    Water activity of Czech dry-cured meat products: Influence of sampling point and sample preparation method

    Water activity (aw) is a key factor influencing dry-cured meat products' safety. However, the standards for determining aw do not specify how the laboratory sample for self-analysis should be prepared and adjusted for determination in this type of food. This work aimed to verify whether the place of sampling and method of sample preparation of dry-cured meat products can influence the measured value of aw. For this purpose, samples of dry-cured heat-treated and fermented meat products were purchased from the local market. Samples before analysis were taken from the edges and centre of the meat products, and preparation consisted of: i) homogenisation; ii) dicing (4 × 4 × 4 mm); iii) slicing. The results of this work indicate that aw is significantly affected by both the part of the product from which the sample is taken and the method of preparation of the sample itself (P < 0.05). The highest measured values of aw were determined in samples prepared by slicing, and the lowest values were determined in homogenised samples. The place of sampling significantly affects the aw, especially for dry-cured heat-treated products

    Bacteriophages as a Strategy to Protect Potato Tubers against <i>Dickeya dianthicola</i> and <i>Pectobacterium carotovorum</i> Soft Rot

    The protective effect of bacteriophage suspensions (Ds3CZ + Ds20CZ and PcCB7V + PcCB251) on phytopathogenic bacteria causing soft rot of potato tubers, namely Dickeya dianthicola (D50, D200) and Pectobacterium carotovorum (P87, P224), was observed in ex vivo and in vitro experiments. Ex vivo tests were performed (with air access) on potato slices, on cylindrical cuts from the center of the tubers, and directly in whole potato tubers. In vitro experiments were carried out in a liquid medium using RTS-8 bioreactors, where bacterial growth was monitored as optical density. In particular, the inhibitory effects of phages were confirmed in experiments on potato slices, where suppression of rot development was evident at first glance. Phage treatment against selected bacteria positively affected potato hardness. Hardness of samples treated with bacteria only was statistically significantly reduced (p p < 0.001 for D200 and P87). Ex vivo experiments confirmed significant inhibition of P87 symptom development, partial inhibition of D200 and D50 in phage-treated tubers, and no effect was observed for P224. The inhibitory effect of phages against bacteria was not observed in the in vitro experiment

    Field Use of Protective Bacteriophages against Pectinolytic Bacteria of Potato

    The pectinolytic Dickeya solani bacterium is an important pathogen found in potatoes. We conducted laboratory and field experiments mimicking severe and mild Dickeya spp. infection and investigated the application of a mixture of two lytic bacteriophages before and after bacterial infection to protect the plants. Application of the phage solution to tuber disks and wounded tubers did not completely eliminate the infection but reduced the development of soft rot symptoms by 59.5–91.4%, depending on the phage concentration. In the field trial, plants treated with bacteriophages after severe Dickeya infection had 5–33% greater leaf cover and 4–16% greater tuber yield compared to untreated plants. When simulating a mild infection, leaf cover was 11–42% greater, and tuber yield was 25–31% greater compared to untreated plants. We conclude that the phage mixture has the potential to protect potatoes ecologically from D. solani

    The Effects of New Alibernet Red Wine Extract on Nitric Oxide and Reactive Oxygen Species Production in Spontaneously Hypertensive Rats

    We aimed to perform a chemical analysis of both Alibernet red wine and an alcohol-free Alibernet red wine extract (AWE) and to investigate the effects of AWE on nitric oxide and reactive oxygen species production as well as blood pressure development in normotensive Wistar Kyoto (WKY) and spontaneously hypertensive rats (SHRs). Total antioxidant capacity together with total phenolic and selected mineral content was measured in wine and AWE. Young 6-week-old male WKY and SHR were treated with AWE (24,2 mg/kg/day) for 3 weeks. Total NOS and SOD activities, eNOS and SOD1 protein expressions, and superoxide production were determined in the tissues. Both antioxidant capacity and phenolic content were significantly higher in AWE compared to wine. The AWE increased NOS activity in the left ventricle, aorta, and kidney of SHR, while it did not change NOS activity in WKY rats. Similarly, increased SOD activity in the plasma and left ventricle was observed in SHR only. There were no changes in eNOS and SOD1 expressions. In conclusion, phenolics and minerals included in AWE may contribute directly to increased NOS and SOD activities of SHR. Nevertheless, 3 weeks of AWE treatment failed to affect blood pressure of SHR

    Design and verification of additional filtration for the application of ecological transmission and hydraulic fluids in tractorc

    This contribution presents the design and function verification of additional filtration. It is intended for the common transmission and hydraulic oil filling of tractors. The main role of this filtration concept is to ensure a high level of oil cleanness as a condition for the application of ecologic fluids in tractors. The next one is to decrease the wear of lubricated tractor components, the degradation of oil and eventually to extend the interval of oil change. The designed additional filtering is characterized by ease installation through the use of quick couplings and hoses to the external hydraulic circuit. Therefore, the filtration is suitable for various tractor types. Filter element has been designed with the filter ability 1micron and the ability to separate to 0.5 dm3 of water from oil. Function of additional filtration was verified during the 150 engine hours of tractor operation. During this time period the oil contamination was evaluated on the basis of chemical elements content such as Fe, Cu, Si, Al, Ni, Mo and Cr. The additive concentration was evaluated on the basis of chemical elements content such as Ca, P and Zn. During the test operation of tractor the concentration decrease of chemical elements reached the values 25.53 % (Fe), 23.53 % (Si), 25 % (Al) and 5.5 % (Cu). The decrease of additive concentration reached only medium level (6.6 %). Therefore, the designed additional filtration doesn’t remove additives from oil. Based on the evaluation of the content of chemical elements (that representing contamination and additives), we can say that the designed filtering method is suitable for use in agricultural tractors
