    The [Tc(N)(PNP)]2+ metal fragment labeled cholecystokinin-8 (CCK8) peptide for CCK-2 receptors imaging: in vitroand in vivo studies

    The radiolabeling of the natural octapeptide CCK8, derivatized with a cysteine residue (Cys-Gly-CCK8), by using the metal fragment [99mTc(N)(PNP3)]2+ (PNP3 = N,N-bis(dimethoxypropylphosphinoethyl)methoxyethylamine) is reported. The [99mTc(N)(NS-Cys-Gly-CCK8)(PNP3)]+ complex was obtained according to two methods (one-step or two-step procedure) that gave the desired compound in high yield. The complex is stable in aqueous solution and in phosphate buffer. In vitro challenge experiments with an excess of cysteine and glutathione indicate that no transchelation reactions occur, confirming the high thermodynamic stability and kinetic inertness of this compound. Stability studies carried out in human and mouse serum, as well as in mouse liver homogenates, show that the radiolabeled compound remains intact for prolonged incubation at 37 degrees C. Binding properties give Kd (19.0 +/- 4.6 nmol/l) and Bmax (approximately 10(6) sites/cell) values in A431 cells overexpressing the CCK2-R. In vivo evaluation of the compound shows rapid and specific targeting to CCK2-R, a fourfold higher accumulation compared to nonreceptor expressing tumors

    A triple-blinded crossover study to evaluate the short-term safety of sweet manioc starch for the treatment of glycogen storage disease type Ia

    BACKGROUND: Glycogen storage disease type 1a (GSD Ia) is characterized by severe fasting hypoglycemia. The clinical management includes the administration of uncooked cornstarch (UCCS). Although such a diet approach is effective in achieving euglycemia, its impact on the quality of life of patients should be considered. In vitro analyses suggest a longer release of glucose when using sweet manioc starch (SMS). METHODS: We compared the efficacy and safety of the administration of SMS and UCCS during a short-fasting challenge in patients with GSD Ia in a randomized, triple-blind, phase I/II, cross-over study. GSD Ia patients aged ≥ 16 years and treated with UCCS were enrolled. Participants were hospitalized for two consecutive nights, receiving UCCS or SMS in each night. After the administration of the starches, glucose, lactate and insulin levels were measured in 1-h interval throughout the hospitalization period. The procedures were interrupted after 10 h of fasting or in a hypoglycemic episode ( 25 kg/m(2)) participated in the study. The average fasting period was 8.2 ± 2.0 h for SMS and 7.7 ± 2.3 h for UCCS (p = 0.04). SMS maintained euglycemia for a greater period over UCCS. Increased lactate concentrations were detected even in absence of hypoglycemia, not being influenced by the different starches investigated (p = 0.17). No significant difference was found in total cholesterol, HDL, triglycerides and uric acid levels in both arms. None of the patients showed severe adverse events. CONCLUSIONS: SMS appears to be non-inferior to UCCS in the maintenance of euglycemia, thus emerging as a promising alternative to the treatment of GSD Ia

Two sections were detailed on the theme: a theoretical and an empirical one. The theoretical section proposes, from a literature review, the presentation of a reflection on the complexity of forming psychic human beings, and the importance of the attention offered by the parental figures, mainly in the children‟s first years of life. Therefore, the concepts of paternal and maternal functions were explored, taking into consideration what concerns the subjectivity building, as well as invariable elements in the psychism formation. Psychoanalysis theoretical contributions were applied in an attempt of obtaining a deep comprehension on the theme, highlighting the effects of parental function demands. It is also noteworthy the need that relations between parents and children be ruled by asymmetry, by the exercise of symbolical authority, and by presenting boundaries to establish possibilities of meetings supported by the recognition of otherness. The empirical section, from qualitative research method, investigated what comprehension the parental figures make of their children‟s infraction act, aiming to explore the paternal and maternal functions modalities in their children‟s situation in conflict with the law, as well as the meanings assigned to fatherhood and motherhood. Semi-structured interviews were assigned with four mothers, three fathers, and one stepfather, responsible for adolescents between 15 and 18 years old, who had been involved in Justice hearings. The material obtained in the interviews were analyzed and discussed via Content Analysis. For data interpretation, the psychoanalytical theory was used. Four final categories were identified: From the parental optics: a reading from impasses to implications, Unique nuances of familiar configurations and relations, Uncovering the parental universe, and, at last, Social and institutional spaces: increment factors to parental abandonment. It was verified the existence of parents‟ life histories marked by frailty in the relations, and by difficulty in establishing relational modalities of care with themselves and with others. It was evidenced the parental figures‟ precarious condition of investing emotionally in their children, and, several times, they ignored important issues related to their children‟s lives. It was also possible to verify the premise of symmetrical family configurations, in which there was not the constitution, by the parents, of a needed symbolic authority in the presentation of boundaries to their children. It was possible to observe that the parental figures assigned the infraction act committed by their children to external elements, as well as presented a vision vaguely connected to the real dimension of their children‟s problem. It was also possible to broaden the comprehension on the theme from the consideration of effects coming from precarious social conditions, and the participants‟ complaints regarding some institutions inefficiency. A Se??o Emp?rica, a partir do m?todo qualitativo de pesquisa, investigou a compreens?o que as figuras parentais fazem do ato infracional do filho, buscando explorar as modalidades de fun??es paterna e materna na situa??o de filhos em conflito com a lei, bem como os significados atribu?dos ? paternidade e ? maternidade. Foram realizadas entrevistas semiestruturadas com quatro m?es, tr?s pais e um padrasto, respons?veis por adolescentes entre 15 e 18 anos de idade, que se encontravam envolvidos com audi?ncias no sistema judici?rio. O material obtido nas entrevistas foi analisado e discutido por meio da An?lise de Conte?do. Para interpretar os dados, utilizou-se o referencial te?rico psicanal?tico. Identificaram-se quatro categorias finais: A partir da ?tica parental: uma leitura dos impasses ? implica??o no cuidado, Nuances singulares das configura??es e das rela??es familiares, Descortinando o universo parental e, por ?ltimo, Espa?os sociais e institucionais: fatores de incremento ao desamparo parental. Constatou-se a exist?ncia de hist?rias de vida parentais marcadas pela fragilidade nas rela??es e pela dificuldade em estabelecer modalidades relacionais de cuidado consigo e com o outro. Evidenciou-se a prec?ria condi??o das figuras parentais de investirem afetivamente em seus filhos, sendo que, muitas vezes, desconheciam quest?es importantes referentes as suas vidas. Se p?de verificar ainda o estabelecimento de configura??es familiares sim?tricas, nas quais n?o havia por parte dos pais o estabelecimento de uma autoridade simb?lica necess?ria na apresenta??o de limites aos filhos. Foi poss?vel observar que as figuras parentais atribu?am o ato infracional cometido pelos filhos a elementos externos, bem como apresentavam uma vis?o pouco conectada com a real dimens?o da problem?tica explicitada por seus filhos. Tamb?m foi poss?vel ampliar a compreens?o da tem?tica a partir da considera??o dos efeitos oriundos de prec?rias condi??es sociais e da queixa dos participantes a respeito da inefici?ncia de algumas institui??es.     New Tc-99m radiopharmaceuticals with the ligand (o-hydroxyphenyl)diphenylphosphine have been prepared and their biodistributions evaluated in rats. The monoxo Tc(V) complex [99mTc(0)Cl(PO),J, the Tc(IV) complex [99mTc(OH),(PO),], the Tc(IJI) complex [99mTc(PO),], and the nitrido Tc(V) complex [99”Tc(N)(PO),] have been characterized by TLC and HPLC chromatography, and their chemical structure elucidated by comparison with the corresponding complexes obtained using the P-emitting isotope Tc-99g.     Reactions of the precursor complexes [99gTc(N)Cl2(PPh3)2] and [99gTc(N)Cl4]- with phosphine-thiol ligands (HLn) of the type R2PCH2CH2SH (R ) phenyl, methoxypropyl), R2¢PCH2CH2CH2SH (R¢ ) phenyl, tolyl), and R2¢¢Po- C6H4SH (R¢¢ ) phenyl) afforded the five-coordinated, disubstituted nitrido technetium(V) complexes [99gTc- (N)(Ln)2]. The complexes were characterized by elemental analysis, 1H and 31P NMR spectroscopy, FT IR, and positive FAB MS spectra. Structural characterization of [99gTc(N)(L1)2] (1) [HL1 ) (C6H5)2PCH2CH2SH] and [99gTc(N)(L5)2] (5) [HL5 ) (o-CH3C6H4)2PCH2CH2CH2SH] showed that the bidentate phosphino-thiol ligands are coordinated to the technetium center through the neutral phosphorus atom and the deprotonated thiol sulfur atom. These complexes possess an uncommon trigonal bipyramidal geometry with the two phosphorus atoms occupying the two transaxial positions and the two sulfur atoms on the equatorial plane along with the nitrido nitrogen atom. Compound 1 crystallizes in the monoclinic space group C2/c, a ) 24.84(2) Å, b ) 7.327(6) Å, c ) 31.52(2) Å, â ) 111.06(10)°, and Z ) 8.
