    Characterization of Nepalese rice (Oryza sativa L.) landraces for qualitative traits

    The characterization of rice (Oryza sativa L.) landraces enables to identify phenotypically unique variables which certainly aid in rice breeding program. So, an experiment was conducted in alpha designed to characterize 188 rice landraces from NAGRC (National Agriculture Genetic Resources center) Nepal for their qualitative agromorphologies in research farm of Agriculture and Forestry University (AFU), Rampur, Chitwan in 2020 AD. Twenty-nine qualitative variables viz; twelve leaf characters, six culm characters, four panicle character and seven grain characters were observed and 26 characters revealed diverse trait expressions for each variable in experimented 188 rice accessions. Two leaf characters namely ligule colour and flag leaf attitude for early observation and one grain character (stigma colour for early observation) showed no variation among studied rice accessions. The intensity of green colour of leaf blade, culm lodging resistance and culm habit, secondary branching of panicle, and lemma and palea colour, lemma apiculus colour and sterile lemma colour, elucidated the higher variation in studied characters. The distinction revealed in qualitative characters approves the presence of abundant phenotypic diversity in the landraces assemblage and that eventually signifies the efficient and effective utilization of landrace in rice breeding programs

    RNA-seq provides insights into potato deubiquitinase responses to drought stress in seedling stage

    Ubiquitination is a specific protein degradation and reversible post-translational modification process that can be reversed by deubiquitinase (DUBs). DUBs can hydrolyze and release ubiquitin in the substrate protein so that the substrate can avoid degradation or change its activity, and it has an impact on plant growth and development, cell cycle, abiotic stress response, and other biological processes. Transcript sequences of potato varieties “DM1-3”, “Atlantic” and “Cooperation-88” downloaded from Potato Genome Resources were used for genome-wide identification of the DUB gene family using Hidden Markov Models and verified in the NCBI CD-Search tool. The characteristics of DUB genes from different potato varieties were analyzed including subcellular localization, gene structural motifs, phylogenetic tree, and sequence homology. Polyethylene glycol 6000 (PEG6000) induced drought stress transcriptome analysis was performed on the “Atlantic”, and differentially expressed genes were screened, with emphasis on the characterization of deubiquitinase. DUB genes have a complex gene structure, often with a large number of exons and alternative splicing. Their promoters contain abundant abiotic stress-responsive elements, such as 425 MYC, 325 ABRE, and 320 MYB. There are also a large number of orthologous genes in the DUBs of the three potato varieties, and these genes are often clustered in similar regions on the genome. We performed transcriptome sequencing of the potato under PEG-induced drought stress and analyzed it for the first time using the Atlantic as a reference genome. We identified a total of 6067 down-regulated differentially expressed genes (DEGs) and 4950 up-regulated DEGs under PEG-induced drought stress. We screened the expression of DUBs and observed that 120 DUBs were up-regulated where most of them functioned in the nucleus, and the interacting proteins of DUBs were also localized in the nucleus. We have comprehensively identified and analyzed potato DUBs, and the accurately aligned transcriptome data which will further deepen the understanding of DUBs involved in the regulation of osmotic stress

    Estimation of heterosis in yield and yield attributing traits in single cross hybrids of maize

    A field experiment was conducted at National Maize Research Program, Rampur, Chitwan, Nepal during winter season from 6th October, 2015 to 5th March 2016 to estimate different heterosis on single cross maize hybrids . Thirteen maize hybrids were tested randomized complete block design with three replications. Hybrid namely RML-98/RL-105 gave the highest standard heterosis (57.5%) for grain yield over CP-666 followed by RML-4/NML-2 (32.6%), RML-95/RL-105 (29%) and RML-5/RL-105 (20.6%). The hybrid RML-98/RL-105 produced the highest standard heterosis (75.1%) for grain yield over Rajkumar followed by RML-4/NML-2(50.2%), RML-95/RL-105(46.6%), RML-5/RL-105 and (35.7%). Mid and better parent heterosis were significantly higher for yield and yield attributes viz. ear length, ear diameter, no of kernel row per ear, no of kernel per row and test weight. The highest positive mid-parent heterosis for grain yield was found in RML-98/RL-105 followed by RML-5/RL-105, RML-95/RL-105, and RML-4/NML-2. For the grain yield the better parent heterosis was the highest in RML-98/RL-105, followed by RML-5/RL-105, RML-95/RL-105, and RML-4/NML-2. These results suggested that maize production can be maximized by cultivating hybrids namely RML-98/RL-105, RML-5/RL-105, RML-95/RL-105, and RML-4/NML-2

    Review of Indications of FDA-Approved Immune Checkpoint Inhibitors per NCCN Guidelines with the Level of Evidence

    Cancer is associated with higher morbidity and mortality and is the second leading cause of death in the US. Further, in some nations, cancer has overtaken heart disease as the leading cause of mortality. Identification of molecular mechanisms by which cancerous cells evade T cell-mediated cytotoxic damage has led to the modern era of immunotherapy in cancer treatment. Agents that release these immune brakes have shown activity to recover dysfunctional T cells and regress various cancer. Both cytotoxic T-lymphocyte-associated protein 4 (CTLA-4) and Programmed Death-1 (PD-1) play their role as physiologic brakes on unrestrained cytotoxic T effector function. CTLA-4 (CD 152) is a B7/CD28 family; it mediates immunosuppression by indirectly diminishing signaling through the co-stimulatory receptor CD28. Ipilimumab is the first and only FDA-approved CTLA-4 inhibitor; PD-1 is an inhibitory transmembrane protein expressed on T cells, B cells, Natural Killer cells (NKs), and Myeloid-Derived Suppressor Cells (MDSCs). Programmed Death-Ligand 1 (PD-L1) is expressed on the surface of multiple tissue types, including many tumor cells and hematopoietic cells. PD-L2 is more restricted to hematopoietic cells. Blockade of the PD-1 /PDL-1 pathway can enhance anti-tumor T cell reactivity and promotes immune control over the cancerous cells. Since the FDA approval of ipilimumab (human IgG1 k anti-CTLA-4 monoclonal antibody) in 2011, six more immune checkpoint inhibitors (ICIs) have been approved for cancer therapy. PD-1 inhibitors nivolumab, pembrolizumab, cemiplimab and PD-L1 inhibitors atezolizumab, avelumab, and durvalumab are in the current list of the approved agents in addition to ipilimumab. In this review paper, we discuss the role of each immune checkpoint inhibitor (ICI), the landmark trials which led to their FDA approval, and the strength of the evidence per National Comprehensive Cancer Network (NCCN), which is broadly utilized by medical oncologists and hematologists in their daily practice

    Stroke epidemiology and outcomes of stroke patients in Nepal: a systematic review and meta-analysis

    Abstract Background With an increasing burden of stroke, it is essential to minimize the incidence of stroke and improve stroke care by emphasizing areas that bring out the maximum impact. The care situation remains unclear in the absence of a national stroke care registry and a lack of structured hospital-based data monitoring. We conducted this systematic review and meta-analysis to assess the status of stroke care in Nepal and identify areas that need dedicated improvement in stroke care. Methods A systematic literature review was conducted to identify all studies on stroke epidemiology or stroke care published between 2000 and 2020 in Nepal. Data analysis was done with Statistical Package for Social Sciences (SPSS) and Comprehensive Meta-analysis (CMA-3). Results We identified 2533 studies after database searching, and 55 were included in quantitative and narrative synthesis. All analyses were done in tertiary care settings in densely populated central parts of Nepal. Ischemic stroke was more frequent (70.87%) than hemorrhagic (26.79%), and the mean age of stroke patients was 62,9 years. Mortality occurred in 16.9% (13-21.7%), thrombolysis was performed in 2.39% of patients, and no studies described thrombectomy or stroke unit care. Conclusion The provision of stroke care in Nepal needs to catch up to international standards, and our systematic review demonstrated the need to improve access to quality stroke care. Dedicated studies on establishing stroke care units, prevention, rehabilitation, and studies on lower levels of care or remote regions are required
