Platelet derived growth factor B gene expression in the Xenopus laevis developing central nervous system
Platelet-derived growth factor B (PDGF-B) belongs to the mitogen and growth factor family and like the other members it has many roles in cell differentiation, proliferation and migration during development, adult life and in pathological conditions. Among them it has been observed that aberrant PDGF signalling is frequently linked to glioma development and progression, and Pdgf-b over-expression in mouse neural progenitors leads to the formation of gliomas. Despite this evidence, the mechanisms underlying PDGF-B driven tumorigenesis and its role during brain development are not fully understood. In order to contribute to clarifying possible new roles of pdgf-b signalling, we present here the embryonic gene expression pattern of pdgf-b, so far unknown in early vertebrate development. By using Xenopus laevis as a model system we performed qRT-PCR and whole mount in situ hybridization. Pdgf-b mRNA is expressed in discrete regions of the developing central nervous system, in the cranial nerve placodes and in the notochord. We also compared the gene expression of pdgf-b with that of its receptor pdgfr-a suggesting so far unsuspected roles for this signalling pathway during the development of specific embryonic structures
Comparison in Serotonin Receptor 1A and 2A expressions along the antero-posterior axis of human hippocampus in Major Depressed patients with and without medications and psychiatrically healthy controls, and Serotonin Receptor 4 expression pattern analysis
Major Depressive Disorder, often referred as Major Depression, is the second cause of death in people aged between 19 and 25-years-old, but despite the increasing awareness, the molecular mechanisms of depression are still poorly understood. Since Major Depression can affect hippocampal structures, and since antidepressants treatment involving increasing amount of serotonin in synaptic cleft due to the blockage of serotonin reuptake, as the action exerted by the first-line antidepressant Selective Serotonin Reuptake Inhibitor, can reverse depressive affection, serotonin receptors have been highlighted as mediator of serotonin role in depressive pathogenesis and etiology.
Particularly, the expression patterns of Serotonin Receptor 1A (5-HT1A), 2A (5-HT2A) and 4 (5-HT4), widely reported to be most likely involved in Major Depression and suicide pathophysiology, have been studied in human hippocampal sections, collected post-mortem from unmedicated and SSRIs-medicated subjects with Major Depressive Disorder, and compared to age- and gender-matched psychiatrically healthy controls. Each serotonin receptor expression has been localized on the hippocampal sections using specific [35S]-radiolabeled riboprobe, and it has been quantified by using Densita software for autoradiographic quantification.
5-HT4 expression has been detected in human hippocampus for the first time, and together with 5-HT1A show an evolutionary preserved expression pattern in mammal hippocampus, and an apparent co-localization with 5-HT1A in Dentate Gyrus, while 5-HT2A displays a neat expression in human Dentate Gyrus, that has not been observed in rodent and primate ones. 5-HT1A and 5-HT2A expressions in human hippocampus have been analized along the antero-posterior axis, with significant p-values for 5-HT1A expression gradient but low R2 values in psychiatrically healthy controls and SSRIs-medicated subjects in Dentate Gyrus (respectively, p=0.0412, R2=0.1924; p=0.000266, R2=0.4397), in Cornu Ammonis 3 (respectively, p=0.0287, R2=0.0971; , p=0.00959, R2=0.3699) and in Cornu Ammonis 1 (respectively, p=0.0353, R2= 0.203; p=0.0144, R2=0.2641), while subjects 5-HT2A antero-posterior expression doesn’t have any peculiar p-value or R2 value. Age of SSRIs-medicated subjects has some inference on 5-HT1A expression in anterior (p=0.0436; R2=0.6828) and mid (p=0.0301; R2=0.8597) Dentate Gyrus, while the affection on 5-HT2A expression by Age is clearly present in psychiatrically healthy controls in anterior (p=0.0301; R2=0.7044) and mid (p=0.00996; R2=0.8913) Dentate Gyrus and in anterior (p=0.00158; R2=0.9461) Cornu Ammonis 1. Since not all the samples have a normal distribution, comparisons of 5-HT1A and 5-HT2A expression between controls and unmedicated and medicated depressed subjects have been analyzed using, side-by-side, a parametric and a non-parametric statistical analysis, considering also the Age factor as a covariate: Kruskall-Wallis (non-parametric) test has highlighted significant or
close-to-significativity p-values in anterior Dentate Gyrus (p=0.06475) and anterior Cornu Ammonis 3 (p=0.04952) only for 5-HT2A expression, while One-Way Anova with Age as covariate as found significant comparisons in anterior (p=0.0391) and mid (p=0.024) Dentate Gyrus for 5-HT1A expression, and significant comparisons in anterior Dentate Gyrus (p=0.0044), anterior Cornu Ammonis 3 (p=0.00644) and anterior Cornu Ammonis 1 (p=0.0181).
Even considering the strongest limitation of this study, which is the small number of available subjects, observing neat differences in 5-HT1A and 5-HT2A expressions between younger controls, SSRIs-medicated and unmedicated major depressed subjects is still possible. These data may suggest a major sensitivity of younger subjects to mood disorders
Validazione in vivo della correlazione funzionale tra segnalazione del PDGF-B e l?espressione della R-caderina e di altre caderine riscontrata in gliomi di alto grado
Gli HGG (High Grade Glioma) sono i principali tumori del Sistema Nervoso Centrale.
Originano da cellule della glia (da cui il nome), e sono costituiti da cellule anaplastiche.
Una loro peculiarità è l’abilità di infiltrarsi nel parenchima cerebrale, estendendo così i
confini del tumore. Gli approcci terapeutici di tipo chirurgico e/o farmacologico o
radioterapico non risultano ad oggi risolutivi, rendendo questo tipo di tumori pressoché
incurabile.
Una strategia, con lo scopo di uccidere selettivamente queste cellule o comunque inibirne
le capacità invasive, potrebbe essere cruciale per combattere questa tipologia di gliomi
maligni.
Una percentuale di gliomi è caratterizzata dalla sovraespressione del fattore di crescita
PDGF-B ed è stato dimostrato, in modello murino, che il PDGF-B è necessario al tumore
per mantenere la tumorigenicità in termini di grado di proliferazione e capacità di infiltrare
il parenchima cerebrale probabilmente annullando il fenomeno dell’inibizione da contatto
cellula-cellula. Esperimenti condotti dal Dott. Paolo Malatesta dell’Istituto Italiano Tumori
di Genova, hanno suggerito una possibile associazione tra la sovraespressione del PDGF-B
e la sovraespressione di una particolare proteina d’adesione nota come Caderina 4 o R-
caderina (così chiamata perché identificata per la prima volta nella retina di pollo). In
particolare, nei gliomi che presentano una sovraespressione del PDGF-B, si osserva un
fenomeno di delocalizzazione della N-caderina a livello delle giunzioni aderenti e la
sovraespressione a livello di membrana della R-caderina durante l’acquisizione di motilità
nelle cellule tumorali. Quello da me trattato durante il mio internato di tesi, è uno dei
molteplici
aspetti
dell’interazione
tra
PDGF-B
e
invasività
cellulare
tramite
sovraespressione indotta di tale fattore nelle cellule delle creste neurali (NCC) di Xenopus
laevis, e di come questa alterazione possa influire sull'espressione tanto della R-caderina,
quanto di altre molecole di adesione. Le NCC costituiscono un ottimo modello di analisi
per lo switch delle caderine e per la motilità cellulare, dal momento che è quello che
normalmente avviene durante lo sviluppo e specificatamente durante la transizione epitelio
mesenchimatica. Ho quindi clonato il cDNA per la R-caderina umana in uno specifico
vettore di espressione (pCS2+), da cui ho trascritto il relativo capped mRNA poi
microiniettato in embrioni di X. laevis allo stato di 2-4 cellule. Gli embrioni con la
sovraespressione della R-caderina sono stati analizzati mediante ibridazione in situ “whole
mount” a vari stadi di sviluppo utilizzando markers specifici delle creste neurali. I fenotipi
ottenuti sono stati correlati a quelli ottenuti precedentemente con la sovraespressione del
PDGF-B. Per verificare, invece, l’effettiva correlazione tra signalling del PDGF-B ed R-
caderina, sono state fatte sia una qPCR con primers progettati per la caderina 4 di Xenopus
sul cDNA ottenuto da embrioni in cui era stata indotta la sovraespressione di PDGF-B, sia
un’ibridazione in situ trascrivendo il probe specifico per l’mRNA di XCDH4 e testandolo
su embrioni microiniettati con il PDGF-B. È attualmente in corso anche la ricerca di altre
molecole di adesione coinvolte con la migrazione delle NCC che possano essere modulate
dalla sovraespressione del PDGF-B. I livelli di espressione di queste molecole sono
analizzati sia quantitativamente che qualitativamente tramite qPCR ed Ibridazione In Situ
“whole mount”.
I risultati ottenuti, ancora in corso di completamento, supportano l’idea di una correlazione
funzionale tra il signalling del PDGF-B e l’R-caderina anche in un modello animale in
vivo. E’ auspicabile che l’insieme di questi dati possa contribuire a chiarire le basi
molecolari della tumorigenicità indotta dal PDGF-B nell’ottica di futuri approcci
terapeutici
Antibodies targeting the European lobster (Palinurus elephas) vitellogenin developed by mRNA isolation and in-silico-designed antigenic peptides
Vitellogenin is an essential protein involved in ovary maturation in many animals. Detection of this protein correlated with reproductive capacity may be important if carried out on marine organisms such as the red spiny lobster Palinurus elephas, a crustacean economically important crop from wild fish catches. Moreover, in recent years, vitellogenin has assumed an important role as a possible biomarker of marine environmental pollution, as its expression levels can be influenced by the presence of similar oestrogen pollutants and affect the reproductive sphere of marine organisms such as crustaceans. The P. elephas vitellogenin protein and its coding gene have never been isolated, so there is little information about its presence in this lobster. The aim of the present study was to develop a molecular strategy to create, for the first time, an antibody for the detection and quantization of vitellogenin in P. elephas
