    Fluorescent optical fiber sensors for cell viability monitoring

In contrast to common methods for cell viability control, which are usually either destructive (like flow-type counters or dead cells coloring and counting), or hardly quantitative like fluorescent microscopy, the method described is automated, does not require the removal of cells from their growth area and is sensitive enough to deal with as low as tens of cells

    Hidden in plain sight: bias towards sick patients when sampling patients with sufficient electronic health record data for research

    Background: To demonstrate that subject selection based on sufficient laboratory results and medication orders in electronic health records can be biased towards sick patients. Methods: Using electronic health record data from 10,000 patients who received anesthetic services at a major metropolitan tertiary care academic medical center, an affiliated hospital for women and children, and an affiliated urban primary care hospital, the correlation between patient health status and counts of days with laboratory results or medication orders, as indicated by the American Society of Anesthesiologists Physical Status Classification (ASA Class), was assessed with a Negative Binomial Regression model. Results: Higher ASA Class was associated with more points of data: compared to ASA Class 1 patients, ASA Class 4 patients had 5.05 times the number of days with laboratory results and 6.85 times the number of days with medication orders, controlling for age, sex, emergency status, admission type, primary diagnosis, and procedure. Conclusions: Imposing data sufficiency requirements for subject selection allows researchers to minimize missing data when reusing electronic health records for research, but introduces a bias towards the selection of sicker patients. We demonstrated the relationship between patient health and quantity of data, which may result in a systematic bias towards the selection of sicker patients for research studies and limit the external validity of research conducted using electronic health record data. Additionally, we discovered other variables (i.e., admission status, age, emergency classification, procedure, and diagnosis) that independently affect data sufficiency

    Dacha dwellers and gardeners: garden plots and second homes in Europe and Russia

    Abstract One of the ways to solve the problems associated with rapid growth of urban population and the development of industry in Western Europe in the 19th century was the creation of collective gardens and vegetable plots, which could be used to grow food for personal consumption. The peak of their popularity was during the First and Second World Wars. In the second half of the 20th century, as food shortages decrease, the number of garden plots in Western Europe sharply decreased. The revival of interest in gardening at the end of the 20 century is connected with the development of nature protection movement and ecological culture. In Eastern Europe, most of the collective gardens and vegetable plots appeared after the Second World War in a planned economy, they were most popular during the periods of economic recession. In some countries – Russia, Poland – gardeners have now become one of the largest land users. The article deals with the history and main factors that influenced the development of collective gardening and vegetable gardening in Europe and analyzes the laws presently regulating the activity of gardeners. The change of functions of garden plots in European countries in the 19–21 centuries is shown. The article presents comparative statistics on the number of second homes in Northern and Southern Europe. On the example of these two regions, the peculiarities of distribution of second homes are revealed and the main areas of their location are shown

    Institutional and regional features of organized second home development in Russia

    The article studies the features of second home land development in Russia. Basing on the data of the All-Russian agricultural censuses, the author shows that organized second home development is represented by two quantitatively and qualitatively different processes, namely: development of residential dachas, observed in horticultural non-profit partnerships (HNP) with dominant residential, recreational and agricultural functions, and development of agricultural dachas (vegetable gardens), observed in gardening non-profit partnerships (GNP), performing only an agricultural function. Despite regional differentiation, the general developmental trend might be described as gradual strengthening of the recreational and residential functions of the HNPs, which is facilitated by institutional regulation. The absolute and relative scales of the GNPs extension are gradually decreasing, but due to the unique localization and long-term traditions of subsidiary gardening, they remain relevant, especially during periods of crisis. Therefore, in some regions, their share in the total land of garden and dacha formations substantially exceeds the national average. The desire of the owners to supplement the functions of the GNPs with a permit for the construction of houses leads to the transfer of gardening lands to other categories, but in the absence of such an opportunity, Russian GNPs remain an analogue of foreign «allotment gardens», where one cannot spend the night, but can do gardening

    Development of dacha settlements in the Moscow region in the second half of the 19th – first half of the 20th century

    One of the results of the dacha development of a territory is the emergence of specific types of settlements – dacha settlements. Their role in today’s national economic complex of the Moscow region is different, largely due to peculiarities of formation at different stages of socio-economic development. In this context, the pre-war decades are interesting as a period of adaptation of the pre-revolutionary dacha market trends to meet needs of the new planned economy, however, the analysis is complicated by inconsistency of a lot of statistical data. Data supplementary with reference and literary sources makes it possible to show that the pre–war dacha settlements were formed in the following two ways originated after reforming in1861: «the urban type» – through the processes of urbanization occurring in rural settlements close to the city limits, and «the developer type» – through the construction of dacha settlements on specially allocated lands. This led to the formation of a network of dacha settlements in the pre-war Moscow region, which became local centers of economic development, capable of meeting the macroeconomic needs in a mobile way, and strengthening prerequisites for the post-war development of the dacha settlements

    Impact of p53 Knockout on Protein Data Set of HaCaT Cells in Confluent and Subconfluent Conditions

    The immortalized keratinocytes, HaCaT, are a popular model for skin research (toxicity, irritation, allergic reactions, or interaction of cells). They maintain a stable keratinocyte phenotype and respond to keratinocyte differentiation stimuli. However, programs of stratification and expression of differentiation markers in HaCaT keratinocytes are aberrant. In HaCaT cells, there are two mutant p53 alleles (i.e., R282Q and H179Y) that contain gain-of-function (GOF) mutations resulting from spontaneous immortalization (mutp53). At the same time, mutp53 acts as a transcription factor and also affects the interaction of p63 protein with its transcription targets. Proteins of the p53 family are crucial for regulation of proliferation and differentiation processes in human keratinocytes, although the involvement of mutp53 in these processes is not fully clear. We present data sets obtained as a result of high-performance proteomic analysis of immortalized HaCaT keratinocytes with p53 knockout in two different states, subconfluent and confluent, which are characterized by different intensites of cell differentiation processes. To obtain the proteomic profiles of the cells, we applied LC-MS/MS measurements processed with MaxQuant software (version 1.6.3.4)

    Self-isolation at the dacha: Can’t? Can? Have to?

    Measures taken by most countries to limit the coronavirus infection spread include self-isolation. An option of voluntary restriction of personal contacts for citizens is to move to the country (second or third) houses, which have a particular name in Russia – “dacha”. The demand for country estates as places of self-isolation can be assessed as the emergence of a new sanitary-epidemic function in second homes. Institutional management of such movements in connection with the coronavirus pandemic varies by country, ranging from prohibition (Norway) to encouragement (Belarus), and quantitative indicators (mass character or singleness) fluctuate according to lifestyle, national traditions, characteristics of settlement, urban housing policy, public health opportunities and many other factors. For Russians, the migration of residents of megalopolises from the city to country houses was a reaction to the pandemic, a characteristic social-group strategy of health-preserving behaviour. Several million Muscovites, Petersburgers, as well as residents of other megacities of Russia moved outside the cities immediately after the outbreak of the pandemic. “Half-townspeople” – internal migrant workers and “seasonal workers” (workers living in villages or small towns but working in metropolises in watch mode) also moved to rural areas. The mass nature of centrifugal spatial-migratory deurbanization model of behaviour of Russians during the pandemic is determined by the specifics of the spatial distribution of the population in Russia, historical features of urbanization and deurbanization processes, in particular, the widespread distribution of second (and third) country houses (dachas) among the citizens. Russia leads both in relative and absolute number of dacha dwellers among the European countries. The number of country houses in Russia is estimated by specialists at 17–20 million, and the number of dacha dwellers at 50–60 million; at least half of the citizens have second (and often third) country houses. Massive movements of citizens into out-of-town spaces had both positive and negative consequences. A significant share of citizens reduced risks of infection and were able to avoid “imprisonment” within the apartment with accompanying socio-psychological overload and physical inactivity. However, mass movements also contributed to the rapid spread of coronavirus beyond the original foci. The article considers the approaches of European countries to countryside self-isolation, describes chronicles of restriction on movement of citizens in Russian regions and waves of summer migration during the pandemic, suggests an assessment of dacha migration from the capital, and discusses its short-term socio-economic consequences
