    Detection Of Encoding Gene Extended Spectrum Beta Lactamase On Escherichia Coli Isolated From Broiler Chicken Meat In Traditional Market Surabaya

    This study aims to isolate, to identify, and to seek out fragments of encoding gene Extended Spectrum β-Lactamase on Escherichia coli isolated from swab surface of broiler chicken meat in a number of traditional markets in Surabaya. The result shows that 31 out of 50 samples positively contain Escherichia coli, shown through EMBA isolation media and identified using indole test. Sensitivity test shows that 100% of the isolates are resistant to Ampicilin, 48.4% are resistant to Cephazoline, 13% are resistant to Ceftazidime, 9.6% are resistant to Cefotaxime, 6.4% are resistant to Ceftriaxone and 87.2% are resistant to Tetracycline. 8 out of 8 (100%) samples of E. coli resistant show the presence of band towards blaTEM gene of 861basepair (bp) Key Words: Extended Spectrum β-Lactamase,Escherichia coli,broiler chicken meat,blaTEM gene,Antimicrobial Resistance

    Antimicrobial Resistance Setection Of Citrobacterfreundii Isolated From Rectal Swab In east Java

    The aim of this study was to detect blaTEMgene which the agent of antibiotic resistance in Citrobacterfreundii isolated from rectal swab in East Java. A total of 275 rectal swab samples were collected from 12 district of East Java. Then, samples were isolated with selective media and biochemical tests. Antibiotic Resistance test was using the disc diffusion against amoxycillin, amoxycillin-clavulanic acid, ampicillin-sulbactam, cefotaxime, ceftazidime, sulfametaxole-trimethoprim, and tetracycline and then detection of bla TEM gene used PCR. The number of Citrobacterfreundii was 7 (2,55 %). Antimicrobial resistance showed 2 broilers and 1 dairy cattle from Surabaya resistant to ampicillin-sulbactam and tetracycline, 4 milkfish from Sidoarjo resistant to amoxicillin and ampicillin-sulbactam.     This study aims to phenotypically identify and confirm the presence of ESBL-producing Escherichia coli in the swab surface samples of beef using VITEK-2 method. Swab samples were taken from five wet markets: Pucang market, Wonokromo market, Keputran market, Pabean market, and Manukan market; 10 swab samples of beef were collected from each market. Then, isolation and identification in terms of bacteria using selective media and biochemical test were conducted. Resistance testing using disc diffusion method was performed on 6 types of antibiotics: Ampicillin, Cefazolin, Ceftriaxone, Cefotaxime, Ceftazidime, and Tetracyclin. Positive isolates resistant to ≥ 2 types of β-Lactam antibiotics using disc diffusion method then were tested using VITEK-2 method. Positive samples containing Escherichia coli are found in 29 samples out of 50 swab samples. Of the 29 Escherichia coli isolates, 17 isolates are found resistant to the disc diffusion method. After testing those 17 isolates using VITEK-2, 5% (1/17) of ESBL-producing Escherichia coli are obtained. Besides, this study also reveals that 35% (6/17) of the Escherichia coli are positive multidrug resistance. Those E.coli are found to be resistant towards Amoxycillin of 35% (6/17), Gentamycin of 5% (1/17), Ciprofloxacin of 35% (6/17), Trimethophrim- Sulfamethoxazole of 35% (6/17), and Ampicillin-sulbactam of 5% (1/17)

    This study aimed to assess the immunostimulatory potential of Spirulina platensis extract tohistopathological change of gouramy fish kidney because Aeromonas hydrophila infection. The research method used five treatments and four replications. Twenty gouramy fish with average weight 20-30 grams were randomly divided into five treatments, including P0-, P0+, P1, P2 and P3. P0- was the group of fish without treatment, P0+was the group of fish infected by Aeomonas hydrophila, P1 was the group of fish dipping withSpirulina platensis extract at 200 mg L-1, P2 was the groupof fish dipping with Spirulina platensis extract at 400mg L-1, and P3 was the group of fish dipping with Spirulina platensis extract at 600mg L-1 and then after 24 hours infected with 106 CFU ml- 1Aeromonas hydrophila. Data were collected on the fourth day after infection. The results of Kruskall Wallis test indicates that there are significant difference (p<0.05) among the treatments, the further test used Mann Whitney can be seen there are no effect of glomerular and tubular kidney necrosis of gouramy fish in all treatment P1, P2 and P3. The conclusion isthe immersed with Spirulina platensis extract and challenged with Aeromonas hydrophila can decrease the damage of the kidney tubular, glomerular and hemopoitic tissue of gouramy fish
