    Studies on Combining Ability for Yield and Quality Traits in Brinjal (Solanum melongena L.)

    The experimental material in brinjal (eggplant) comprised 19 parents (15 lines + 4 testers), 60 F1 crosses and two standard checks (BH-1 and BH-2). This was grown in Randomized Block Design, with three replications. Lines vs. testers showed significance for all characters except plant height, plant spread, number of primary branches, dry matter, total sugars and total phenol. Analysis for parents vs. hybrids showed significance for all the characters except average fruit weight, number of fruits per plant, plant spread and number of primary branches. Analysis of Variance for combining ability revealed mean squares due to lines and testers were significant for all the characters except plant height, plant spread, number of primary branches, total sugars, total phenol and content of anthocyanins. The ratio of variance due to specific combining ability and general combining ability (σ2SCA: σ2GCA) was greater than unity, indicating non-additive genetic control for all traits except plant spread and total phenols. Among the females, Punjab Barsati, PBR-91-1, RCMBL-1-1, BSR-11; and among the males, BB-93-C and U-8-61-3 were best general combiners for yield and yield components. Punjab Barsati was the best combiner for days to 50% flowering, days to first fruit harvest, number of fruits per plant and number of primary branches. The cross JBR-3-16 × PB-64 manifested best SCA effects for days to 50% flowering; PBR-91-1JBSR-98-2 for average fruit weight; BSR-11 × PB-64 for fruit length; BSR-11× U-8-61-3 for fruit girth, and the cross HABL-1 × JBSR-98-2 for yield per plant and per hectare

    Cassia auriculata: Aspects of Safety Pharmacology and Drug Interaction

    Safety pharmacology studies help in identifying preclinical adverse drug reactions. We carried out routine safety pharmacology with focus on cardiovascular variables and pharmacokinetic herb-drug interaction studies on rats fed with standardized traditional hydro-alcoholic extract and technology-based supercritical extract of Cassia auriculata for 12 weeks. Our studies indicate that both these extracts are pharmacologically safe and did not show any significant adverse reactions at the tested doses. The traditional hydro-alcoholic extract did not show any significant effect on pharmacokinetics; however, the technology-based supercritical extract caused a significant reduction in absorption of metformin. Our results indicate the need to include pharmacokinetic herb-drug interaction studies as evidence for safety especially for technology-based extracts

    Techno-economic viability of bio-based methyl ethyl ketone production from sugarcane using integrated fermentative and chemo-catalytic approach: process integration using pinch technology

    Butanediols are versatile platform chemicals that can be transformed into a spectrum of valuable products. This study examines the techno-commercial feasibility of an integrated biorefinery for fermentative production of 2,3-butanediol (BDO) from sucrose of sugarcane (SC), followed by chemo-catalytic upgrading of BDO to a carbon-conservative derivative, methyl ethyl ketone (MEK), with established commercial demand. The techno-economics of three process configurations are compared for downstream MEK separation from water and co-product, isobutyraldehyde (IBA): (I) heterogeneous azeotropic distillation of MEK-water and extractive separation of (II) MEK and (III) MEK-IBA from water using p-xylene as a solvent. The thermal efficiency of these manufacturing processes is further improved using pinch technology. The implementation of pinch technology reduces 8% of BDO and 9–10% of MEK production costs. Despite these improvements, raw material and utility costs remain substantial. The capital expenditure is notably higher for MEK production from SC than BDO alone due to additional processing steps. The extraction based MEK separation is the simplest process configuration despite marginally higher capital requirements and utility consumption with slightly higher production costs than MEK-water azeotropic distillation. Economic analysis suggests that bio-based BDO is cost-competitive with its petrochemical counterpart, with a minimum gross unitary selling price of US$ 1.54, assuming a 15% internal rate of return over five-year payback periods. However, renewable MEK is approximately 16–24% costlier than the petrochemical route. Future strategies must focus on reducing feedstock costs, improving BDO fermentation efficacy, and developing a low-cost downstream separation process to make renewable MEK commercially viable

    Assessing long term impact of nutrient management and rainfall variability on the agroecological resilience of maize (Zea mays)- wheat (Triticum aestivum) system in NW India

    A long-term (2000-2010) field experiment was carried out in the lower Shiwalik foothills of Punjab to study the carry over effect of organic manures and fertilizers on the productivity of maize (Zea mays L.)- wheat (Triticum aestivum L.) cropping system for efficient N management and resource use under rainfed conditions and to develop predictive models describing relationship between yields and seasonal rainfall. N management strategies involving combined application of 15 kg N/ha either through compost or leucaena loppings along with 20 kg N/ha through inorganic fertilizer for maize-wheat cropping sequence utilized growth resources most efficiently and maintained stable yield performance culminating in significantly higher system productivity, better resource use efficiencies and sustainable yield index, suggesting partial N substitution through compost or locally available plant material. The regression models developed to predict the effects of N sources on crop yields using monthly rainfall would be of interest to estimate the yield at a given level of rainfall with the likely fluctuation (as error) particularly under rainfed conditions

    Effect of Mucin13 gene polymorphism on diarrhoeagenic <i>E. coli</i> adhesion pattern and its expression analysis in native Indian pigs

    We identified genetic polymorphism in Mucin13 gene affecting E. coli adhesion patterns using (local isolate) diarrhoeagenic E. coli in Indian desi pigs. Five SNPs and one indel previously reported to be associated with enterotoxigenic E. coli (ETEC) F4ab/ac adhesion pattern were examined by designing PCR-RFLP protocol. The genotypic frequencies of only one SNP (g.22304A&thinsp; &gt; &thinsp;G) differed significantly (at P ≤ 0.05) in adhesive, non-adhesive and weakly adhesive population. The AA (306&thinsp;sbp, 231&thinsp;bp), AG (306, 231, 108, 198&thinsp;bp) and GG (231, 198&thinsp;bp, 108&thinsp;b) genotypes of g.22304A&thinsp; &gt; &thinsp;G locus were observed with frequencies 50.0&thinsp;%, 21.25&thinsp;% and 28.75&thinsp;%, respectively and AG genotype was significantly (P ≤ 0.05) associated with a non-adhesive pattern. The polymorphism information content of SNPs ranged from 17.67 (g.22124T&thinsp; &gt; &thinsp;C) to 37.36&thinsp;% (g.21471C&thinsp; &gt; &thinsp;T) loci. Three loci (g.21471C&thinsp; &gt; &thinsp;T, g.22124T&thinsp; &gt; &thinsp;C and g.22304A&thinsp; &gt; &thinsp;G) were significantly departed from Hardy–Weinberg equilibrium. The linkage disequilibrium analysis revealed locus g.22124T&thinsp; &gt; &thinsp;C and g.22304A&thinsp; &gt; &thinsp;G were significantly (P ≤ 0.05) associated with each other. Expression profiling of target gene in jejuna of animals having AA, AG and GG genotypes revealed differences in various genotypes with the highest in the AA, moderate in the GG and low levels in the AG genotype, although they were statistically non-significant (at P ≤ 0.05). The absence of significant effect of genotypes on MUC13 mRNA expression indicates no direct functional role, although the structural role can not be ignored as the putative receptor gene is located within targeted genomic region. Further, reports of same SNP association with an ETEC F4ab/ac adhesion pattern indicate the target gene's role in diarrhoea even caused by other strains of E. coli which is not ETEC.</p

    Molekularni dokaz toksinskih tipova bakterije Clostridium perfringens, enteropatogenih sojeva Escherichia coli te rotavirusa i koronavirusa u uzorcima proljeva kod neonatalne jaradi

    In the present study, out of 1156 neonatal goat kids, 238 showing clinical diarrhea were used for detection of toxinotypes of Clostridium perfringens, Enteropathogenic E. coli (EPEC), Group A rotavirus (GARV) and Bovine coronavirus (BCV). Isolation and toxinotyping of isolates were done by multiplex Polymerase chain reaction (PCR) using primers for cpa, cpb, cpb2, etx and iap genes. For EPEC, isolation and identification were done using bfpA gene and SYBR green based real time PCR (qPCR). GARV and BCV were detected, by one-step RT-PCR (osRT-PCR). The incidence of C. perfringens was 15.13% with 75% isolates toxinotype A, 25% type D and 61.11% of isolates carrying the β2-toxin gene. The incidence of EPEC was 68.07% based on qPCR, whereas 21.85% were positive for GARV and 15.97% for BCV by osRT-PCR. There was mixed infection of C. perfringens and EPEC in 11.76% and 3.78% for C. perfringens and GARV and 2.1% of C. perfringens and BCV. EPEC and GARV was 19.74% and EPEC plus BCV positivity was 11.34%. GARV and BCV was 5.88%, and 4.20% had mixed infection of EPEC, GARV and BCV. Of the total diarrheic kids sampled, 0.84% had mixed infection of C. perfringens, GARV, BCV and EPEC. On the basis of the above findings, it may be concluded that isolation, multiplex PCR and real time PCR facilitated the characterization of circulating C. perfringens toxinotypes and EPEC in goats reared under semi-arid conditions. The importance of enteritis caused by GARV and BCV and their role in mixed infection in goats requires extensive screening and pathogenicity studies to associate the symptoms with disease.U populaciji od 1156 neonatalnih jarića 238 je pokazivalo kliničke znakove proljeva. Od njih su uzeti uzorci izmeta za dokazivanje toksinskih tipova bakterije Clostridium perfringens, enteropatogenih sojeva bakterije E. coli (engl. enteropathogenic E. coli, EPEC), rotavirusa skupine A (engl. group A rotavirus, GARV) i goveđeg koronavirusa (engl. bovine coronavirus, BCV). Izdvajanje i toksinska tipizacija izolata provedeni su višestrukom lančanom reakcijom polimerazom (PCR) upotrebom početnica za gene cpa, cpb, cpb2, etx i iap. Izdvajanje i identifikacija EPEC-a provedeni su pretragom na gen bfpA i PCR-om u stvarnom vremenu, uz upotrebu SYBR zelenila (qPCR). Za dokaz rotavirusa skupine A i goveđeg koronavirusa upotrijebljena je RT-PCR (osRT-PCR). Incidencija bakterije C. perfringens iznosila je 15,13 %. Od toga je 75 % izolata pripadalo toksinskom tipu A, 25 % tipu D, dok je 61,11 % izolata imalo gen za toksin β2. Incidencija EPEC-a iznosila je 68,07 %, a 21,85 % pretraženih uzoraka bilo je pozitivno na GARV te 15,97 % na BCV. Mješovita infekcija bakterijom C. perfringens i EPEC-om utvrđena je u 11,76 % uzoraka, C. perfringens i GARV u 3,78 % te C. perfringens i BCV u 2,1 % pretraženih uzoraka. Mješovita infekcija EPEC-om i GARV-om utvrđena je u 19,74 %, a EPEC-om i BCV-om u 11,34 %. Mješovita infekcija rotavirusom i koronavirusom bila je ustanovljena u 5,88 %, a mješovita infekcija EPEC-om, GARV-om i BCV-om u 4,20 % uzoraka. Od ukupnog broja pretražene jaradi s proljevom u njih 0,84 % dokazana je mješovita infekcija bakterijom C. perfringens, rotavirusom skupine A, goveđim koronavirusom i EPEC-om. Na osnovi prikazanih rezultata može se zaključiti da izdvajanje, višestruki PCR te PCR u stvarnom vremenu omogućuju karakterizaciju i praćenje kolanja toksinskih tipova bakterija C. perfringens i EPEC-a u koza uzgajanih u sušnim uvjetima. Važnost enteritisa uzrokovanog rotavirusom skupine A i goveđim koronavirusom te njihova uloga kod mješovitih infekcija u koza zahtijevaju pojačan nadzor i istraživanje patogenosti radi povezivanja kliničkih znakova s ustanovljenom bolešću
