    VALIDATED REVERSE PHASE HIGH PERFORMANCE LIQUID CHROMATOGRAPHIC METHOD FOR THE SIMULTANEOUS DETERMINATION OF ARTEMETHER AND LUMEFANTRINE IN FIXED COMBINED DOSAGE FORM

    ABSTRACTObjective: The present aim is to develop simple, precise, and accurate reverse phase high performance liquid chromatographic method (RP-HPLC) forthe simultaneous assay of artemether and lumefantrine in fixed combined dosage form.Methods: The chromatographic study was carried out on Hypersil C18 column (250×4.6 mm, 5 μ) with mobile phase containing a mixture of KH2PO4buffer (pH-3.5) and acetonitrile in the ratio of 45:55% v/v at a flow rate of 1.0 ml/minute with ultraviolet detection at 218 nm in ambient columntemperature.Results: Using the optimized chromatographic conditions artemether and lumefantrine eluted with retention times of 2.207 and 3.733 minutes,respectively. The method was validated according to ICH guidelines with good reproducibility and linear responses, y=60.813.x+629.53 (r²=0.9982)for artemether and y=88.3108.x+2370.2 (r²=0.9912). The % relative standard deviations of intra-day precision was ranged 0.378% and 1.26% forartemether and 0.459% and 1.15% for lumefantrine, respectively. The percentage recoveries were ranged from 99.96% to 100.02% for artemetherand 99.96-99.97% for lumefantrine, respectively.Conclusions: The developed RP-HPLC method was validated as per ICH guidelines and was found to be best suitable for pharmacokinetic studies ofthese mentioned drugs.Keywords: Artemether, Lumefantrine, ICH guidelines

    SIMULTANEOUS DETERMINATION OF FLUPENTIXOL AND NORTRIPTYLINE HCl USING RP-HPLC WITH PDA DETECTOR

    Objective: In the current investigation, to separated and validate the cancer healing drugs (Nortriptyline HCl and Flupentixol) through the HPLC (e-2695) instrument containing a PDA detector. Methods: A simple, selective, validated and well-defined stability that shows isocratic RP-HPLC methodology for the quantitative determination of Nortriptyline HCl and Flupentixol. The chromatographic strategy utilized Agilent eclipse XDB column of dimensions 250x4.6 mm, 5 micron, using isocratic elution with a mobile phase of Methanol and 0.1% orthophosphoric acid (40:60). A flow rate of 1 ml/min and a detector wavelength of 250 nm utilizing the PDA detector were given in the instrumental settings. Validation of the proposed method was carried out according to an international conference on harmonization (ICH) guidelines. Results: LOD and LOQ concentrations for Flupentixol were 0.015 µg/ml, 0.05 µg/ml and for Nortriptyline HCl were 0.3 µg/ml, 1.0 µg/ml. The calibration charts plotted were linear with a regression coefficient of R2>0.999. Recovery, specificity, linearity, accuracy, robustness, ruggedness were determined as a part of method validation and the results were found to be within the acceptable range. Conclusion: The proposed method to be fast, simple, feasible and affordable in assay condition. During stability tests, it can be used for routine analysis of production samples and to verify the quality of drug samples during stability studies

    Objective: The authors report two simple, accurate and economic spectrophotometric methods A and B for the determination of Itopride hydrochloride in bulk and dosage forms.Methods: The proposed methods are based on the formation of chloroform soluble ion-associates in the presence of acidic dyes namely BPB (Method A) and BCP (Method B) exhibiting lmax at 418 and 418 nm respectively.Results: Beer's law is found to be obeyed in the concentration range of 2.0-10.0 µg/ml and 2.0-10.0 µg/ml. The molar absorptivities are found to be 1.42x104 and 9.61x103L/mol. cm for methods A and B. These methods are successfully applied for the assay of Itopride hydrochloride in pharmaceutical formulations

    Experimental and theoretical study of the correlated compound YbCdSn: Evidence for large magnetoresistance and mass enhancement

    The unusual features of topological semimetals arise from its nontrivial band structure. The impact of strong electron correlations on the topological states remains largely unexplored in real materials. Here, we report the magnetotransport properties of YbCdSn single crystals. We found two fundamental experimental evidences of electron correlations through magnetic susceptibility and specific heat. The electron correlations in this compound lead to an intermediate valence state and enhance the effective mass of the charge carriers. This correlated state exhibits large nonsaturating magnetoresistance, low carrier density, magnetic field induced metal-semiconductor-like crossover and a plateau in resistivity at low temperatures. This compound also shows a cusp-like magnetoconductivity at low magnetic field which indicates the presence of weak antilocalization effect. Our band structure calculations of Yb2+^{2+} state predict YbCdSn to be a topological nodal-line semimetal.Comment: 10 Pages, 8 Figures, Published, Phys. Rev.

    A series of novel alkynyl substituted 3,4-dihydropyrimidin-2(1H)-one (DHPM) derivatives were designed, synthesized and evaluated in vitro as potential inhibitors of chorismate mutase (CM). All these compounds were prepared via a multi-component reaction (MCR) involving sequential I2-mediated Biginelli reaction followed by Cu-free Sonogashira coupling. Some of them showed promising inhibitory activities when tested at 30 μM. One compound showed dose dependent inhibition of CM with IC50 value of 14.76 ± 0.54 μM indicating o-alkynylphenyl substituted DHPM as a new scaffold for the discovery of promising inhibitors of CM
