    Arsenite-Induced Alterations of DNA Photodamage Repair and Apoptosis After Solar-Simulation UVR in Mouse Keratinocytes in Vitro

    Our laboratory has shown that arsenite markedly increased the cancer rate caused by solar-simulation ultraviolet radiation (UVR) in the hairless mouse skin model. In the present study, we investigated how arsenite affected DNA photodamage repair and apoptosis after solar-simulation UVR in the mouse keratinocyte cell line 291.03C. The keratinocytes were treated with different concentrations of sodium arsenite (0.0, 2.5, 5.0 μM) for 24 hr and then were immediately irradiated with a single dose of 0.30 kJ/m(2) UVR. At 24 hr after UVR, DNA photoproducts [cyclobutane pyrimidine dimers (CPDs) and 6–4 photoproducts (6-4PPs)] and apoptosis were measured using the enzyme-linked immunosorbent assay and the two-color TUNEL (terminal deoxynucleotide transferase dUTP nick end labeling) assay, respectively. The results showed that arsenite reduced the repair rate of 6-4PPs by about a factor of 2 at 5.0 μM and had no effect at 2.5 μM. UVR-induced apoptosis at 24 hr was decreased by 22.64% at 2.5 μM arsenite and by 61.90% at 5.0 μM arsenite. Arsenite decreased the UVR-induced caspase-3/7 activity in parallel with the inhibition of apoptosis. Colony survival assays of the 291.03C cells demonstrate a median lethal concentration (LC(50)) of arsenite of 0.9 μM and a median lethal dose (LD(50)) of UVR of 0.05 kJ/m(2). If the present results are applicable in vivo, inhibition of UVR-induced apoptosis may contribute to arsenite’s enhancement of UVR-induced skin carcinogenesis

    ASSESSMENT OF LEVELS OF PHYSICAL ACTIVITY IN PERIMENOPAUSAL WOMEN

    Aim: Various physical, psychological and hormonal changes during the perimenopause may affect their quality of life. It results in Joint pain, stiffness and urinary incontinence may limit the ability to take the exercise and to socialize. Poor sleep and mood changes can have an impact on women`s ability to work effectively. Objective: To categories level of physical activity in terms of light, moderate and vigorous through RAPA scale. Methodology: Questionnaire based survey was carried out in 100 perimenopausal women aged 45 to 55 years. After the anthropometric and demographic assessment RAPA was administered and then data was analyzed. Result: Out of 100 subjects only 17 women were categorized as active with moderate physical activity for 30 minutes on 5 or more days of week and 10 did vigorous physical activity for 20 minutes for 3 or more days a week and were called as active. Out of the 10 active only 4 did flexibility and no strengthening. Conclusion: Only 10 perimenopausal women could reach second level of RAPA. 22 were eliminated in question no. 1 i.e. they were inactive apart from daily household activities .     The skeleton: an overlooked regulator of systemic glucose metabolism in cancer?

    Recent discoveries demonstrated the skeleton’s role as an endocrine organ regulating whole-body glucose homeostasis. Glucose metabolism is critical for rapid cell proliferation and tumour growth through increasing glucose uptake and fermentation of glucose to lactate despite being in an aerobic environment. This hypothesis paper discusses emerging evidence on how bones can regulate whole-body glucose homeostasis with potential to impact on tumour growth and proliferation. Moreover, it proposes a clinical link between bone glucose metabolism and prognosis of cancer based on recent clinical trial data. Targeting metabolic pathways related with classic glucose metabolism and also bone metabolism, novel methods of cancer therapy and treatment could be developed. This paper objective is to highlight the need for future research on this altered metabolism with potential to change future management of cancer patients

    We report an 11-month-old boy who brought with status epilepticus. The presence of bilateral retinal hemorrhages and generalizedcerebral atrophy indicating ischemia revealed on imaging tests lead us to keep a differential diagnosis of shaken baby syndrome (SBS).On repeated questioning, mother gave the history of shaking by a relative which further supported our diagnosis. Thus in a child with anonspecific febrile convulsive state, the treating physician should keep SBS also as a differential diagnosis

    CORRELATION OF ACSM\u27S WALKING EQUATION AND DIRECTLY MEASURED VO2MAX IN INDIAN POPULATION: A PILOT STUDY

    Purpose of study: American College of Sports and Medicine equation is applied in many cardiovascular centers and sports medicine laboratory. It is used in majority of the commercial software available for exercise testing ACSM has published several metabolic equations for the indirect estimation of VO2max. So the purpose of the study was to find the applicability of ACSM walking equation in Indian population. Aim and Objective: To find out the correlation between actual VO2max and predicted VO2max 18-25 years female in Nagpur. Methodology: All the participants were selected according to sample of convenience and given a thorough explanation of the procedure in the language they understand. Each participant signed written informed consent before participating in the study in the language they could comprehend. Subjects were screened on the basis of selection criteria and PAR-q & you questionnaire. Total 20 participants were selected and demographic and anthropometric data was recorded. In this VO2 max was calculated directly by gas analyzer using AD instrument and indirectly by ACSM\u27s walking equation and data was analyzed. Result: The mean and standard deviation of directly measured VO2max (ml/kg/min) is 8.6354 and ±4.5557 this shows the range of VO2max for the studied population and VO2max predicted by ACSM walking equation is 38.1891 and ±5.4691. The pearson\u27s correlation for direct and indirect measurement of VO2max suggested that there is no co-relations with the value (r=0.11, p=, 0.05). Conclusion: The present study concluded that there is no correlation between actual and predicted VO2 max in the studied population and need to develop new regression equation for studied population i.e;ACSM equation is not applicable to the studied population.
