    A Plane Wave Virtual Element Method for the Helmholtz Problem

    We introduce and analyze a virtual element method (VEM) for the Helmholtz problem with approximating spaces made of products of low order VEM functions and plane waves. We restrict ourselves to the 2D Helmholtz equation with impedance boundary conditions on the whole domain boundary. The main ingredients of the plane wave VEM scheme are: i) a low frequency space made of VEM functions, whose basis functions are not explicitly computed in the element interiors; ii) a proper local projection operator onto the high-frequency space, made of plane waves; iii) an approximate stabilization term. A convergence result for the h-version of the method is proved, and numerical results testing its performance on general polygonal meshes are presented

    How toxic are gold nanoparticles? The state-of-the-art.

    With the growing interest in biotechnological applications of gold nanoparticles and their effects exerted on the body, the possible toxicity is becoming an increasingly important issue. Numerous investigations carried out, in the last few years, under different experimental conditions, following different protocols, have produced in part conflicting results which have leaded to different views about the effective gold nanoparticle safety in human applications. This work is intended to provide an overview on the most recent experimental results in order to summarize the current state-of-the-art. However, rather than to present a comprehensive review of the available literature in this field, that, among other things, is really huge, we have selected some representative examples of both in vivo and in vitro investigations, with the aim of offering a scenario from which clearly emerges the need of an urgent and impelling standardization of the experimental protocols. To date, despite the great potential, the safety of gold nanoparticles is highly controversial and important concerns have been raised with the need to be properly addressed. Factors such as shape, size, surface charge, surface coating and surface functionalization are expected to influence interactions with biological systems at different extents, with different outcomes, as far as gold nanoparticle potentiality in biomedical applications is concerned. Moreover, despite the continuous attempt to establish a correlation between structure and interactions with biological systems, we are still far from assessing the toxicological profile of gold nanoparticles in an unquestionable manner. This review is intended to provide a contribution in this direction, offering some suggestions in order to reach the systematization of data over the most relevant physico-chemical parameters, which govern and control toxicity, at different cellular and organismal levels

    Chemiresistive polyaniline-based gas sensors: a mini review

    This review focuses on some recent advances made in the field of gas sensors based on polyaniline [PANI], a conducting polymer with excellent electronic conductivity and electrochemical properties. Conducting polymers represent an important class of organic materials with an enhanced resistivity towards external stimuli. Among them, PANI polymers have attracted wide interest because of the versatility in their use, combined with the easy of synthesis, high yield and good environmental stability, together with a favorable response to guest molecules at room temperature. Moreover, PANI can be shaped into various structures with different morphologies and the possibility of obtaining nanofibers, in addition to thin films, has opened a rapid development of ultrasensitive chemical sensors, with improved processability and functionality. This review provides a brief description of the current status of gas chemiresistive sensors based on polyaniline and highlights the properties and applications of these devices in diverse range of applications. © 2015 Elsevier B.V.     New Perspectives on Treatment of Hepatitis B Before and After Liver Transplantation

    open5noThe hepatitis B virus (HBV) infects more than 260 million people globally, with increasing incidence, especially in developing countries. Despite antiviral therapies, HBV-related end-stage liver disease remains one of the most important indications for liver transplantation worldwide. Although new available treatments have improved the outcome of patients with both compensated and decompensated liver disease in some specific clinical settings as acute-on-chronic liver failure mortality is still high. Moreover, the incidence of HBV-related hepatocellular carcinoma (HCC) seems to be increasing and represents a major challenge for the transplant team. In the post-transplant setting, combination of anti-HBV immunoglobulins and oral nucleos(t)ides provided significant improvement on graft and patient survival. Furthermore, recent data suggested the possibility of personalized therapeutic algorithms based on pre and post-transplant viral and host risk factors. Finally, liver grafts from HBV core antibody (anti-HBc) positive or hepatitis B surface antigen (HBsAg) donors can be safely used in order to expand the donor pool, considering adequate allocation and tailored prophylaxis after LT.     Malleability of the self: electrophysiological correlates of the enfacement illusion

    Self-face representation is fundamentally important for self-identity and self-consciousness. Given its role in preserving identity over time, self-face processing is considered as a robust and stable process. Yet, recent studies indicate that simple psychophysics manipulations may change how we process our own face. Specifically, experiencing tactile facial stimulation while seeing similar synchronous stimuli delivered to the face of another individual seen as in a mirror, induces 'enfacement' illusion, i.e. the subjective experience of ownership of the other’s face and a bias in attributing to the self, facial features of the other person. Here we recorded visual Event-Related Potentials elicited by the presentation of self, other and morphed faces during a self-other discrimination task performed immediately after participants received synchronous and control asynchronous Interpersonal Multisensory Stimulation (IMS). We found that self-face presentation after synchronous as compared to asynchronous stimulation significantly reduced the late positive potential (LPP; 450-750 ms), a reliable electrophysiological marker of self-identification processes. Additionally, enfacement cancelled out the differences in LPP amplitudes produced by self- and other-face during the control condition. These findings represent the first direct neurophysiological evidence that enfacement may affect self-face processing and pave the way to novel paradigms for exploring defective self-representation and self-other interactions

    Liver transplantation for viral hepatitis in 2015

    Liver transplantation (LT) is a life-saving treatment for patients with end-stage liver disease and for patients with liver cell cancer related to liver disease. Acute and chronic liver diseases related to hepatitis viruses are between the main indications for liver transplantation. The risk of viral reinfection after transplantation is the main limiting factor in these indications. Before the availability of antiviral prophylaxis, hepatitis B virus (HBV) recurrence was universal in patients who were HBV DNA-positive before transplantation. The natural history of recurrent HBV was accelerated by immunosuppression, and it progressed rapidly to graft failure and death. Introduction of post-transplant prophylaxis with immunoglobulin alone first, and associated to antiviral drugs later, drastically reduced HBV recurrence, resulting in excellent long-term outcomes. On the contrary, recurrence of hepatitis C is the main cause of graft loss in most transplant programs. Overall, patient and graft survival after LT for hepatitis C virus (HCV)-associated cirrhosis is inferior compared with other indications. However, successful pretransplant or post transplant antiviral therapy has been associated with increased graft and overall survival. Until recently, the combination of pegylated interferon and ribavirin was the standard of care for the treatment of patients with chronic hepatitis C.     Hypoxia and Inflammation are strictly interconnected with important consequences at clinical and therapeutic level. While cell and tissue damage due to acute hypoxia mostly leads to cell necrosis, in chronic hypoxia, cells that are located closer to vessels are able to survive adapting their phenotype through the expression of a number of genes, including proinflammatory receptors for alarmins. These receptors are activated by alarmins released by necrotic cells and generate signals for master transcription factors such as NFkB, AP1, etc. which control hundreds of genes for innate immunity and damage repair. Clinical consequences of chronic inflammatory reparative response activation include cell and tissue remodeling, damage in the primary site and, the systemic involvement of distant organs and tissues. Thus every time a tissue environment becomes stably hypoxic, inflammation can be activated followed by chronic damage and cell death or repair with vessel proliferation and fibrosis. This pathway can occur in cancer, myocardial infarction and stroke, diabetes, obesity, neurodegenerative diseases, chronic and autoimmune diseases and age-related diseases. Interestingly, proinflammatory gene expression can be observed earlier in hypoxic tissue cells and, in addition, in activated resident or recruited leukocytes. Herewith, the reciprocal relationships between hypoxia and inflammation will be shortly reviewed to underline the possible therapeutic targets to control hypoxia-related inflammation in a number of epidemiologically important human diseases and conditions

    Platelet-activating factor mediates CD40-dependent angiogenesis and endothelial-smooth muscle cell interaction.

    The aim of the present study was to investigate whether stimulation of CD40 expressed by endothelial or smooth muscle cells triggers the synthesis of platelet-activating factor (PAF), an inflammatory mediator with angiogenic properties, and whether PAF contributes to CD40-induced neoangiogenesis. The results obtained indicate that the interaction of CD40 with soluble CD154 or with CD154 expressed on the membrane of leukocytes (CD154-transfected J558 cells) or of activated platelets, stimulated the synthesis of PAF by endothelial cells but not by smooth cells. The synthesis of PAF triggered by activated platelets was inhibited by a soluble CD40-murine Ig fusion protein that prevents the interaction between membrane CD40 and CD154. Studies with specific inhibitors and evaluation of protein phosphorylation indicated the involvement in PAF synthesis of two intracellular signaling pathways leading to cytosolic phospholipase A 2 activation: a phospholipase Cγ-protein kinase C-Raf-p42/p44-mitogen-activated protein kinase (MAPK) and a MAPK kinase-3/6-dependent activation of p38 MAPK. PAF synthesized by endothelial cells after CD40 stimulation was instrumental in the in vitro migration and vessel-like organization of endothelial cells, and in the interaction between endothelial cells and smooth muscle cells, as inferred by the inhibitory effect of two different PAF receptor antagonists, WEB2170 and CV3988. In vivo, blockade of PAF receptors prevented the angiogenic effect triggered by CD40 stimulation in a murine model of s.c. Matrigel implantation. In conclusion, these observations indicate that PAF synthesis induced by stimulation of endothelial CD40 contributes to the formation and organization of new vessels. This may be relevant in the vascular remodeling associated with tumor and inflammatory neoangiogenesis

    Frequent detection of high human papillomavirus DNA loads in oral potentially malignant disorders

    Human papillomavirus (HPV) is estimated to be the cause of 40-80% of the squamous cell carcinoma of the oropharynx but only of a small fraction of the oral cavity cancers. The prevalence of oral HPV infection has significantly increased in the last decade, raising concerns about the HPV role in progression of oral potentially malignant disorders (OPMD) toward squamous cell carcinomas. We sought to study HPV infection in patients with oral lesions, and in control individuals, using non-invasive and site-specific oral brushing and sensitive molecular methods. HPV DNA positivity and viral loads were evaluated in relation to patient data and clinical diagnosis. We enrolled 116 individuals attending Dental Clinics: 62 patients with benign oral lesions (e.g. fibromas, papillomatosis, ulcers) or OPMD (e.g. lichen, leukoplakia) and 54 controls. Oral cells were collected with Cytobrush and HPV-DNA detected with quantitative real-time PCR (qPCR) for the more common high-risk (HR) and low-risk (LR) genotypes. HPV detection rate, percentage of HR HPVs and HPV-DNA loads (namely HPV16 and in particular, HPV18) were significantly higher in patients than in controls. Lichen planus cases had the highest HPV positive rate (75.0%), hairy leukoplakia the lowest (33.3%). This study detected unexpectedly high rates of HPV infection in cells of the oral mucosa. The elevated HR HPV loads found in OPMD suggest the effectiveness of qPCR in testing oral lesions. Prospective studies are needed to establish whether elevated viral loads represent a clinically useful marker of the risk of malignant progression
