    The effect of N-acetyl cysteine on H2O2 mediated oxidative stress in Whartonʼs jelly derived mesenchymal stem cells

    Background: Hypoxic stress is a crucial factor for retaining the cell survival in injured tissue. Overcoming this issue is the key for successful cellular regenerative therapy. Therefore the purpose of this study was to investigate whether the in-vitro pretreatment of Whartonʼs Jelly (WJ) derived Mesenchymal stem cells (WJ-MSCs) with an antioxidant, namely N-acetylcysteine (NAC), can improve the efficacy of WJ-MSCs for transplantation purpose.Methods: WJ-MSCs were cultured with or without NAC at different concentrations (0.1mM, 1mM and 10mM). To simulate oxidative stress conditions, cultures were exposed to hydrogen peroxide (H2O2) 100 µM for 1 hour. Cytoprotective effect of NAC was evaluated by determining cell injury, viability, and proliferation. The oxidative stress is assessed by measuring the activity of glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), and malodialdehyde (MDA).Results: Pretreatment of WJ-MSCs with NAC increased their viability and proliferation in concentration-dependent manner. Furthermore, 10 mM NAC significantly reduced the H2O2 induced oxidative stress by enhancing the activity of GSH, SOD, and CAT and reduced the level of MDAConclusion: The study results indicate that NAC may abrogate H2O2 induced oxidative-stress of WJ-MSCs. This study provides basis to explore NAC effect on WJ-MSCs survival without cytotoxicity

    Consistency of local perceptions of climate change in the Kangchenjunga Himalaya landscape

    The Himalaya is experiencing rapid climate change that is likely to significantly impact local ecosystems, biodiversity, agriculture and human well-being. However, the scientific community has been slow to examine the extent and consequences of climate change. Local communities have been coping with environmental change since millennia. Thus they often have considerable knowledge about environmental change and means to cope with its consequences. We (a) examined the perceptions of local communities about climate change and its impacts on ecosystems, biodiversity, agriculture and livelihoods in the Kangchenjunga Himalayas region; (b) analysed the consistency of perceptions across geographical regions, and (c) assessed the conformity between local perceptions and scientific evidence. Our study is based on surveys conducted in 576 households, focus group discussions, key informant surveys and direct observations. The results show that people in the Kangchenjunga Himalayas region have considerable knowledge of climate change and its effects on the weather, ecosystems, biodiversity and agriculture. These perceptions are consistent across the region and conform to scientific findings

    Nanoremediation technologies for sustainable remediation of contaminated environments: Recent advances and challenges

    A major and growing concern within society is the lack of innovative and effective solutions to mitigate the challenge of environmental pollution. Uncontrolled release of pollutants into the environment as a result of urbanisation and industrialisation is a staggering problem of global concern. Although, the eco-toxicity of nanotechnology is still an issue of debate, however, nanoremediation is a promising emerging technology to tackle environmental contamination, especially dealing with recalcitrant contaminants. Nanoremediation represents an innovative approach for safe and sustainable remediation of persistent organic compounds such as pesticides, chlorinated solvents, brominated or halogenated chemicals, perfluoroalkyl and polyfluoroalkyl substances (PFAS), and heavy metals. This comprehensive review article provides a critical outlook on the recent advances and future perspectives of nanoremediation technologies such as photocatalysis, nano-sensing etc., applied for environmental decontamination. Moreover, sustainability assessment of nanoremediation technologies was taken into consideration for tackling legacy contamination with special focus on health and environmental impacts. The review further outlines the ecological implications of nanotechnology and provides consensus recommendations on the use of nanotechnology for a better present and sustainable futur

    Stroke Ready: a multi-level program that combines implementation science and community-based participatory research approaches to increase acute stroke treatment: protocol for a stepped wedge trial

    Abstract Background Post-stroke disability is common, costly, and projected to increase. Acute stroke treatments can substantially reduce post-stroke disability, but few patients take advantage of these cost-effective treatments. Practical, cost-efficient, and sustainable interventions to address underutilized acute stroke treatments are currently lacking. In this context, we present the Stroke Ready project, a stepped wedge design, multi-level intervention that combines implementation science and community-based participatory research approaches to increase acute stroke treatments in the predominately African American community of Flint, Michigan, USA. Methods Guided by the Tailored Implementation of Chronic Disease (TICD) framework, we begin with optimization of acute stroke care in emergency departments, with particular attention given to our safety-net hospital partners. Then, we move to a community-wide, multi-faceted, stroke preparedness intervention, with workshops led by peer educators, over 2 years. Measures of engagement of the safety-net hospital and the feasibility and sustainability of the implementation strategy as well as community intervention reach, dose delivered, and satisfaction will be collected. The primary outcome is acute stroke treatment rates, which includes both intravenous tissue plasminogen activator, and endovascular treatment. The co-secondary outcomes are intravenous tissue plasminogen activator treatment rates and the proportion of stroke patients who arrive by ambulance. Discussion If successful, Stroke Ready will increase acute stroke treatment rates through emergency department and community level interventions. The stepped wedge design and process evaluation will provide insight into how Stroke Ready works and where it might work best. By exploring the relative effectiveness of the emergency department optimization and the community intervention, we will inform hospitals and communities as they determine how best to use their resources to optimize acute stroke care.     A mesh network of MnO nanowires and CNTs reinforced by molecularly imprinted structures for the selective detection of para-nitrophenol

    Advanced material architecture can be used to develop tailor-made interfaces for innovative and selective sensor platforms. An intricate mesh structure of manganese oxide nanowires and carbon nanotubes was synthesized. Further, the mesh was strengthened by a molecularly imprinted network to generate template cavities and impart selective recognition. Termed as MIP@MnO:CNT, this mesh structure was used as the receptor interface for microarray transducers. The unique hybrid composition and morphology enhanced binding performance for detection of para-nitrophenol (P-NP), an important pollutant. The sensor showed exceptional sensitivity towards P-NP monitoring with a limit of detection of 3 nM (S/N = 3). Benefitted from the imprinting strategy, the designed sensor exhibited 85–99% selectivity when compared to other aromatic compounds. Moreover, the designed interface was able to detect P-NP in water samples. As demonstrated in this study, other chemical compositions and morphology of multi-dimensional materials can be crafted for the improved and specific detection of analytes. </p

    Myopericytoma of lip: A rare lesion in an unusual location

    Myopericytoma is a rare benign tumor with perivascular myoid differentiation. It shares histomorpholoic features with the so-called hemangiopericytoma, myofibroma and glomus tumor. We hereby report the case of a 14-year-old boy who presented with a slowly growing, painless, firm mass on upper lip, diagnosed as myopericytoma on the basis of histopathology and immunohistochemistry. To the best of our knowledge, this is only the second such reported case
