    Safety and feasibility of robotic-assisted thoracic surgery after neoadjuvant chemoimmunotherapy in non-small cell lung cancer

    ObjectivesThis study aimed to evaluate the safety and feasibility of robotic-assisted thoracic surgery (RATS) after neoadjuvant chemoimmunotherapy in NSCLC.MethodsWe retrospectively collected data for NSCLC patients who received thoracic surgery after neoadjuvant chemoimmunotherapy from May 2020 to August 2022. Surgery details, pathological response, and perioperative outcome were compared between video-assisted thoracic surgery (VATS) group and RATS group. Inverse probability of treatment weighting (IPTW) was used to equal the baseline characteristics.ResultsA total of 220 patients were divided into 78 VATS patients and 142 RATS patients. There was no 90-day mortality in either group. RATS patients demonstrated better results in conversion rate to thoracotomy (VATS vs. RATS: 28.2% vs. 7.5%, P < 0.001), number of lymph node stations harvested (5.63 ± 1.75 vs. 8.09 ± 5.73, P < 0.001), number of lymph nodes harvested (13.49 ± 9.325 vs. 20.35 ± 10.322, P < 0.001), yield pathologic-N (yp-N) assessment (yp-N0, 88.5% vs. 67.6%; yp-N1, 7.6% vs. 12.6%; yp-N2, 3.8% vs. 19.7%; P < 0.001), and visual analog scale pain score after surgery (4.41 ± 0.93 vs. 3.77 ± 1.21, P=0.002). However, there were no significant differences in pathological response evaluation for neoadjuvant chemoimmunotherapy (P = 0.493) and complication rate (P = 0.803). After IPTW-adjustment, these results remained constant.ConclusionsRATS reduced the risk of conversion to thoracotomy, provided a better yp-N stage evaluation, and improved pain score; this suggests that RATS is safe and feasible for NSCLC patients after neoadjuvant chemoimmunotherapy

    The Oncogene IARS2 Promotes Non-small Cell Lung Cancer Tumorigenesis by Activating the AKT/MTOR Pathway

    A limited number of studies have indicated an association between isoleucyl-tRNA synthetase 2 (IARS2) and tumorigenesis. We evaluated IARS2 protein expression in lung tumor tissues and paired non-tumor tissues. We found higher IARS2 expression in the tumor tissues, which was associated with the late Tumor and Node stages of the Tumor, Node, Metastasis staging system. Silencing IARS2 inhibited the activity of A549 and H1299 cells, resulting in G0/G1 stasis of A549 cells and mitochondrial apoptosis. IARS2 silencing was also found to inhibit NSCLC tumor growth in nude mice. Complementary DNA microarray analysis revealed 742 differentially expressed genes (507 upregulated and 235 downregulated) in IARS2-silenced A549 cells compared to controls. Ingenuity Pathway Analysis of the differential expression data suggested that multiple pathways are associated with IARS2 silencing in NSCLC cells; upstream analysis predicted the activation or inhibition of transcriptional regulators. Correlation analysis revealed that AKT and MTOR activities were significantly inhibited in IARS2-silenced cells, but were partially restored by the AKT-stimulating agent SC79. IARS2 appears to regulate lung cancer cell proliferation via the AKT/MTOR pathway. Our results help clarify the complex roles of IARS2 in tumorigenesis and suggest that it may be a novel regulator of lung cancer development

    Objective: Investigate the status quo of the elderly living in the nursing home and their satisfaction and needs of nursing services, and analyze the service needs of the elderly living in the nursing home of Zhengzhou City. Methods: Randomly select 312 elderly people from 84 nursing homes for questionnaire survey and in-depth interview. Results: More than 60% of the respondents show their satisfaction on the current life in the nursing home. Sex, occupation, physical condition, dietary level, accommodation level, service level and health care conditions of the nursing home have a significant impact on the satisfaction of the elderly. Conclusion: With the growth of the aging of population, improving the level of service for the elderly and improve the service facilities is an essential way to improve the satisfaction and urgent needs of the elderly

    Hydrogen peroxide (H2O2) formed via the two-electron oxygen reduction reaction (2e-ORR) on Fe-N-C catalysts at elevated temperatures can cause catalyst degradation in fuel cells. In this work, we studied the effects of temperature on the selectivity of Fe-N-C catalysts for the 2e- and 4e-ORRs in acidic electrolytes. The results showed that the H2O2 yield increased with increasing temperature, and prolonged high-temperature exposure under ORR polarization conditions led to catalyst degradation and higher H2O2 yield. These effects were further explained by means of a kinetic model describing the competitive reactions and the thermodynamics of the system

    To study the regulation effect of apple polyphenol on fat metabolism in mice. The mice were randomly divided into 5 groups, except the basic control group, the other 4 groups were given high fat diet, body weight was measured once a week, and the obesity model was established. The basal control group and the obesity model group were intragastrically given normal saline (4.2mg/(kgbw.d), The three dose groups were gavaged separately with apple polyphenol 4.2, 8.4, 12.6 mg/(kg bw. d). After 4 consecutive weeks, the body weight of mice, the contents of total cholesterol (TC), total triglyceride (TG), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), malondialdehyde (MDA) in serum and the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), liver lipase (HL) and lipoprotein lipase (LPL) in serum. should be measured respectively. Apple polyphenol can significantly reduce the body weight, TG, LDL-C, TC and MDA contents, coupled with the atherosclerosis index (AI) of obese mice. It can increase the serum HDL-C content of obese mice, and effectively increase the activities of SOD, GSH-Px, HL and LPL in mice. Apple Polyphenol can reduce blood lipid and prevent atherosclerosis via regulating fat metabolism in mice

    Abstract Precision control of stereochemistry in radical reactions remains a formidable challenge due to the prevalence of incidental racemic background reactions resulting from undirected substrate oxidation in the absence of chiral induction. In this study, we devised an thoughtful approach—electricity-driven asymmetric Lewis acid catalysis—to circumvent this impediment. This methodology facilitates both asymmetric dienylation and allylation reactions, resulting in the formation of all-carbon quaternary stereocenters and demonstrating significant potential in the modular synthesis of functional and chiral benzoxazole-oxazoline (Boox) ligands. Notably, the involvement of chiral Lewis acids in both the electrochemical activation and stereoselectivity-defining radical stages offers innovative departures for designing single electron transfer-based reactions, significantly underscoring the relevance of this approach as a multifaceted and universally applicable strategy for various fields of study, including electrosynthesis, organic chemistry, and drug discovery

    Genome-wide identification of papain-like cysteine proteases in Citrus sinensis and expression analysis in response to Candidatus Liberibacter asiaticus

    ABSTRACTPapain-like cysteine proteases (PLCPs) play important roles in plant-pathogen interactions. In citrus, PLCPs are targeted by effectors from Candidatus Liberibacter asiaticus (CLas), the causal agent of Huanglongbing (HLB). To study interactions between Citrus sinensis and CLas, we identified 33 candidate CsPLCP genes by HMMsearch with the Peptidase_C1 domain (PF00112), and further confirmed them using local BLAST with PLCP sequences from Arabidopsis thaliana. The 33 CsPLCPs were classified into 9 subfamilies. Analysis of the cis-acting elements in their promoters indicated that CsPLCPs are responsive to various phytohormones and stressors. Further qPCR analysis indicated that several CsPLCPs were up-regulated during CLas infection. Subcellular localization analysis indicated that five CsPLCPs were localized to the cytoplasm and nucleus, while CsXBCP2 was localized only to the cytoplasm. In summary, this study provides a sound basis for understanding the roles of CsPLCPs in the Citrus-CLas interaction

    Complete remission through icotinib treatment in Non-small cell lung cancer epidermal growth factor receptor mutation patient with brain metastasis: A case report

    Brain metastasis (BM) has been universally recognized as a poor prognostic factor in non-small cell lung cancer (NSCLC). Epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors (TKIs) have shown efficacy in treating BM with an EGFR mutation. This paper reports a case of BM patient with EGFR-mutated NSCLC. According to the findings, a complete remission (CR) of the BM was achieved by icotinib treatment without conducting a radiotherapy, which was followed by a resection of the primary lung cancer lesion and lymph nodes. After one-year follow-up, the disease progressed to liver metastasis and liver lesion biopsy showed a T790M mutation. The patient responded well to the combination treatment of AZD9291 and icotinib after the failure of transcatheter arterial chemoembolization (TACE). This case report suggests that icotinib has a sustainable anticancer response to BM and the combination with icotinib and AZD9291 is effective for liver metastasis with T790M

    Sign-extension avoidance and word-length optimization by positive-offset representation for FIR filter design

    This brief proposes a new approach to utilizing positive-offset representation for sign-extension avoidance in shift-and-add implementation of a finite-impulse response filter. Affine arithmetic is used to model the excess offsets in order to curtail the word-length (WL) expansion problem. Tighter probabilistically justified WL bounds are determined to enable further offset to be removed from each tap. The approach is applicable even after the redundant adders in the multiplier block of the filter have been minimized. Our simulation results show an average power reduction of about 19% over and above the savings achieved by sharing of adders in multiple constant multiplication.Accepted versio

    IntroductionEpisyrphus balteatus is one representative Syrphidae insect which can provide extensive pollination and pest control services. To date, the symbiont composition and potential acquisition approaches in Syrphidae remain unclear.MethodsHerein, we investigated microbiota dynamics across developmental stages, different living states, and two sexes in E. balteatus via full-length 16S rRNA genes sequencing, followed by an attempt to explore the possibility of symbiont transmission from prey Megoura crassicauda to the hoverfly.ResultsOverall, Proteobacteria and Firmicutes were the dominant bacteria phyla with fluctuating relative abundances across the life stage. Cosenzaea myxofaciens is dominant in adulthood, while Enterococcus silesiacus and Morganella morganii dominate in larvae and pupae of E. balteatus, respectively. Unexpectedly, Serratia symbiotica, one facultative endosymbiont commonly harbored in aphids, was one of the predominant bacteria in larvae of E. balteatus, just behind Enterococcus silesiacus. In addition, S. symbiotica was also surprisingly most dominated in M. crassicauda aphids (92.1% relative abundance), which are significantly higher than Buchnera aphidicola (4.7% relative abundance), the primary obligate symbiont of most aphid species. Approximately 25% mortality was observed among newly emerged adults, of which microbiota was also disordered, similar to normally dying individuals. Sexually biased symbionts and 41 bacteria species with pairwise co-occurrence in E. balteatus and 23 biomarker species for each group were identified eventually. Functional prediction showed symbionts of hoverflies and aphids, both mainly focusing on metabolic pathways. In brief, we comprehensively explored the microbiome in one Syrphidae hoverfly using E. balteatus reared indoors on M. morganii as the model, revealed its dominated symbiont species, identified sexually biased symbionts, and found an aphid facultative endosymbiont inhabited in the hoverfly. We also found that the dominated symbiotic bacteria in M. crassicauda are S. symbiotica other than Buchnera aphidicola.DiscussionTaken together, this study provides new valuable resources about symbionts in hoverflies and prey aphids jointly, which will benefit further exploring the potential roles of microbiota in E. balteatus.</p
