    Forebrain Ischemia Triggers GABAergic System Degeneration in Substantia Nigra at Chronic Stages in Rats

    The long-term consequences of forebrain ischemia include delayed Parkinson's syndrome. This study revealed delayed neurodegeneration in the substantia nigra 8 weeks after 12.5 minutes of global ischemia in rat brain. Following neuronal loss of 30–40% in central and dorsolateral striatum at day 3, neuronal damage in the substantia nigra (SN) was assessed at 4–8 weeks using immunohistochemistry for glutamate decarboxylase 67 (GAD67), vesicular GABA transporter (VGAT), and calretinin (CR). At day 56, the optical density of GAD67-, but not VGAT-, immunoreactivity in substantia nigra pars reticulata (SNR)—significantly decreased. CR-neurons concentrated in substantia nigra pars compacta (SNC) were reduced by 27% from day 3 (n = 5) to day 56 (n = 7, ANOVA, p < .01). Movement coordination was impaired at day 56, as evaluated using beam-walking test (time-to-traverse 5.6 ± 1.2 sec versus 11.8 ± 5.4 sec; sham versus ischemia, p < .05, n = 5, and 7, resp.). Our results demonstrate delayed impairment of the GABAergic system components in SN and associated with movement deficits after global ischemia

    Rotating dust solutions of Einstein's equations with 3-dimensional symmetry groups, Part 3: All Killing fields linearly independent of u^{\alpha} and w^{\alpha}

    This is the third and last part of a series of 3 papers. Using the same method and the same coordinates as in parts 1 and 2, rotating dust solutions of Einstein's equations are investigated that possess 3-dimensional symmetry groups, under the assumption that each of the Killing vectors is linearly independent of velocity uαu^{\alpha} and rotation wαw^{\alpha} at every point of the spacetime region under consideration. The Killing fields are found and the Killing equations are solved for the components of the metric tensor in every case that arises. No progress was made with the Einstein equations in any of the cases, and no previously known solutions were identified. A brief overview of literature on solutions with rotating sources is given.Comment: One missing piece, signaled after eq. (10.7), is added after (10.21).     Uniformly Accelerated Charge in a Quantum Field: From Radiation Reaction to Unruh Effect

    We present a stochastic theory for the nonequilibrium dynamics of charges moving in a quantum scalar field based on the worldline influence functional and the close-time-path (CTP or in-in) coarse-grained effective action method. We summarize (1) the steps leading to a derivation of a modified Abraham-Lorentz-Dirac equation whose solutions describe a causal semiclassical theory free of runaway solutions and without pre-acceleration patholigies, and (2) the transformation to a stochastic effective action which generates Abraham-Lorentz-Dirac-Langevin equations depicting the fluctuations of a particle's worldline around its semiclassical trajectory. We point out the misconceptions in trying to directly relate radiation reaction to vacuum fluctuations, and discuss how, in the framework that we have developed, an array of phenomena, from classical radiation and radiation reaction to the Unruh effect, are interrelated to each other as manifestations at the classical, stochastic and quantum levels. Using this method we give a derivation of the Unruh effect for the spacetime worldline coordinates of an accelerating charge. Our stochastic particle-field model, which was inspired by earlier work in cosmological backreaction, can be used as an analog to the black hole backreaction problem describing the stochastic dynamics of a black hole event horizon.Comment: Invited talk given by BLH at the International Assembly on Relativistic Dynamics (IARD), June 2004, Saas Fee, Switzerland. 19 pages, 1 figur

    Model of LPG Refrigerator: A Literature Review

    This work investigates the result of an experimental study carried out to determine the Coefficient of performance of domestic refrigerator when a propane-butane mixture is liquefied petroleum gas (LPG) which is available and comprises 56.4% butane, 24.4%propane, and 17.2% isobutene. This paper also presented an experimental investigation of COP by the effect of changing capillary tube length, capillary tube inner diameter and capillary coil diameter on the mass flow rate of refrigerant in an adiabatic helical capillary tube. Large amount of electricity supply is not available easily in large part of underdevelopment country like India. It will also prove to be an effective for remote area such as research sites, mines, & deserts where electricity is generally not available. The LPG is cheaper and possesses an environmental free in nature with no ozone depletion potential (ODP). Also LPG is available as a side product in local refineries. The results of the present work indicate the successful use of this propane-butane mixture as an alternative refrigerant to CFCs and HFCs in domestic refrigerator. It would include Experimental setup of working model and detailed observation of the LPG refrigerator and represents its application in refinery, hotel, chemical industries where requirement of LPG is more.     Quantum Theory of Non-Relativistic Particles Interacting with Gravity

    We investigate the effects of the gravitational field on the quantum dynamics of non-relativistic particles. We consider N non-relativistic particles, interacting with the linearized gravitational field. Using the Feynman - Vernon influence functional technique, we trace out the graviton field, to obtain a master equation for the system of particles to first order in GG. The effective interaction between the particles, as well as the self-interaction is non-local in time and in general non-markovian. We show that the gravitational self-interaction cannot be held responsible for decoherence of microscopic particles due to the fast vanishing of the diffusion function. For macroscopic particles though, it leads to diagonalization to the energy eigenstate basis, a desirable feature in gravity induced collapse models.     EF1α and RPL13a represent normalization genes suitable for RT-qPCR analysis of bone marrow derived mesenchymal stem cells

    <p>Abstract</p> <p>Background</p> <p>RT-qPCR analysis is a widely used method for the analysis of mRNA expression throughout the field of mesenchymal stromal cell (MSC) research. Comparison between MSC studies, both <it>in vitro </it>and <it>in vivo</it>, are challenging due to the varied methods of RT-qPCR data normalization and analysis. Therefore, this study focuses on putative housekeeping genes for the normalization of RT-qPCR data between heterogeneous commercially available human MSC, compared with more homogeneous populations of MSC such as MIAMI and RS-1 cells.</p> <p>Results</p> <p>Eight genes including; <it>ACTB, B2M, EF1α, GAPDH, RPL13a, YWHAZ, UBC </it>and <it>HPRT1 </it>were tested as possible housekeeping genes based on their expression level and variability. <it>EF1α </it>and <it>RPL13a </it>were validated for RT-qPCR analysis of MIAMI cells during expansion in varied oxygen tensions, endothelial differentiation, neural precursor enrichment, and during the comparison with RS-1 cells and commercially available MSC. <it>RPL13a </it>and <it>YWHAZ </it>were validated as normalization genes for the cross-species analysis of MIAMI cells in an animal model of focal ischemia. <it>GAPDH</it>, which is one of the most common housekeeping genes used for the normalization of RT-qPCR data in the field of MSC research, was found to have the highest variability and deemed not suitable for normalization of RT-qPCR data.</p> <p>Conclusions</p> <p>In order to make comparisons between heterogeneous MSC populations, as well as adult stem cell like MSC which are used in different laboratories throughout the world, it is important to have a standardized, reproducible set of housekeeping genes for RT-qPCR analysis. In this study we demonstrate that <it>EF1α</it>, <it>RPL13a </it>and <it>YWHAZ </it>are suitable genes for the RT-qPCR analysis and comparison of several sources of human MSC during <it>in vitro </it>characterization and differentiation as well as in an <it>ex vivo</it> animal model of global cerebral ischemia. This will allow for the comparative RT-qPCR analysis of multiple MSC populations with the goal of future use in animal models of disease as well as tissue repair.</p
