    On the Survival of Short-Period Terrestrial Planets

    The currently feasible method of detection of Earth-mass planets is transit photometry, with detection probability decreasing with a planet's distance from the star. The existence or otherwise of short-period terrestrial planets will tell us much about the planet formation process, and such planets are likely to be detected first if they exist. Tidal forces are intense for short-period planets, and result in decay of the orbit on a timescale which depends on properties of the star as long as the orbit is circular. However, if an eccentric companion planet exists, orbital eccentricity (eie_i) is induced and the decay timescale depends on properties of the short-period planet, reducing by a factor of order 105ei210^5 e_i^2 if it is terrestrial. Here we examine the influence companion planets have on the tidal and dynamical evolution of short-period planets with terrestrial structure, and show that the relativistic potential of the star is fundamental to their survival.Comment: 13 pages, 2 figures, accepted for publication in Ap

    Self-Assessment and Planned Change of Placement and Career Services Center

    In the 1990s, college and university career services and placement units face many challenges which may influence their success including (a) increased budgetary constraints, (b) changing student demographics, (c) increased availability of computer technologies, (d) new service delivery models, and (e) changing employer recruitment practices. In an effort to address these issues and examine its role within the university (Roth, 1994), the Placement and Career Information Center (PCIC) at Central Michigan University undertook a program of applied research, self-assessment and planned change. The purpose of this article is to briefly report our experiences in conducting this program of applied research. The assessment strategy is presented in the first part of this report. Next, the methods used to collect data and assessment, sampling procedures and response rates are described. Following this, highlights of the assessment results are presented including a summary of some changes already made and those planned for the future. The results of this study are presented in detail in Adams, et at. (1994)

    Synthesis and Cell Adhesive Properties of Linear and Cyclic RGD Functionalized Polynorbornene Thin Films

    Described herein is the efficient synthesis and evaluation of bioactive arginine-glycine-aspartic acid (RGD) functionalized polynorbornene-based materials for cell adhesion and spreading. Polynorbornenes containing either linear or cyclic RGD peptides were synthesized by ring-opening metathesis polymerization (ROMP) using the well-defined ruthenium initiator [(H_(2)IMes)(pyr)_(2)(Cl)_(2)Ru═CHPh]. The random copolymerization of three separate norbornene monomers allowed for the incorporation of water-soluble polyethylene glycol (PEG) moieties, RGD cell recognition motifs, and primary amines for postpolymerization cross-linking. Following polymer synthesis, thin-film hydrogels were formed by cross-linking with bis(sulfosuccinimidyl) suberate (BS^3), and the ability of these materials to support human umbilical vein endothelial cell (HUVEC) adhesion and spreading was evaluated and quantified. When compared to control polymers containing either no peptide or a scrambled RDG peptide, polymers with linear or cyclic RGD at varying concentrations displayed excellent cell adhesive properties in both serum-supplemented and serum-free media. Polymers with cyclic RGD side chains maintained cell adhesion and exhibited comparable integrin binding at a 100-fold lower concentration than those carrying linear RGD peptides. The precise control of monomer incorporation enabled by ROMP allows for quantification of the impact of RGD structure and concentration on cell adhesion and spreading. The results presented here will serve to guide future efforts for the design of RGD functionalized materials with applications in surgery, tissue engineering, and regenerative medicine

    Alterations in vasodilator-stimulated phosphoprotein (VASP) phosphorylation: associations with asthmatic phenotype, airway inflammation and ÎČ\u3csub\u3e2\u3c/sub\u3e-agonist use

    Background Vasodilator-stimulated phosphoprotein (VASP) mediates focal adhesion, actin filament binding and polymerization in a variety of cells, thereby inhibiting cell movement. Phosphorylation of VASP via cAMP and cGMP dependent protein kinases releases this brake on cell motility. Thus, phosphorylation of VASP may be necessary for epithelial cell repair of damage from allergen-induced inflammation. Two hypotheses were examined: (1) injury from segmental allergen challenge increases VASP phosphorylation in airway epithelium in asthmatic but not nonasthmatic normal subjects, (2) regular in vivo ÎČ2-agonist use increases VASP phosphorylation in asthmatic epithelium, altering cell adhesion. Methods Bronchial epithelium was obtained from asthmatic and non-asthmatic normal subjects before and after segmental allergen challenge, and after regularly inhaled albuterol, in three separate protocols. VASP phosphorylation was examined in Western blots of epithelial samples. DNA was obtained for ÎČ2-adrenergic receptor haplotype determination. Results Although VASP phosphorylation increased, it was not significantly greater after allergen challenge in asthmatics or normals. However, VASP phosphorylation in epithelium of nonasthmatic normal subjects was double that observed in asthmatic subjects, both at baseline and after challenge. Regularly inhaled albuterol significantly increased VASP phosphorylation in asthmatic subjects in both unchallenged and antigen challenged lung segment epithelium. There was also a significant increase in epithelial cells in the bronchoalveolar lavage of the unchallenged lung segment after regular inhalation of albuterol but not of placebo. The haplotypes of the ÎČ2-adrenergic receptor did not appear to associate with increased or decreased phosphorylation of VASP. Conclusion Decreased VASP phosphorylation was observed in epithelial cells of asthmatics compared to nonasthmatic normals, despite response to ÎČ-agonist. The decreased phosphorylation does not appear to be associated with a particular ÎČ2-adrenergic receptor haplotype. The observed decrease in VASP phosphorylation suggests greater inhibition of actin reorganization which is necessary for altering attachment and migration required during epithelial repair

    Tsetse fly (Glossina pallidipes) midgut responses to Trypanosoma brucei challenge

    Abstract Background Tsetse flies (Glossina spp.) are the prominent vector of African trypanosome parasites (Trypanosoma spp.) in sub-Saharan Africa, and Glossina pallidipes is the most widely distributed species in Kenya. This species displays strong resistance to infection by parasites, which are typically eliminated in the midgut shortly after acquisition from the mammalian host. Although extensive molecular information on immunity for the related species Glossina morsitans morsitans exists, similar information is scarce for G. pallidipes. Methods To determine temporal transcriptional responses of G. pallidipes to Trypanosoma brucei brucei challenge, we conducted Illumina based RNA-seq on midgut organ and carcass from teneral females G. pallidipes at 24 and 48 h post-challenge (hpc) with T. b. brucei relative to their respective controls that received normal blood meals (without the parasite). We used a suite of bioinformatics tools to determine differentially expressed and enriched transcripts between and among tissues, and to identify expanded transcripts in G. pallidipes relative to their orthologs G. m. morsitans. Results Midgut transcripts induced at 24 hpc encoded proteins were associated with lipid remodelling, proteolysis, collagen metabolism, apoptosis, and cell growth. Midgut transcripts induced at 48 hpc encoded proteins linked to embryonic growth and development, serine endopeptidases and proteosomal degradation of the target protein, mRNA translation and neuronal development. Temporal expression of immune responsive transcripts at 48 relative to 24 hpc was pronounced, indicative of a gradual induction of host immune responses the following challenge. We also searched for G. m. morsitans orthologous groups that may have experienced expansions in the G. pallidipes genome. We identified ten expanded groups in G. pallidipes with putative immunity-related functions, which may play a role in the higher refractoriness exhibited by this species. Conclusions There appears to be a lack of strong immune responses elicited by gut epithelia of teneral adults. This in combination with a compromised peritrophic matrix at this stage during the initial phase of T. b. brucei challenge may facilitate the increased parasite infection establishment noted in teneral flies relative to older adults. Although teneral flies are more susceptible than older adults, the majority of tenerals are still able to eliminate parasite infections. Hence, robust responses elicited at a later time point, such as 72 hpc, may clear parasite infections from the majority of flies. The expanded G. m. morsitans orthologous groups in G. pallidipes may also be functionally associated with the enhanced refractoriness to trypanosome infections reported in G. pallidipes relative to G. m. morsitans

    Quantifying annual variation in catchability for commercial and research fishing

    Biomass indices, from commercial catch per unit of effort (CPUE) or random trawl surveys, are commonly used in fisheries stock assessments. Uncertainty in such indices, often ex-pressed as a coefficient of variation (CV), has two components: observation error, and annual variation in catchability. Only the former can be estimated directly. As a result, the CVs used for these indices either ignore the annual-variation component or assume a value for it (often implicitly). Two types of data for New Zealand stocks were examined: 48 sets of residuals and catchability estimates from stock assessments using either CPUE or trawl survey indices; and biomass estimates from 17 time series of trawl surveys with between 4 and 25 species per time series. These data show clear evidence of significant annual variation in catchability. With the trawl survey data, catchability was detectably extreme for many species in about one year in six. The assessment data suggest that this annual variability typically has a CV of about 0.2. For commercial CPUE the variability is slightly less, and a typical total CV (including both components) of 0.15 to 0.2. This is much less than the values of 0.3 to 0.35 that have commonly been assumed in New Zealand. Some estimates of catchability are shown to be implausible

    Discovery of Recent Star Formation in the Extreme Outer Regions of Disk Galaxies

    We present deep Halpha images of three nearby late-type spiral galaxies (NGC628, NGC1058 and NGC6946), which reveal the presence of HII regions out to, and beyond, two optical radii (defined by the 25th B-band isophote). The outermost HII regions appear small, faint and isolated, compared to their inner disk counterparts, and are distributed in organized spiral arm structures, likely associated with underlying HI arms and faint stellar arms. The relationship between the azimuthally--averaged Halpha surface brightness (proportional to star formation rate per unit area) and the total gas surface density is observed to steepen considerably at low gas surface densities. We find that this effect is largely driven by a sharp decrease in the covering factor of star formation at large radii, and not by changes in the rate at which stars form locally. An azimuthally--averaged analysis of the gravitational stability of the disk of NGC6946 reveals that while the existence of star formation in the extreme outer disk is consistent with the Toomre-Q instability model, the low rates observed are only compatible with the model when a constant gaseous velocity dispersion is assumed. We suggest that observed behaviour could also be explained by a model in which the star formation rate has an intrinsic dependence on the azimuthally-averaged gas volume density, which decreases rapidly in the outer disk due to the vertical flaring of the gas layer.Comment: 10 pages, 2 embedded postscript files, 3 jpeg images; accepted for publication in ApJ Letter

    Gut-seeded α-synuclein fibrils promote gut dysfunction and brain pathology specifically in aged mice

    Parkinson’s disease is a synucleinopathy that is characterized by motor dysfunction, death of midbrain dopaminergic neurons and accumulation of α-synuclein (α-Syn) aggregates. Evidence suggests that α-Syn aggregation can originate in peripheral tissues and progress to the brain via autonomic fibers. We tested this by inoculating the duodenal wall of mice with α-Syn preformed fibrils. Following inoculation, we observed gastrointestinal deficits and physiological changes to the enteric nervous system. Using the AAV-PHP.S capsid to target the lysosomal enzyme glucocerebrosidase for peripheral gene transfer, we found that α-Syn pathology is reduced due to the increased expression of this protein. Lastly, inoculation of α-Syn fibrils in aged mice, but not younger mice, resulted in progression of α-Syn histopathology to the midbrain and subsequent motor defects. Our results characterize peripheral synucleinopathy in prodromal Parkinson’s disease and explore cellular mechanisms for the gut-to-brain progression of α-Syn pathology

    Estrogen effects on triglyceride metabolism in analbuminemic rats.BackgroundTriglyceride (TG) levels are normally lower in female rats, while the opposite is the case in the Nagase analbuminemic rats (NAR). Increased TG levels in normal males are caused by a testosterone-mediated decrease in postheparin (PH) lipoprotein lipase (LpL). Castration of males reduces TG, while castration of females is without effect. TG levels are reduced by castration of the female NAR, suggesting that estrogen rather than testosterone causes hypertriglyceridemia in this strain. The mechanism for this increase is unknown.MethodsWe measured secretion of very-low density lipoprotein (VLDL) TG using Triton WR 1339 clearance as the disappearance from blood of 3H-trioleate and 14C-cholesterol–labeled chylomicrons (CM), and the activity of the PH lipases: LpL and hepatic lipase (HL). All were determined in Sprague-Dawley (SD) and NAR female, male, and ovariectomized (OVX) rats.ResultsTG levels were significantly greater in female NAR in comparison to all other groups. Ovariectomy of NAR significantly ameliorated hypertriglyceridemia. VLDL TG secretion was significantly greater in intact female NAR compared with all other groups. There were no other differences in VLDL TG secretion among the other groups. The clearance of CM was greatest in female SD rats, and OVX had no effect. NAR cleared CM less well than did SD rats (P < 0.001), but among NAR, clearance was greatest in OVX NAR and male NAR (P < 0.002). Both PH LpL activity and HL activity were lowest in female NAR (P < 0.05). Ovariectomy partially corrected the defect in HL (P < 0.05).ConclusionTG levels in female NAR are in part a result of increased VLDL-TG secretion, an effect mediated by estrogen. The presence of an estrogen-mediated catabolic defect that was alleviated by OVX was also observed. This catabolic defect is likely a result of an estrogen-mediated decrease both in LpL and HL expressed only in the presence of analbuminemia
