respect is the PageRank. In this work, we focus on another
class of centrality derived from the Laplacian of the network. We extend the
Laplacian-based centrality, which has mainly been applied to strongly connected
networks, to the case of general directed networks such that we can
quantitatively compare arbitrary nodes. Toward this end, we adopt the idea used
in the PageRank to introduce global connectivity between all the pairs of nodes
with a certain strength. Numerical simulations are carried out on some
networks. We also offer interpretations of the Laplacian-based centrality for
general directed networks in terms of various dynamical and structural
properties of networks. Importantly, the Laplacian-based centrality defined as
the stationary density of the continuous-time random walk with random jumps is
shown to be equivalent to the absorption probability of the random walk with
sinks at each node but without random jumps. Similarly, the proposed centrality
represents the importance of nodes in dynamics on the original network supplied
with sinks but not with random jumps.Comment: 7 figure
Perception of nonnative tonal contrasts by Mandarin-English and English-Mandarin sequential bilinguals
This study examined the role of acquisition order and crosslinguistic similarity in influencing transfer at the initial stage of perceptually acquiring a tonal third language (L3). Perception of tones in Yoruba and Thai was tested in adult sequential bilinguals representing three different first (L1) and second language (L2) backgrounds: L1 Mandarin-L2 English (MEBs), L1 English-L2 Mandarin (EMBs), and L1 English-L2 intonational/non-tonal (EIBs). MEBs outperformed EMBs and EIBs in discriminating L3 tonal contrasts in both languages, while EMBs showed a small advantage over EIBs on Yoruba. All groups showed better overall discrimination in Thai than Yoruba, but group differences were more robust in Yoruba. MEBs’ and EMBs’ poor discrimination of certain L3 contrasts was further reflected in the L3 tones being perceived as similar to the same Mandarin tone; however, EIBs, with no knowledge of Mandarin, showed many of the same similarity judgments. These findings thus suggest that L1 tonal experience has a particularly facilitative effect in L3 tone perception, but there is also a facilitative effect of L2 tonal experience. Further, crosslinguistic perceptual similarity between L1/L2 and L3 tones, as well as acoustic similarity between different L3 tones, play a significant role at this early stage of L3 tone acquisition.Published versio
Cyclooxygenase in biology and disease
Cyclooxygenase (COX), the key enzyme required for the conversion of arachidonic acid to prostaglandins was first identified over 20 years ago. Drugs, like aspirin, that inhibit cyclooxygenase activity have been available to the public for about 100 years. In the past decade, however, more progress has been made in understanding the role of cyclooxygenase enzymes in biology and disease than at any other time in history. Two cyclooxygenase isoforms have been identified and are referred to as COX‐1 and COX‐2. Under many circumstances the COX‐1 enzyme is produced constitutively (i.e., gastric mucosa) whereas COX‐2 is inducible (i.e., sites of inflammation). Here, we summarize the current understanding of the role of cyclooxygenase‐1 and ‐2 in different physiological situations and disease processes ranging from inflammation to cancer. We have attempted to include all of the most relevant material in the field, but due to the rapid progress in this area of research we apologize that certain recent findings may have been left out.—DuBois, R. Ophthalmic Artery Chemosurgery for Less Advanced Intraocular Retinoblastoma: Five Year Review
BACKGROUND: Ophthalmic artery chemosurgery (OAC) for retinoblastoma was introduced by us 5 years ago for advanced intraocular retinoblastoma. Because the success was higher than with existing alternatives and systemic side effects limited we have now treated less advanced intraocular retinoblastoma (Reese-Ellsworth (RE) I-III and International Classification Retinoblastoma (ICRB) B and C). METHODOLOGY/PRINCIPAL FINDINGS: Retrospective review of 5 year experience in eyes with Reese Ellsworth (Table 1) I (7 eyes), II (6 eyes) or III (6 eyes) and/or International Classification (Table 2) B (19 eyes) and C (11 eyes) treated with OAC (melphalan with or without topotecan) introduced directly into the ophthalmic artery. Patient survival was 100%. Ocular event-free survival was 100% for Reese-Ellsworth Groups I, II and III (and 96% for ICRB B and C) at a median of 16 months follow-up. One ICRB Group C (Reese-Ellsworth Vb) eye could not be treated on the second attempt for technical reasons and was therefore enucleated. No patient required a port and only one patient required transfusion of blood products. The electroretinogram (ERG) was unchanged or improved in 14/19 eyes. CONCLUSIONS/SIGNIFICANCE: Ophthalmic artery chemosurgery for retinoblastoma that was Reese-Ellsworth I, II and III (or International Classification B or C) was associated with high success (100% of treatable eyes were retained) and limited toxicity with results that equal or exceed conventional therapy with less toxicity
Evolution of Cooperation and Coordination in a Dynamically Networked Society
Situations of conflict giving rise to social dilemmas are widespread in
society and game theory is one major way in which they can be investigated.
Starting from the observation that individuals in society interact through
networks of acquaintances, we model the co-evolution of the agents' strategies
and of the social network itself using two prototypical games, the Prisoner's
Dilemma and the Stag Hunt. Allowing agents to dismiss ties and establish new
ones, we find that cooperation and coordination can be achieved through the
self-organization of the social network, a result that is non-trivial,
especially in the Prisoner's Dilemma case. The evolution and stability of
cooperation implies the condensation of agents exploiting particular game
strategies into strong and stable clusters which are more densely connected,
even in the more difficult case of the Prisoner's Dilemma.Comment: 18 pages, 14 figures. to appea
Auditory Enhancement and Second Language Experience in Spanish and English Weighting of Secondary Voicing Cues
The role of secondary cues in voicing categorization was investigated in three listener groups: Monolingual English (n=20) and Spanish speakers (n=20), and Spanish speakers with significant English experience (n=16). Results showed that, in all three groups, participants used onset f0 in making voicing decisions only in the positive voice onset time (VOT) range (short lag and long lag tokens), while there was no effect of onset f0 on voicing categorization within the negative VOT range (voicing lead tokens) for any of the participant groups. These results support an auditory enhancement view of perceptual cue weighting: Onset f0 serves as a secondary cue to voicing only in the positive VOT range where it is not overshadowed by the presence of pre-voicing. Moreover, results showed that Spanish learners of English gave a significantly greater weight to onset f0 in their voicing decisions than did listeners in either of the other two groups. This result supports the view that learners may overweight secondary cues to distinguish between non-native categories that are assimilated to the same native category on the basis of a primary cue
SGAS 143845.1+145407: A Big, Cool Starburst at Redshift 0.816
We present the discovery and a detailed multi-wavelength study of a
strongly-lensed luminous infrared galaxy at z=0.816. Unlike most known lensed
galaxies discovered at optical or near-infrared wavelengths this lensed source
is red, r-Ks = 3.9 [AB], which the data presented here demonstrate is due to
ongoing dusty star formation. The overall lensing magnification (a factor of
17) facilitates observations from the blue optical through to 500micron, fully
capturing both the stellar photospheric emission as well as the re-processed
thermal dust emission. We also present optical and near-IR spectroscopy. These
extensive data show that this lensed galaxy is in many ways typical of
IR-detected sources at z~1, with both a total luminosity and size in accordance
with other (albeit much less detailed) measurements in samples of galaxies
observed in deep fields with the Spitzer telescope. Its far-infrared spectral
energy distribution is well-fit by local templates that are an order of
magnitude less luminous than the lensed galaxy; local templates of comparable
luminosity are too hot to fit. Its size (D~7kpc) is much larger than local
luminous infrared galaxies, but in line with sizes observed for such galaxies
at z~1. The star formation appears uniform across this spatial scale. In this
source, the luminosity of which is typical of sources that dominate the cosmic
infrared background, we find that star formation is spatially extended and well
organised, quite unlike the compact merger-driven starbursts which are typical
for sources of this luminosity at z~0.Comment: 18 pages, 10 figure
Patient and physician discordance of global disease assessment in juvenile dermatomyositis: Findings from the Childhood Arthritis & Rheumatology Research Alliance Legacy Registry
© 2020 The Author(s). Background: Global disease activity scores (gVAS) capture patient or family (PF) and physician (MD) assessments of disease. This study sought to measure discordance between PF and MD global activity scores in juvenile dermatomyositis (JDM), and determine factors associated with discordance. Methods: Patients with JDM were included from the Childhood Arthritis and Rheumatology Research Alliance (CARRA) Legacy Registry (N = 563). PF and MD gVAS were assessed for discordance, defined as a ≥ 2-point difference. Factors associated with discordant gVAS were compared in univariate analysis. Multivariable regression analysis was used to identify predictors of discordance. Results: Almost 40% (N = 219) of PF and MD gVAS were discordant. Among discordant scores, 68% of PF rated gVAS ≥2-points above MD, which was associated with calcinosis and lower quality of life and functional scores (p \u3c 0.01). MD gVAS rated ≥2-points above PF in 32%, which was associated with abnormal laboratory results, weakness, arthritis, rash and other skin changes, and current intravenous steroid treatment (p \u3c 0.01). In multivariate analysis, predictors for higher PF rating included calcinosis, lower quality of life and functional scores, while predictors for higher MD rating included rash, calcinosis, nailfold capillaroscopy changes, and current intravenous steroid treatment. Conclusions: Discordance between PF and MD gVAS was common in this JDM cohort. Overall, higher PF rating was associated with poorer patient reported outcome (PRO) scores, while higher MD rating was associated with poorer objective measures. This suggests PF and MD assessments of gVAS may be measuring different aspects of disease, highlighting the importance of integrating PROs into clinical practice and research
