    Maximising the number of cycles in graphs with forbidden subgraphs

    Fix k≥2k \ge 2 and let HH be a graph with χ(H)=k+1\chi(H) = k+1 containing a critical edge. We show that for sufficiently large n,n, the unique nn-vertex HH-free graph containing the maximum number of cycles is Tk(n)T_k(n). This resolves both a question and a conjecture of Arman, Gunderson and Tsaturian \cite{Gund1}

    Sensitivity to radiation-induced chromosome damage may be a marker of genetic predisposition in young head and neck cancer patients

    We previously showed that levels of chromosome damage induced by ionizing radiation were, on average, higher in G 2 and G 0 lymphocytes of breast cancer patients than of normal healthy controls, but that there was no correlation between the results in the two assays. We proposed that enhanced sensitivity to G 2 or G 0 irradiation was a marker of low-penetrance predisposition to breast cancer, and have recently demonstrated heritability of sensitivity in families of breast cancer cases. We have now applied these assays to patients with head and neck cancers, for whom there is epidemiological evidence of inherited predisposition in addition to environmental causes. The mean frequency of radiation-induced G 2 aberrations was higher in the 42 patients than in 27 normal controls, but not significantly so. However, cases less than 45 years old were significantly more sensitive than normals of the same age range (P = 0.046), whereas there was no difference between patients and normals of less than 45 years. Also, there was an inverse correlation between G 2 sensitivity and age for patients but not for normals. Radiation-induced micronuclei in G 0 cells were more frequent in 49 patients than in 31 normals (P = 0.056) but, as with the G 2 assay, the greatest difference was seen between early-onset patients and young normals. Again there was an inverse correlation with age for patients but not for normals. Six patients with enhanced toxicity to radiotherapy were G 2 tested and four other such patients were G 0 tested; levels of chromosome damage were not significantly greater than in patients with normal reactions. Both assays were used on 64 individuals (39 patients, 25 normals) and there was no significant correlation between the results. We suggest that a proportion of early-onset head and neck cancer patients are genetically predisposed and that each of the two assays detects a different subset of these cases. © 2001 Cancer Research Campaign http://www.bjcancer.co

    Resolution of Nodular Fasciitis in the Upper Arm

    AbstractNodular fasciitis is a benign fibroblastic lesion that was historically misdiagnosed as a malignant neoplasm. Patients present with pain and swelling of relatively brief duration. The clinical presentation is suggestive of an aggressive lesion, usually occurring in muscle fascia. Histologic features can cause it to be mistaken for sarcoma. After the diagnosis is established histologically, observation is the suggested treatment. We present the case of a patient who had a large soft-tissue tumor in the upper arm with a clinical picture indicative of sarcoma, which ultimately was diagnosed as nodular fasciitis. The patient was treated with anti-inflammatory agents and observation. Within 7 months, the mass almost completely resolved, as documented by magnetic resonance imaging

    Radiation-induced micronucleus induction in lymphocytes identifies a high frequency of radiosensitive cases among breast cancer patients: a test for predisposition?

    Enhanced sensitivity to the chromosome-damaging effects of ionizing radiation is a feature of many cancer-predisposing conditions. We previously showed that 42% of an unselected series of breast cancer patients and 9% of healthy control subjects showed elevated chromosomal radiosensitivity of lymphocytes irradiated in the G2 phase of the cell cycle. We suggested that, in addition to the highly penetrant genes BRCA1 and BRCA2, which confer a very high risk of breast cancer and are carried by about 5% of all breast cancer patients, there are also low-penetrance predisposing genes carried by a much higher proportion of breast cancer patients, a view supported by recent epidemiological studies. Ideally, testing for the presence of these putative genes should involve the use of simpler methods than the G2 assay, which requires metaphase analysis of chromosome damage. Here we report on the use of a simple, rapid micronucleus assay in G0 lymphocytes exposed to high dose rate (HDR) or low dose rate gamma-irradiation, with delayed mitogenic stimulation. Good assay reproducibility was obtained, particularly with the HDR protocol, which identified 31% (12 out of 39) of breast cancer patients compared with 5% (2 out of 42) of healthy controls as having elevated radiation sensitivity. In the long term, such cytogenetic assays may have the potential for selecting women for intensive screening for breast cancer

    Long-term Survival Outcomes for Men Who Provided Ejaculate Specimens for Prostate Cancer Research: Implications for Patient Management

    Background: Determining whether men diagnosed with early prostate cancer (PCa) will live long enough to benefit from interventions with curative intent is difficult. Although validated instruments for predicting patient survival are available, these do not have clinical utility so are not used routinely in practice. Objective: To test the hypothesis that volunteers who provided ejaculate specimens had a high survival rate at 10 and 15 yr and beyond. Design, setting, and participants: A total of 290 patients investigated because of high serum prostate-specific antigen donated ejaculate specimens for research between January 1992 and May 2003. The median age at the time of ejaculation was 63.5 yr. 153 of the donors were diagnosed with PCa and followed up to December 31, 2013. Outcome measurements and statistical analysis: Survival outcomes were compared with those for the whole population, as indicated by life expectancy tables up to 20 yr. Results and limitations: Men in the PCa group had life expectancies comparable with values listed in life expectancy tables for the whole population. Overall, PCa-specific and relative survival were significantly better for men in the non-PCa and PCa groups in comparison with men diagnosed with PCa in Queensland during the same period. Relative survival for those aged 20-49, 50-64, and ≥65 yr was >100% for ejaculate donors and 81.5%, 82.7%, and 65.2%, respectively, for the Queensland Cancer Registry reference at 10 yr. These findings for this highly selected patient cohort support the hypothesis that an ability to provide an ejaculate specimen is associated with a high likelihood of surviving 10-20 yr after donation, whether or not PCa was detected. Conclusion: Life expectancy tables may serve as a quick and simple life expectancy indicator for biopsy patients who donate ejaculate.
