    The Effect of NAG–thiazoline on Morphology and Surface Hydrophobicity of Escherichia Coli

    The β-hexosaminidase inhibitor and structural analog of the putative oxazolium reaction intermediate of lytic transglycosylases, N-acetylglucosamine thiazoline (NAG–thiazoline), was synthesized in 46% overall yield and tested as an inhibitor of Escherichia coli growth. NAG–thiazoline, at concentrations up to 1 mg/ml, was not found to affect the viability of E. coli DH5α

    More than ever, human factors engineers and ergonomists need to justify our practice’s value to management. How can we effectively communicate with management? How should we present a Return on Investment (ROI) that leadership will find useful that addresses company profits, cost savings, productivity, first time quality, and turnover? What else does management care about other than ROI? This second panel in a two panel series will specifically highlight case studies in which presenters give examples of situations in which ROI for ergonomics was investigated from a business value. The session will start with four case study lectures followed by a panel discussion led by the moderators. The audience will be encouraged to participate with their own questions and comments

    Outcome following valve surgery in Australia: Development of an enhanced database module

    Background: Valvular heart disease, including rheumatic heart disease (RHD), is an important cause of heart disease globally. Management of advanced disease can include surgery and other interventions to repair or replace affected valves. This article summarises the methodology of a study that will incorporate enhanced data collection systems to provide additional insights into treatment choice and outcome for advanced valvular disease including that due to RHD. Methods: An enhanced data collection system will be developed linking an existing Australian cardiac surgery registry to more detailed baseline co-morbidity, medication, echocardiographic and hospital separation data to identify predictors of morbidity and mortality outcome following valve surgery. Discussion: This project aims to collect and incorporate more detailed information regarding pre and postoperative factors and subsequent morbidity. We will use this to provide additional insights into treatment choice and outcome. © 2017 The Author(s)

    Authenticating Hybrid Cell Lines

    Hybrid (both intra-species and inter-species) cell lines arise through intentional or nonintentional fusion of somatic cells having different origins. Hybrid cell lines can pose a problem for authentication testing to confirm cell line identity, since the results obtained may not conform to the results expected for the two parental cell types. Thus, depending on the identity testing methodology, a hybrid cell may display characteristics of one of the parental cell type or of both. In some instances, the hybrid cell line may display characteristics that are different from those displayed by either parental cell type; these differences may not necessarily indicate cellular cross-contamination. Testing should be performed as soon as possible after an intended fusion has occurred, so that a baseline reference profile is available for later comparison. In this article, we describe the various approaches that have been used for identifying hybrid cell lines and the results that might be expected when using various technologies for this purpose

    Role of Ser216 in the mechanism of action of membrane-bound lytic transglycosylase B: Further evidence for substrate-assisted catalysis

    AbstractLytic transglycosylases cleave the β-(1→4)-glycosidic bond in the bacterial cell wall heteropolymer peptidoglycan between the N-acetylmuramic acid (MurNAc) and N-acetylglucosamine (GlcNAc) residues with the concomitant formation of a 1,6-anhydromuramoyl residue. Based on sequence alignments, Ser216 in Pseudomonas aeruginosa membrane-bound lytic transglycosylase B (MltB) was targeted for replacement with alanine to delineate its role in the enzyme’s mechanism of action. The specific activity of the Ser216→Ala MltB derivative was less than 12% of that for the wild-type enzyme, while its substrate binding affinity remained virtually unaltered. These data are in agreement with a role of Ser216 in orienting the N-acetyl group on MurNAc at the −1 subsite of MltB for its participation in a substrate-assisted mechanism of action
