    The Willmore Flow of Tori of Revolution

    We study long-time existence and asymptotic behavior for the L2-gradient flow of the Willmore energy, under the condition that the initial datum is a torus of revolution. We show that if an initial datum has Willmore energy below 8π8\pi then the solution of the Willmore flow converges for tt \rightarrow \infty to the Clifford Torus, possibly rescaled and translated. The energy threshold of 8π8\pi turns out to be optimal for such a convergence result. We give an application to the conformally constrained Willmore minimization problem.Comment: 43 page

    How to Predict the Impact of Methylphenidate on Cardiovascular Risk in Children with Attention Deficit Disorder: Methylphenidate Improves Autonomic Dysfunction in Children with ADHD

    Background. Although stimulants have long been touted as treatments for attention deficit disorder with or without hyperactivity (ADHD), in recent years, increasing concerns have been raised about the cardiovascular safety of these medications. We aimed to prove if measurements of autonomic function with time domain analysis of heart rate variability (HRV) in 24-hour Holter ECG are useful to predict the risk of sudden cardiac death in ADHD children and adolescents. Methods. We analysed HRV obtained from children with the diagnosis of ADHD prior to (N = 12) or during medical therapy (N = 19) with methylphenidate (MPH), aged 10.8 ± 2.0 years (mean ± SD), who were referred to our outpatient Paediatric Cardiology Clinic to rule out heart defect. As a control group, we compared the HRV data of 19 age-matched healthy children without heart defect. Results. Average HRV parameters from 24-hour ECG in the ADHD children prior to MPH showed significant lower values compared to healthy children with respect to rMSSD (26 ± 4 ms versus 44 ± 10 ms, P ≤ 0.0001) and pNN50 (6.5 ± 2.7% versus 21.5 ± 9.0%, P ≤ 0.0001). These values improved in MPH-treated children with ADHD (RMSSD: 36 ± 8 ms; pNN50: 14.2 ± 6.9%). Conclusion. Children who suffer from ADHD show significant changes in HRV that predominantly reflects diminished vagal tone, a well-known risk factor of sudden cardiac death in adults.     Impact of reduced tillage on greenhouse gas emissions and soil carbon stocks in an organic grass-clover ley - winter wheat cropping sequence

    Organic reduced tillage aims to combine the environmental benefits of organic farming and conservation tillage to increase sustainability and soil quality. In temperate climates, there is currently no knowledge about its impact on greenhouse gas emissions and only little information about soil organic carbon (SOC) stocks in these management systems. We therefore monitored nitrous oxide (N2O) and methane (CH4) fluxes besides SOC stocks for two years in a grass-clover ley – winter wheat – cover crop sequence. The monitoring was undertaken in an organically managed long-term tillage trial on a clay rich soil in Switzerland. Reduced tillage (RT) was compared with ploughing (conventional tillage, CT) in interaction with two fertilisation systems, cattle slurry alone (SL) versus cattle manure compost and slurry (MC). Median N2O and CH4 flux rates were 13 μg N2O-N m−2 h−1 and −2 μg CH4C m−2 h−1, respectively, with no treatment effects. N2O fluxes correlated positively with nitrate contents, soil temperature, water filled pore space and dissolved organic carbon and negatively with ammonium contents in soil. Pulse emissions after tillage operations and slurry application dominated cumulative gas emissions. N2O emissions after tillage operations correlated with SOC contents and collinearly to microbial biomass. There was no tillage system impact on cumulative N2O emissions in the grass-clover (0.8–0.9 kg N2O-N ha−1, 369 days) and winter wheat (2.1–3.0 kg N2O-N ha−1, 296 days) cropping seasons, with a tendency towards higher emissions in MC than SL in winter wheat. Including a tillage induced peak after wheat harvest, a full two year data set showed increased cumulative N2O emissions in RT than CT and in MC than SL. There was no clear treatment influence on cumulative CH4 uptake. Topsoil SOC accumulation (0–0.1 m) was still ongoing. SOC stocks were more stratified in RT than CT and in MC than SL. Total SOC stocks (0–0.5 m) were higher in RT than CT in SL and similar in MC. Maximum relative SOC stock difference accounted for +8.1 Mg C ha−1 in RT-MC compared to CT-SL after 13 years which dominated over the relative increase in greenhouse gas emissions. Under these site conditions, organic reduced tillage and manure compost application seems to be a viable greenhouse gas mitigation strategy as long as SOC is sequestered

    Rapid Decline of Serum Proprotein Convertase Subtilisin/Kexin 9 (PCSK9) in Non-Cirrhotic Patients with Chronic Hepatitis C Infection Receiving Direct-Acting Antiviral Therapy.

    Direct-acting antivirals (DAAs) efficiently eradicate the hepatitis C virus (HCV). Low-density lipoprotein (LDL) levels increase rapidly upon DAA treatment. Proprotein convertase subtilisin/kexin 9 (PCSK9) induces degradation of the hepatic LDL receptor and thereby elevates serum LDL. The aim of this study was to determine serum PCSK9 concentrations during and after DAA therapy to identify associations with LDL levels. Serum PCSK9 was increased in 82 chronic HCV-infected patients compared to 55 patients not infected with HCV. Serum PCSK9 was low in HCV patients with liver cirrhosis, but patients with HCV-induced liver cirrhosis still exhibited higher serum PCSK9 than patients with non-viral liver cirrhosis. Serum PCSK9 correlated with measures of liver injury and inflammation in cirrhotic HCV patients. In patients without liver cirrhosis, a positive association of serum PCSK9 with viral load existed. Serum PCSK9 was not different between viral genotypes. Serum PCSK9 did not correlate with LDL levels in HCV patients irrespective of cirrhotic status. Serum PCSK9 was reduced, and LDL was increased at four weeks after DAA therapy start in non-cirrhotic HCV patients. Serum PCSK9 and LDL did not change upon DAA treatment in the cirrhotic group. The rapid decline of PCSK9 after the start of DAA therapy in conjunction with raised LDL levels in non-cirrhotic HCV patients shows that these changes are not functionally related

    Prevention of M2 polarization and temporal limitation of differentiation in monocytes by extracellular ATP

    BACKGROUND Elevated levels of extracellular adenosine triphosphate (ATP) modulate immunologic pathways and are considered to be a danger signal in inflammation, lung fibrosis and cancer. Macrophages can be classified into two main types: M1 macrophages are classically activated, pro-inflammatory macrophages, whereas M2 macrophages are alternatively activated, pro-fibrotic macrophages. In this study, we examined the effect of ATP on differentiation of native human monocytes into these macrophage subtypes. We characterized M1 and M2 like macrophages by their release of Interleukin-1beta (IL-1β) and Chemokine (C-C motif) ligand 18 (CCL18), respectively. RESULTS Monocytes were stimulated with ATP or the P2X7 receptor agonist Benzoylbenzoyl-ATP (Bz-ATP), and the production of various cytokines was analyzed, with a particular focus on CCL18 and IL-1β, along with the expression of different purinergic receptors. Over a 72 h period of cell culture, monocytes spontaneously differentiated to M2 like macrophages, as indicated by an increased release of CCL18. Immediate stimulation of monocytes with ATP resulted in a dose-dependent reduction in CCL18 release, but had no effect on the concentration of IL-1β. In contrast, delayed stimulation with ATP had no effect on either CCL18 or IL-1β release. Similar results were observed in a model of inflammation using lipopolysaccharide-stimulated human monocytes. Stimulation with the P2X7 receptor agonist Bz-ATP mimicked the effect of ATP on M2-macrophage differentiation, indicating that P2X7 is involved in ATP-induced inhibition of CCL18 release. Indeed, P2X7 was downregulated during spontaneous M2 differentiation, which may partially explain the ineffectiveness of late ATP stimulation of monocytes. However, pre-incubation of monocytes with PPADS, Suramin (unselective P2X- and P2Y-receptor blockers) and KN62 (P2X7-antagonist) failed to reverse the reduction of CCL18 by ATP. CONCLUSIONS ATP prevents spontaneous differentiation of monocytes into M2-like macrophages in a dose- and time-dependent manner. These effects were not mediated by P2X and P2Y receptors

    Genetic variation of introduced red oak (Quercus rubra) stands in Germany compared to North American populations

    Although Northern red oak (Quercus rubra L.) is the most important introduced deciduous tree species in Germany, only little is known about its genetic variation. For the frst time, we describe patterns of neutral and potentially adaptive nuclear genetic variation in Northern red oak stands across Germany. For this purpose, 792 trees were genotyped including 611 trees from 12 stands in Germany of unknown origin and 181 trees from four populations within the natural distribution area in North America. Our marker set included 12 potentially adaptive (expressed sequence tag-derived simple sequence repeat=EST SSR) and 8 putatively selectively neutral nuclear microsatellite (nSSR) markers. Our results showed that German stands retain comparatively high levels of genetic variation at both EST-SSRs and nSSRs, but are more similar to each other than to North American populations. These fndings are in agreement with earlier chloroplast DNA analyses which suggested that German populations originated from a limited geographic area in North America. The comparison between potentially adaptive and neutral microsatellite markers did not reveal diferences in the analyzed diversity and diferentiation measures for most markers. However, locus FIR013 was identifed as a potential outlier locus. Due to the absence of signatures of selection in German stands, we suggest that introduced populations were established with material from provenances that were adapted to environmental conditions similar to those in Germany. However, we analyzed only a limited number of loci which are unlikely to be representative of adaptive genetic diferences among German stands. Our results suggest that the apparent introduction from a limited geographic range in North America may go along with a reduced adaptive potential

    The screening power of methylenetetrahydrofolate reductase C677T polymorphism versus plasma homocysteine concentration in patients with stenosis of the internal carotid artery

    BACKGROUND: Hyperhomocysteinemia is an important and independent risk factor for vascular disease. About 35% of patients with stroke and 47% of patients with peripheral arterial disease have elevated plasma homocysteine (HCY) concentrations. The relationship between plasma HCY and the methylentetrahydrofolate reductase (MTHFR) C677T polymorphism is still unclear, especially in regard to screening/diagnostic power. METHODS: This case-control study was performed on 96 patients, who underwent surgery due to asymptomatic or symptomatic high grade stenosis of the internal carotid artery (ICA), and 96 healthy age and sex-matched, controls. Plasma HCY concentration was determined using a commercial kit for fully automated analysis (AxSYM, Abbott). The C677T polymorphism of the MTHFR-gene was assessed by PCR. RESULTS: The mean plasma HCY concentration was significantly higher in the group with stenosis of ICA compared to the controls, 12.43 ± 6.96 μM and 10.16 ± 3.16 μM, respectively, (p < 0.05). An HCY plasma concentration of 1.5 SD above the mean value of the control group, was defined as cut-off for a pathological versus physiological plasma concentration. The sensitivity and specificity of HCY was 0.27 and 0.94, respectively. The positive predictive value was 0.82. There was no significant difference in the frequency of the MTHFR 677 CT and TT genotype between patients and controls (47% vs. 47% and 8.3% vs. 11.4%, respectively). Carriers of the T-allele (CT and TT genotypes) have significantly higher plasma HCY concentrations than CC patients, 14.1 ± 7.6 μM and 10.29 ± 5.2 μM, respectively, p < 0.05. Sensitivity and specificity of the MTHFR C677T polymorphism (T-allele) were 0.56 and 0.40, respectively. The positive predictive value was 0.48. There was no significant difference in plasma HCY or genotype frequency of the MTHFR C677T polymorphism between asymptomatic and symptomatic patients. CONCLUSION: Our study shows that in a population with a given pretest disease probability of 50%, the determination of plasma HCY concentration, with a positive predictive value of 0.82, is more suitable for screening of patients at risk than analysis of the MTHFR C677T polymorphism

Es wird aufgezeigt, wie ein deutsch-chinesisches Lernfabriknetzwerk durch Zusammenarbeit der TU Darmstadt und Festo Didactic in Deutschland sowie CASICloud-Tech und dem ITEI Institut in China aufgebaut wurde. Auf Basis dieses Lernfabriknetzwerkes wurden neue Schulungsinhalte in den Bereichen Werkerassistenzsysteme, Bauteilrückverfolgbarkeit und Cloud Manufacturing für die Bedarfe chinesischer Unternehmen erstellt und die Vernetzung von internationalen Wertschöpfungsnetzwerken durch Anbindung an eine gemeinsame Cloud gestärkt
