    Innate Immunity in Lobsters: Partial Purification and Characterization of a Panulirus cygnus Anti-A Lectin

    A lectin detected in haemolymph from the Australian spiny lobster Panulirus cygnus agglutinated human ABO Group A cells to a higher titre than Group O or B. The lectin also agglutinated rat and sheep erythrocytes, with reactivity with rat erythrocytes strongly enhanced by treatment with the proteolytic enzyme papain, an observation consistent with reactivity via a glycolipid. The lectin, purified by affinity chromatography on fixed rat-erythrocyte stroma, was inhibited equally by N-acetylglucosamine and N-acetylgalactosamine. Comparison of data from gel filtration of haemolymph (behaving as a 1,800,000 Da macromolecule), and polyacrylamide gel electrophoresis of purified lectin (a single 67,000 Da band), suggested that in haemolymph the lecin was a multimer. The purified anti-A lectin autoprecipitated unless the storage solution contained chaotropic inhibitors (125 mmol/L sucrose: 500 mmol/L urea). The properties of this anti-A lectin and other similar lectins are consistent with a role in innate immunity in these invertebrates

    FAN1 controls mismatch repair complex assembly via MLH1 retention to stabilize CAG repeat expansion in Huntington's disease.

    CAG repeat expansion in the HTT gene drives Huntington's disease (HD) pathogenesis and is modulated by DNA damage repair pathways. In this context, the interaction between FAN1, a DNA-structure-specific nuclease, and MLH1, member of the DNA mismatch repair pathway (MMR), is not defined. Here, we identify a highly conserved SPYF motif at the N terminus of FAN1 that binds to MLH1. Our data support a model where FAN1 has two distinct functions to stabilize CAG repeats. On one hand, it binds MLH1 to restrict its recruitment by MSH3, thus inhibiting the assembly of a functional MMR complex that would otherwise promote CAG repeat expansion. On the other hand, it promotes accurate repair via its nuclease activity. These data highlight a potential avenue for HD therapeutics in attenuating somatic expansion

    Comparative Absorption and Emission Abundance Analyses of Nebulae: Ion Emission Densities for IC 418

    Recent analyses of nebular spectra have resulted in discrepant abundances from CNO forbidden and recombination lines. We consider independent methods of determining ion abundances for emission nebulae, comparing ion emission measures with column densities derived from resonance absorption lines viewed against the central star continuum. Separate analyses of the nebular emission lines and the stellar UV absorption lines yield independent abundances for ions, and their ratio can be expressed in terms of a parameter n_e_{em}, the ``emission density'' for each ion. Adequate data for this technique are still scarce, but separate analyses of spectra of the planetary nebula and central star of IC 418 do show discrepant abundances for several ions, especially Fe II. The discrepancies are probably due to the presence of absorbing gas which does not emit and/or to uncertain atomic data and excitation processes, and they demonstrate the importance of applying the technique of combining emission- and absorption-line data in deriving abundances for nebulae.Comment: 25 pages, 3 figures, accepted for publication in PAS

    Albert Pierrepoint and the cultural persona of the twentieth-century hangman

    Albert Pierrepoint was Britain’s most famous 20th-century hangman. This article utilises diverse sources in order to chart his public representation, or cultural persona, as hangman from his rise to prominence in the mid-1940s to his portrayal in the biopic Pierrepoint(2005). It argues that Pierrepoint exercised agency in shaping this persona through publishing his autobiography and engagement with the media, although there were also representations that he did not influence. In particular, it explores three iterations of his cultural persona – the Professional Hangman, the Reformed Hangman and the Haunted Hangman. Each of these built on and reworked historical antecedents and also communicated wider understandings and contested meanings in relation to capital punishment. As a hangman who remained in the public eye after the death penalty in Britain was abolished, Pierrepoint was an important, authentic link to the practice of execution and a symbolic figure in debates over reintroduction. In the 21st century, he was portrayed as a victim of the ‘secondary trauma’ of the death penalty, which resonated with worldwide campaigns for abolition

    Energy Resolution Performance of the CMS Electromagnetic Calorimeter

    The energy resolution performance of the CMS lead tungstate crystal electromagnetic calorimeter is presented. Measurements were made with an electron beam using a fully equipped supermodule of the calorimeter barrel. Results are given both for electrons incident on the centre of crystals and for electrons distributed uniformly over the calorimeter surface. The electron energy is reconstructed in matrices of 3 times 3 or 5 times 5 crystals centred on the crystal containing the maximum energy. Corrections for variations in the shower containment are applied in the case of uniform incidence. The resolution measured is consistent with the design goals

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    Eavesdropping on community members has immediate and clear benefits. However, little is known regarding its importance for the organisation of cross-taxa community structure. Furthermore, the possibility that eavesdropping could allow species to coexist with a predator and access risky foraging habitat, thereby expanding their realised niche, has been little considered. Kalahari Tree Skinks (Trachylepis spilogaster) associate with Sociable weaver (Philetairus socius) colonies, as do African pygmy falcons (Polihierax semitorquatus), a predator of skinks and weavers. We undertook observational and experimental tests to determine if skinks eavesdrop on Sociable weavers to mitigate any increase in predation threat that associating with weaver colonies may bring. Observations reveal that skinks use information from weavers to determine when predators are nearby; skinks were more active, more likely to forage in riskier habitats and initiated flight from predators earlier in the presence of weavers, compared to when weavers were absent. Playback of weaver alarm calls caused skinks to increase vigilance and flee, confirming that skinks eavesdrop on weavers. Furthermore, skinks at Sociable weaver colonies were more likely to flee than skinks at non-colony trees, suggesting that learning is mechanistically important for eavesdropping behaviour. Overall, it appears that eavesdropping allows skinks at colony trees to gain an early warning signal of potential predators, expand their realised niche and join communities, whose predators may otherwise exclude them.<br
