    Costruzione di un clone molecolare di un isolato del virus dell'immunodeficienza felina e sua caratterizzazione in vitro

    Questa tesi si è incentrata nella costruzione di un clone molecolare dell’isolato locale FIV M2 di sottotipo B e nella sua caratterizzazione in vitro. Il DNA provirale è stato estratto da diversi tessuti linfoidi prelevati da un gatto infettato da FIV M2 e deceduto per complicazioni respiratorie. Sono state sviluppate idonee strategie di amplificazione e clonaggio dell’intero genoma provirale. Per analizzare la capacità replicativa in vitro del clone prodotto, quest’ultimo è stato trasfettato sulla linea cellulare di fibroblasti felini Crandell. Il virus rilasciato nel surnatante è stato raccolto ed usato per infettare la linea MBM di linfoblasti felini. Il rapido e costante aumento della p25 nel sovranatante ha suggerito che il clone molecolare prodotto è replicazione competente.     Further Development of Testing Procedures for High Temperature Surface Tension Measurements

    A new testing procedure combining in one test two methods for surface tension measurements of liquid metals [the pendant drop (PD) and the sessile drop] and carried out in the same device is discussed. The attention is focused on methodological aspects of the PD method due to the novelty of its application for high temperature metallic systems. It has been claimed that under the conditions applied in the present study, this method can be considered as a quasi-containerless one. Surface tension measurements of pure Cu, Ni, Al, and Fe performed using the new procedure are described. To confirm the validity of this procedure, the experimental results are discussed in the framework of the available literature data, particularly those obtained by the containerless methods. © 2013 The Author(s)

    The effective oxidation pressure of indium-oxygen system

    A theoretical model on oxygen transport at the surface of liquid metals has been validated by dynamic surface tension measurements performed on liquid Indium as test metal. The oxygen contamination conditions have been obtained at different oxygen partial pressures under both low total pressure (Knudsen regime) and inert atmospheric pressure (Fick regime) conditions, confirming that an oxide removal regime occurs under an oxygen partial pressure much higher than the equilibrium one (the "Effective Oxidation Pressure"). Experimental results are reported which give a further insight on the relative importance of the various processes due to the oxygen mass transport between the liquid metal and the gas phase. The critical aspects involved in surface tension measurements of liquid metals, related to the problem of liquid metal-oxygen interactions, are also underlined

    The introduction of immunotherapeutic agents has provided renewed hope for Chronic lymphocytic leukemia fludarabine-refractory patients. Several clinical trials have shown that alemtuzumab is a more effective option compared to combination chemotherapy for treatment of patients who have relapsed or who are refractory to fludarabine, including those with poor prognostic factors. Although there are significant potential toxicities associated with alemtuzumab, such as infusional reactions and the risk of cytomegalovirus (CMV) reactivation, most are manageable. Pre-treatment anti-pyretics and anti-histamines are recommended to prevent or mitigate the acute infusional reactions associated with intravenous infusion. Recent use of alemtuzumab via the subcutaneous route has been shown to be well tolerated and has yielded similar response rates to the infusional method of administration. Prophylaxis with thrimethoprim/sulphamethoxazole (TMP/SMZ) as well as valacyclovir or a similar anti-viral can prevent many of the opportunistic infections seen in early trials. Reactivation of CMV infection can be effectively managed with monitoring and early treatment. Chemo-immunotherapy combination with alemtuzumab has been tested and demonstrated unprecedented clinical results in relapsed and refractory patients. The use of this agent earlier in the algorithm of patients with these characteristics should be considered. Future areas of research will include the use of alemtuzumab in combination with other monoclonal antibodies and other targeted therapies

    The spectrum of use of rituximab in chronic lymphocytic leukemia

    The monoclonal chimeric anti-CD20 antibody, rituximab, has considerably improved therapeutic outcome in B-cell chronic lymphocytic leukemia. Rituximab has limited clinical activity when used as a single agent. The combination of the monoclonal antibody with fludarabine-based regimens clearly demonstrated, in Phase II and randomized trials, an increase in clinical efficacy in previously untreated and pretreated patients. Furthermore the addition of rituximab enabled the eradication of minimal residual disease, which is correlated with the prognosis in a high proportion of patients. Although the combination of rituximab with fludarabine-based regimens increased myelosuppression and immunosuppression, incidence of infections did not increase. The benefit of adding rituximab to other purine analogs or other chemotherapeutic combination regimens has also been explored. Moreover there could be a role for achieving better quality of responses with the combination of different monoclonal antibodies, considering that they target different antigens and exert different mechanism of action. Although the role of rituximab as maintenance therapy in low grade non-Hodgkin’s lymphomas has been determined, the benefit and optimal schedule in chronic lymphocytic leukemia are still under investigation.     Dolphins Stranded along the Tuscan Coastline (Central Italy) of the “Pelagos Sanctuary”: A Parasitological Investigation

    Parasite monitoring is considered a necessary step for cetacean management and conservation. Between February 2013 and July 2015, 26 dolphins (15 Stenella coeruleoalba, 10 Tursiops truncatus, and one Grampus griseus) stranded along the Tuscan coastline of the protected marine area “Pelagos Sanctuary”, were examined. Organs, tissues, and faecal and blood samples taken from all animals were analysed by parasitological, immunological, and molecular techniques. Twenty-one out of 26 dolphins (80.77%) tested positive for at least one parasite species, and 13/15 (86.7%) S. coeruleoalba, 7/10 (70%) T. truncatus, and the single G. griseus were found positive. Identified parasites included the nematodes Skrjabinalius guevarai (7.69%, 2/26), Halocercus lagenorhynchi (3.85%, 1/26), Halocercus delphini (7.69%, 2/26), Stenurus ovatus (7.69%, 2/26), Crassicauda spp. (7.69%, 2/26); the trematodes Pholeter gastrophilus (26.92%, 7/26), Campula palliata (3.85%, 1/26); the cestodes Phyllobothrium delphini (42.31%, 11/26), Monorygma grimaldii (23.08%, 6/26), Tetrabothrium forsteri (7.69%, 2/26), Strobilocephalus triangularis (7.69%, 2/26), and the acanthocephalan Bolbosoma vasculosum (7.69%, 2/26). Moreover, 6/26 (23%) animals scored positive to Toxoplasma gondii at serology, but PCR confirmed the infection (T. gondii Type II genotype) in a single animal.
