Development of control strategies for bacteria and fungi associated with a micropropagated new cultivar of orange-fleshed sweet potato (Ipomoea batatas cv. Agrosavia–Aurora)
Vegetative propagation of sweet potato lead to the accumulation of diseases from generation to generation, which represents a threat to both productivity and conservation of genetic resources. In vitro techniques can help to overcome phytosanitary problems by applying plant material cleaning strategies. The objective of this study was to develop in vitro micropropagation strategies for the production of high-quality plant material of an orange-fleshed variety of sweet potato recently released in Colombia. Molecular identification of contaminating microorganisms was performed by sequencing the 16S rRNA gene for bacteria and ITS for fungi. Five disinfection protocols were evaluated, three of which were previously developed for sweet potato and included disinfection with 0.5, 1, and 2% sodium hypochlorite respectively, while two protocols are proposed in this work and included washing with povidone-iodine, disinfection with sodium hypochlorite 2%; one of these two new protocols also contains acetic acid and quaternary ammonium. For the evaluation of the viability of in vitro plants after disinfection, they were acclimatized in a greenhouse, reintroduced, and a molecular testing by PCR of 16S rRNA gene and ITS was carried out to verify the phytosanitary status of the material. The contaminating microorganisms found were filamentous fungi of the genera Fusarium, Sarocladium, Cladosporium and Aspergillus, yeasts of the genera Pseudozyma and Moesziomyces, and the actinobacterium Curtobacterium sp. The results indicated that washing with povidone-iodine and disinfection with 2% sodium hypochlorite, acetic acid and quaternary ammonium was the most efficient disinfection protocol, reducing the number of contaminated cultures by up to 10% and eradicating 70% of contaminants. The in vitro plants established in the greenhouse remained healthy and, after reintroduction, the molecular test for bacteria and fungi was negative. These results allowed the generation of an optimized protocol that can be incorporated into the in vitro micropropagation process to generate contamination-free sweet potato seeds

Rapid access to tetracyclic ring system of lennoxamine type natural product by combined use of a novel three-component reaction and Pummerer cyclization
From readily available allylamine, aldehyde and isocyanoacetamide, a three-component reaction followed by a Pummerer cyclization provided tetracyclic ring systems [6-7-5-6 (I)and 6-6-5-6 (II)] in excellent overall yields. [on SciFinder (R)
Truncated diastereoselective Passerini reaction, a rapid construction of polysubstituted oxazole and peptides having an alpha -hydroxy-beta -amino acid component
The reaction of aldehydes and ketones, including aliph. and arom. ones, with amides of alpha -isocyano-beta -phenylpropionic acid in toluene in the presence of lithium bromide gives 2,4,5-trisubstituted oxazoles in good to excellent yield. Protected chiral alpha -amino aldehydes participate in this reaction to give, after hydrolysis of the oxazoles, norstatine-contg. peptides in good overall yield. The nucleophilic addn. of isonitriles to N,N-dibenzylphenylalanal is investigated for the first time and is found to be stereoselective leading predominantly to the anti-adduct (dr = 9/1). On the other hand, the reaction between the N-Boc phenylalanal and isonitrile is non-stereoselective. [on SciFinder (R)
Multicomponent domino process to oxa-bridged polyheterocycles and pyrrolopyridines, structural diversity derived from work-up procedure
Novel three-component domino processes to polyheterocycles are developed. Reaction of an allylamine, an aldehyde and an alpha -isocyanoacetamide in methanol at room temp. provides an efficient access to an oxa-bridged tricycle as a single diastereoisomer. In this one-pot process, one C-N, one C-O and three C-C bonds are formed with concomitant creation of five asym. centers. While isolable, the oxa-bridged tricycles can be cleanly in-situ fragmented to pyrrolopyridines under acidic conditions (trifluoroacetic acid, -78 DegC), providing thus an unusual work-up derived structural diversity. The operational simplicity and excellent chem. yield make these novel heterocycle syntheses valuable in diversity-oriented high throughput synthesis. [on SciFinder (R)

Palladium-Catalyzed Multicomponent Synthesis of Fully Substituted Alkylidene Furanones
In the presence of a catalytic amount of Pd(OAc)(2) and XantPhos, the three-component reaction of ynones, imines, and aryl iodides affords fully substituted alkylidene-furan-3(2H)-ones via a sequence of the Mannich reaction followed by chemo- and regioselective oxypalladation and reductive elimination. One carbon-oxygen and two carbon-carbon bonds are generated to afford the heterocycle in good to excellent yield
