    Automated satellite remote sensing of giant kelp at the Falkland Islands (Islas Malvinas)

Plos One, 17(1), (2022): e0257933, https://doi.org/10.1371/journal.pone.0257933.Giant kelp populations that support productive and diverse coastal ecosystems at temperate and subpolar latitudes of both hemispheres are vulnerable to changing climate conditions as well as direct human impacts. Observations of giant kelp forests are spatially and temporally uneven, with disproportionate coverage in the northern hemisphere, despite the size and comparable density of southern hemisphere kelp forests. Satellite imagery enables the mapping of existing and historical giant kelp populations in understudied regions, but automating the detection of giant kelp using satellite imagery requires approaches that are robust to the optical complexity of the shallow, nearshore environment. We present and compare two approaches for automating the detection of giant kelp in satellite datasets: one based on crowd sourcing of satellite imagery classifications and another based on a decision tree paired with a spectral unmixing algorithm (automated using Google Earth Engine). Both approaches are applied to satellite imagery (Landsat) of the Falkland Islands or Islas Malvinas (FLK), an archipelago in the southern Atlantic Ocean that supports expansive giant kelp ecosystems. The performance of each method is evaluated by comparing the automated classifications with a subset of expert-annotated imagery (8 images spanning the majority of our continuous timeseries, cumulatively covering over 2,700 km of coastline, and including all relevant sensors). Using the remote sensing approaches evaluated herein, we present the first continuous timeseries of giant kelp observations in the FLK region using Landsat imagery spanning over three decades. We do not detect evidence of long-term change in the FLK region, although we observe a recent decline in total canopy area from 2017–2021. Using a nitrate model based on nearby ocean state measurements obtained from ships and incorporating satellite sea surface temperature products, we find that the area of giant kelp forests in the FLK region is positively correlated with the nitrate content observed during the prior year. Our results indicate that giant kelp classifications using citizen science are approximately consistent with classifications based on a state-of-the-art automated spectral approach.     TARP γ-7 selectively enhances synaptic expression of calcium-permeable AMPARs

    Regulation of calcium-permeable AMPA receptors (CP-AMPARs) is crucial in normal synaptic function and neurological disease states. Although transmembrane AMPAR regulatory proteins (TARPs) such as stargazin (γ-2) modulate the properties of calcium-impermeable AMPARs (CI-AMPARs) and promote their synaptic targeting, the TARP-specific rules governing CP-AMPAR synaptic trafficking remain unclear. We used RNA interference to manipulate AMPAR-subunit and TARP expression in γ-2–lacking stargazer cerebellar granule cells—the classic model of TARP deficiency. We found that TARP γ-7 selectively enhanced the synaptic expression of CP-AMPARs and suppressed CI-AMPARs, identifying a pivotal role of γ-7 in regulating the prevalence of CP-AMPARs. In the absence of associated TARPs, both CP-AMPARs and CI-AMPARs were able to localize to synapses and mediate transmission, although their properties were altered. Our results also establish that TARPed synaptic receptors in granule cells require both γ-2 and γ-7 and reveal an unexpected basis for the loss of AMPAR-mediated transmission in stargazer mice

    A citizen science approach to teaching climate change in introductory-level undergraduate general science courses

    Hundreds of thousands of undergraduates enroll in general education science courses to fulfill university core requirements. However, many of these lecture-based courses fail to foster high-level data literacy skills. This work details the design, implementation, and analysis of a new climate change-based classroom activity for college students that pairs data interpretation with participation in an online citizen science project called Floating Forests (http://floatingforests.org). We pilot tested our activity in introductory geoscience and biology courses at six universities (∼1,500 students) during the 2020–2021 academic year. Additionally, we developed and validated a survey to assess how engagement with our activity impacted students’ self-reported changes across four factors: (1) perceptions of the impacts of climate change, (2) data literacy self-efficacy, (3) beliefs about the value of citizen science, and (4) beliefs about science engagement. In a pre- to post-test comparison, students who utilized our activity in their courses showed statistically significant increases (p < 0.05) across all four factors. These results highlight the potential benefit of implementing data-driven, citizen science-based activities in introductory-level undergraduate courses.</p
