Aneuploidy among androgenic progeny of hexaploid triticale (XTriticosecale Wittmack).
Doubled haploids are an established tool in plant breeding and research. Of several methods for their production, androgenesis is technically simple and can efficiently produce substantial numbers of lines. It is well suited to such crops as hexaploid triticale. Owing to meiotic irregularities of triticale hybrids, aneuploidy may affect the efficiency of androgenesis more severely than in meiotically stable crops. This study addresses the issue of aneuploidy among androgenic regenerants of triticale. Plant morphology, seed set and seed quality were better predictors of aneuploidy, as determined cytologically, than flow cytometry. Most aneuploids were hypoploids and these included nullisomics, telosomics, and translocation lines; among 42 chromosome plants were nulli-tetrasomics. Rye chromosomes involved in aneuploidy greatly outnumbered wheat chromosomes; in C(0) rye chromosomes 2R and 5R were most frequently involved. While the frequency of nullisomy 2R was fairly constant in most cross combinations, nullisomy 5R was more frequent in the most recalcitrant combination, and its frequency increased with time spent in culture with up to 70% of green plants recovered late being nullisomic 5R. Given that 5R was not involved in meiotic aberrations with an above-average frequency, it is possible that its absence promotes androgenesis or green plant regeneration. Overall, aneuploidy among tested combinations reduced the average efficiency of double haploid production by 35% and by 69% in one recalcitrant combination, seriously reducing the yield of useful lines

Populations of doubled haploids for genetic mapping in hexaploid winter triticale.
To create a framework for genetic dissection of hexaploid triticale, six populations of doubled haploid (DH) lines were developed from pairwise hybrids of high-yielding winter triticale cultivars. The six populations comprise between 97 and 231 genotyped DH lines each, totaling 957 DH lines. A consensus genetic map spans 4593.9 cM is composed of 1576 unique DArT markers. The maps reveal several structural rearrangements in triticale genomes. In preliminary tests of the populations and maps, markers specific to wheat segments of the engineered rye chromosome 1R (RM1B) were identified. Example QTL mapping of days to heading in cv. Krakowiak revealed loci on chromosomes 2BL and 2R responsible for extended vernalization requirement, and candidate genes were identified. The material is available to all parties interested in triticale genetics
Sposoby barwienia i preparowania cietych pedow Molucella laevis L.
Materiałem doświadczalnym były cięte pędy Molucella laevis. W doświadczeniach sprawdzono wpływ pięciu stężeń gliceryny (0,5; 1,0; 2,5; 5,0; 10%) na jakość utrwalonych pędów. Wykazano, że najwyższą jakość uzyskano, stosując 5% roztwór gliceryny. Pędy były elastyczne i równomiernie odbarwione. W doświadczeniach określono także wpływ stężenia barwnika firmy Dillewijn (10, 15 i 20 g·dm⁻³) i czasu barwienia (6 i 24 godz.) na dekoracyjność i trwałość ciętych pędów dzwonków irlandzkich. Najbardziej korzystne okazało się barwienie w stężeniu 10 g·dm⁻³ przez 6 godz. Uzyskane w ten sposób pędy były równomiernie wybarwione i wykazywały największą trwałość po zbiorze. Umieszczenie pędów Molucella laevis po barwieniu do pożywki standardowej, stosowanej do przedłużania kwiatów ciętych, (8HQC + 2% S) nie wpłynęło na ich trwałość.Cut shoots of Molucella laevis were used in the experiments. Effect of five glycerin concentrations (0,5; 1,0; 2,5; 5,0; 10%) on the quality of shoots was tested. Preserving shoots in 5% water solution of glycerin assured their best decorative value: shoots were flexible and uniformly decolored. Next the effect of dye concentration (10, 15 or 20 g·dm⁻³) and duration of dyeing (6 or 24 h) was evaluated. Dyeing for 6 hours in the water dye solution of 10 g·dm⁻³ gave the best results: shoots were uniformly dyed and had the longest vase life. A standard preservative applied for cut flowers (8HQC + 2% S) did not affect the longevity of dyed shoots of Molucella laevis
Wpływ sposobu przechowywania na starzenie się ciętych cantedeskii (Zantedeschia aethiopica Spr.) i funkii (Hosta 'Undulata Erromena')
Materiałem roślinnym były cięte liście cantedeskii etiopskiej (Zantedeschia aethiopica Spr.) i funkii (Hosta ‘Undulata Erromena’). W doświadczeniach sprawdzono wpływ sposobu przechowywania liści przez 3 dni w obniżonej temperaturze (5±1°C): na świetle/na mokro; na świetle/na sucho; w ciemności/na mokro; w ciemności/na sucho, na pozbiorczą ich trwałość oraz na zmiany zawartości chlorofilu w trakcie starzenia. Testowane gatunki nie zareagowały na chłodzenie w temperaturze 5°C: trwałość liści przechowywanych na świetle w wodzie destylowanej była porównywalna do trwałości liści nieprzechowywanych, wstawionych bezpośrednio po zbiorze do 20±1°C. Na starzenie ciętych liści obu gatunków wpływ miał jednak sposób przechowywania w obniżonej temperaturze. Przechowywanie liści cantedeskii w ciemności, chociaż przez stosunkowo krótki okres czasu - 3 dni - przyspieszyło degradację chlorofilu i skróciło trwałość liści po zbiorze. Szczególnie widoczne skrócenie okresu dekoracyjności wystąpiło przy przechowywaniu liści w ciemności, na sucho. W ciętych liściach cantedeskii odnotowano wyraźny negatywny efekt ciemności, a w liściach funkii - suszy. Przechowywanie liści funkii na sucho zarówno w ciemności, jak i na świetle przyspieszyło degradację chlorofilu i skróciło ich pozbiorczą trwałość.Cut leaves of Zantedeschia aethiopica SPR. and Hosta ‘Undulata Erromena’ were subjected to dry or wet storage at 5±1°C, in light or darkness. After 3 days in the above conditions leaf vase life was evaluated at 20±1°C in comparison to unstored leaves, placed into water immediately after harvest. Low temperature did not affect leaf vase life in neither species, however, the method of storage did. Storage in darkness, especially without water, accelerated chlorophyll loss in Zantedeschia and decreased leaf longevity. In Zantedeschia darkness was the factor negatively affecting cut leaves while in Hosta water stress had more affect on leaves. Those dry-stored, both in light and darkness, showed a faster chlorophyll degradation and shorter vase life
Przydatność kwiatów ciętych wybranych odmian powojników (Clematis L.) do wykorzystania we florystyce
Materiałem roślinnym były cięte kwiaty 17 odmian i jednego gatunku powojników (Clematis L.). W doświadczeniach sprawdzono przydatność poszczególnych odmian do uprawy na kwiat cięty i określono wpływ pożywek na trwałość kwiatów po zbiorze. Wykazano, że możliwość wykorzystania powojników na kwiat cięty zależy od odmiany i rodzaju zastosowanej pożywki. Najdłuższą trwałością cechowały się kwiaty odmian z grupy botanicznych: 'Arabella' i wielkokwiatowych: 'Julka' i 'Silver Moon', których trwałość w wodzie destylowanej dochodziła do około 12 dni. Nieco krótszą trwałością (około 10 dni) odznaczały się odmiany: 'Gravetye Beauty', 'Proteus' i gatunek С. x durandii, które także z powodzeniem można polecić do uprawy na kwiat cięty. Większość badanych odmian pozytywnie reagowała na wstawienie ich do pożywki przedłużającej trwałość kwiatów. Najskuteczniejsza okazała się pożywka 8HQC z dodatkiem 2% sacharozy. Wyższe stężenia cukru powodowały przebarwienia kwiatów i skracały ich trwałość.Vase life of 18 cultivars of Clematis L. was evaluated both in water and in 5 preservative solutions. Longevity of Clematis depends on a cultivar and a vase solution. Vase life in water was the longest for the cultivar 'Arabella' (12.6 days), while among cultivars 'Julka' and 'Silver Moon' it was about 12 days. Cultivars 'Gravetye Beauty' and 'Proteus' can also be recommended as cut flowers as well as C. x durandii, all of them having vase life of about 10 days. Most cultivars responded positively to some of the preservatives, the solution of 8HQC and 2% sucrose being the best. Higher sugar concentration caused flower discoloration and shortened their longevity
Występowanie blokad w ciętych pędach powojnika Clematis L.
During vase life of cut flowers obstructions in stem xylem
vessels develop. Such obstructions may restrict water uptake
in stems and its transport towards flowers, thus lowering their
ornamental value and longevity. Clematis is a very attractive
plant which can be used as a cut flower in floral compositions.
However, nothing is known about the histochemical or cytological
nature of xylem blockages occurring in cut stems of this
plant. Observations carried out on Clematis cv. ‘Solidarność’
proved that tyloses appeared as a principal source of xylem
blockage in cut stems. The preservative composed of 200 mg
× dm-3 8-HQC (8-hydroxyquinolin citrate) and 2% sucrose arrested
development of xylem blockage, while the vessels in stems
kept in water were filled with tyloses or an amorphic substance.
PAS reaction proved that polysaccharides were present in the
xylem occlusions, whereas no homogalacturonans were immunolocalized
in tyloses using JIM 5 and JIM 7 antibodies. The
present study provides new information on the origin of xylem
occlusions in clematis and their development in two different
vase solutions. Such information can be useful to develop proper
postharvest treatments aiming to improve keeping qualities
of this new cut flower.Podczas życia w wazonie kwiatów ciętych
mogą pojawić się blokady hamujące swobodny transport
wody w pędach. Blokady te skutecznie utrudniają
swobodny przepływ wody, powodując tym samym obniżenie
wartości kwiatów oraz skracając ich trwałość.
Powojnik jest niezwykle atrakcyjną rośliną wykorzystywaną
jako pnącze ogrodowe, a od niedawna także
w bukietach. Do tej pory nie prowadzono badań nad
trwałością posprzętną tej rośliny oraz nad charakterem
blokad występujących w pędach powojnika. Obserwacje
prowadzone na odmianie ‘Solidarność’ pokazały,
że głównym źródłem blokad pędów u powojnika
są wcistki. Pożywka składająca się z 200 mg × dm-3
× 8-HQC (cytrynian 8 hydroksykinoliny) z dodatkiem
2% sacharozy skutecznie zahamowała rozwój blokad
w pędach, w przeciwieństwie do wody destylowanej.
Reakcja PAS pokazała, że jednym z komponentów
wcistek były wielocukry, jednak nie wykryto wśród
nich pektyn i homogalakturonianów. Przeprowadzone
obserwacje dostarczyły informacji na temat blokad występujących
w ciętych pędach powojników, co pozwoli
na stworzenie precyzyjnych procedur mających na celu
skuteczne przedłużenie trwałości ciętych powojników
Wplyw wybranych substancji chemicznych na akumulacje amoniaku i proliny w czasie starzenia cietych lisci funkii [Hosta 'Undulata Erromena']
W doświadczeniach określono wpływ pożywki standardowo stosowanej do przedłużania trwałości kwiatów ciętych cytrynian 8-hydroksychinoliny + 2% sacharoza (8HQC + 2% S) oraz benzyloadeniny (BA) i kwasu giberelinowego (GA₃) na zawartość amoniaku i proliny w czasie starzenia ciętych liści funkii ‘Undulata Erromena’. Wykazano, iż w czasie starzenia ciętych liści tego gatunku, przetrzymywanych w wodzie destylowanej, następuje prawie 2,5-krotny wzrost zawartości amoniaku, który pociąga za sobą prawie proporcjonalny, bo 2,4-krotny wzrost poziomu proliny. Poziom obu komponentów w liściach funkii uzależniony był jednak od rodzaju zastosowanej substancji chemicznej. Wstawienie liści bezpośrednio po zbiorze do pożywki standardowej spowodowało 4-krotny wzrost poziomu amoniaku i prawie 3-krotny proliny w stosunku do stanu wyjściowego. Zastosowanie BA w formie 24-godzinnego kondycjonowania powstrzymało wzrost zawartości obu komponentów w ciętych liściach funkii, utrzymując ich zawartość na poziomie zbliżonym do wyjściowego. Zawartość amoniaku i proliny była istotnie wyższa przy zastosowaniu GA₃ niż BA.The contents of ammonium and proline in ageing cut leaves of Hosta ‘Undulata Erromena’ affected by the standard preservative used to prolong cut flower longevity (composed of 8HQC 200 mg·dm⁻³ + sucrose 2%) and benzyladenine (BA) or gibberellic acid (GA₃) was examined. In leaves kept held in water the 2.5 fold increase in ammonium content was observed as compared to the initial level and almost a similar rise in proline content occurred in the senescing leaves. In leaves kept held in the preservative solution increase in ammonium and proline contents was 4-fold and 3-fold, respectively. BA application as a 24 hour puls treatment stopped both ammonium and proline accumulation, maintaining their contents at the initial level. BA prevented ammonium and proline accumulation more effectively than GA₃ did
